Project description:The intake of fermented foods is gaining increasing interest due to their health-promoting benefits. Among them, fermented dairy foods have been associated with obesity prevention, and reduction of the risk of metabolic disorders and immune-related pathologies. Fermented foods could lead to these health benefits by providing the consumer with both easily metabolizable nutrients and beneficial microorganisms. Our aim was to evaluate the relationship between the consumption of fermented dairy products and the intestinal microbiota, serum lipid profile, and the pro-oxidant/inflammatory status. 130 healthy adults were evaluated. Dietary fermented food intake was assessed by an annual food frequency questionnaire (FFQ), including 26 fermented dairy products. Levels of the major phylogenetic types of the intestinal microbiota were determined by qPCR, and concentration of fecal short chain fatty acids were assessed by gas chromatography. Serum glucose and lipid profile, as well as serum malondialdehyde (MDA), C-reactive protein (CRP), and leptin levels were determined by standardized protocols. Among fermented dairy foods, natural yogurt, sweetened yogurt and matured/semi-matured cheese were the most consumed. While natural yogurt consumers showed increased fecal levels of Akkermansia with respect to non-consumers, sweetened yogurt intake was associated to lower levels of Bacteroides. Serum levels of CRP were also significantly reduced in yogurt consumers. Our results underline the interest in exploring the potential effects of the different yogurt types and the role the microbiota may play in such effects.

Project description:The complicated biological process of how dairy cows convert indigestible nutrients for humans into nutritious milk is orchestrated by various tissues. Here, we deciphered the single-cell landscapes of 10 bovine tissue types covering 88,013 cells. We characterized the metabolic features of epithelial cell subtypes that exert unique functions to the tissues of residence. We also identified the cell subtypes that are essential for the transport of certain nutrients. Th17 cells were specifically enriched in the rumen, reticulum, and omasum tissues and predominantly interacted with the epithelial cell subtypes with high potential uptake capacities of short-chain fatty acids through IL-17 signaling. The comparison between IL17RAhighIL17RChigh cells and other cell types explained the important role of Th17 cells in regulating the epithelial cellular transcriptional response to nutrients transport in these three tissues. The findings enhance our understanding of ruminants biology and open new avenues for an improved animal production in dairy cows.

Project description:The aim of this study was to determine the effects of unprotected dietary unsaturated fatty acids (UFA) from different plant oils on gene expression