Project description:Pea (Pisum. sativum L.) is a traditional and important edible legume that can be sorted into grain pea and vegetable pea according to their harvested maturely or not. Vegetable pea by eating the fresh seed is becoming more and more popular in recent years. These two type peas display huge variations of the taste and nutrition, but how seed development and nutrition accumulation of grain pea and vegetable pea and their differences at the molecular level remains poorly understood. To understand the genes and gene networks regulate seed development in grain pea and vegetable pea, high throughput RNA-Seq and bioinformatics analysis were used to compare the transcriptomes of vegetable pea and grain pea developing seed. RNA-Seq generated 18.7 G raw data, which was then de novo assembled into 77,273 unigenes with a mean length of 930 bp. Functional annotation of the unigenes was carried out using the nr, Swiss-Prot, COG, GO and KEGG databases. There were 459 and 801 genes showing differentially expressed between vegetable pea and grain pea at early and late seed maturation phases, respectively. Sugar and starch metabolism related genes were dramatically activated during pea seed development. The up-regulated of starch biosynthesis genes could explain the increment of starch content in grain pea then vegetable pea; while up-regulation of sugar metabolism related genes in vegetable pea then grain pea should participate in sugar accumulation and associated with the increase in sweetness of vegetable pea then grain pea. Furthermore, transcription factors were implicated in the seed development regulation in grain pea and vegetable pea. Thus, our results constitute a foundation in support of future efforts for understanding the underlying mechanism that control pea seed development and also serve as a valuable resource for improved pea breeding.

Project description:Ascochyta blight (AB) is a devastating and aggressive disease of field peas caused by multiple pathogens and the occurrence of the combinations of these pathogens forms a disease complex of which Peyronellaea pinodes, is the most aggressive. AB symptoms include necrotic lesions on all above ground parts of the plant, which can reduce crop yield by up to 70% and result in major economic losses globally. Crop resistance to the disease is reported to be quantitative and, although several genes have been reported to be AB response genes in Spanish pea genotypes, the lines or genotypes of the Australian field pea breeding program have not been investigated. Plants have specialized defence mechanisms that enable them respond to pathogen invasion, and this involves a cascade of genes which are differentially regulated upon infection. In this study, the time-course gene expression profiles of four Australian field pea genotypes including commercial varieties, known to have varying levels of AB resistance were investigated in response to P. pinodes infection. Infected and control plants at 0, 6, 24, 48, 72 and 96HPI were sampled for RNA seq. Mapping of transcriptome sequencing data against the field pea genome resulted in about the identification of c. 6,700 differentially expressed genes. Of the 21 AB resistance genes reported in previously published studies, only seven were present on the list of DEGs generated in this study. No significant pattern was identified in gene expression for the seven genes to differentiate resistant from susceptible genotypes. However, four key genes i.e., disease response protein, malate dehydrogenase, isoflavone 2 hydrogenase and Isoflavone reductase were shown to be associated with suppression of AB disease phenotype after 96HPI. Furthermore, GO functional annotation revealed several genes associated with defence including genes associated with the Salicylic acid pathway, reactive oxygen species, programmed cell death and hypersensitive response. Although the overall result showed a lack of strong resistance in the Australian pea genotypes as well as the fact that disease suppression does not directly equal resistance, we however were able to identify potential candidates that could be pyramided from exotic material into the breeding program to assess for improved resistance.

Project description:In angiosperms, the mature seed consists of an embryo (E), a seed coat (SC), and, in many cases, an endosperm. In contrast to knowledge about embryo and endosperm, we have relatively little knowledge of SC, especially at the genomics level. In this study, we analyzed the gene expression during seed development using the panel of cultivated and wild pea genotypes. We report the comprehensive gene expression changes related both to development as well as domestication status. Analysis of seed developmental stages revealed extensive modification of gene expression between wild pea progenitor and cultivated pea crop. A significant difference in gene expression dynamics appeared between early and late developmental stages D1, D2, and D3, D4, D5 in wild pea genotypes, where the expression was increased 3-5-fold and 5-10-fold, respectively. Our work extends knowledge about the role of the seed coat during pea seed development. We described gene expression dynamic resulting in specific metabolic profiles providing new insight into pea domestication.

Project description:Seed development is a complex process controlled by many factors, including genetics, plant growth regulators, and the environment. Understanding the different compositions and abundances of proteins in various types of seeds at different developmental stages is fundamental for improving seed quality and enhancing nutritional and food security. In this study, we applied label-free quantitative proteomics to analyze round and wrinkled pea seeds at five different growth stages: 4, 7, 12, 15 days after anthesis (DAA), and at maturity. Wrinkled peas had lower starch content (29.5%) compared to round peas (~47-55%). Proteomic analysis identified 3,659 protein groups, of which approximately 21-24% proteins were shared across growth stages. Relatively more proteins were identified during early seed development compared to later stages, and the protein profiles were also drastically different between early and late stages. Statistical analysis identified 735 significantly different proteins between wrinkled and round seeds, regardless of the growth stage. These detected proteins were characterized into different functional classes including metabolic enzymes, proteins involved in protein biosynthesis and homeostasis, carbohydrate metabolism, vesicle trafficking, cell division, and cell wall organization. Cell division-related proteins were more abundant in the early stages of seed development, whereas storage proteins and proteins implicated in lipid metabolism were more abundant at the later stages. Wrinkled seeds had a significantly lower abundance of starch-branching enzyme than did round seeds. This enzyme is involved in the biosynthesis of the amylopectin component of starch. Seed storage proteins such as legumin and a form of albumin (PA2) accumulated more in round pea genotypes, whereas vicilin was more abundant in the wrinkled pea genotype. In summary, this study can enhance our understanding of pea seed development, highlighting key differences in protein profiles between round and wrinkled pea seeds.

Project description:Abiotic stresses caused by adverse environmental conditions are responsible for heavy economic losses on pea crop, being drought one of the most important abiotic constraints. Development of pea cultivars well adapted to dry conditions has been one of the major tasks in breeding programs. The increasing food requirements drive the necessity to broaden the molecular basis of tolerance to drought to develop pea cultivars well adapted to dry conditions. We have used a shotgun proteomic approach (nLC-MSMS) to study the tolerance to drought in three pea genotypes which were selected based on differences in the level of water deficit tolerance. Multivariate statistical analysis of data unraveled 367 significant differences of 700 identified when genotypes and/or treatment were compared. More than half of the significantly changed proteins belong to primary metabolism and protein regulation categories. We propose different mechanisms to cope drought in the genotypes studied. Maintenance of the primary metabolism and protein protection seems a strategy for drought tolerance. On the other hand susceptibility might be related to maintenance of the homeostatic equilibrium, a very energy consuming process.

Project description:Knowledge about an organism’s cell and tissue-specific transcriptional repertoire is essential for understanding the gene regulatory circuits that control key developmental events. The shoot apical meristem (SAM) is responsible for development of all the above ground parts of plants. Our understanding of SAM at the molecular level is far from complete. The present work investigates the global gene expression repertoire of SAMs in the garden pea (Pisum sativum). To this end, 10,346 EST sequences representing 7611 unique genes were generated from pea SAM cDNA libraries. These sequences, together with previously reported ESTs, were used to construct a 12K oligonucleotide array used to identify genes exhibiting differential SAM expression, as compared to the axillary meristem, root apical meristem, and non-meristematic tissues. We identified a number of genes that are predominantly expressed in specific cell layers or domains of the SAM, and thus are likely components of the gene networks involved in stem cell maintenance and initiation of lateral organ primordial cells. In situ hybridization confirmed the spatial localisation of some of these key genes within the SAM. Our data also indicate the diversification of some gene expression patterns and functions in legume crop plants.
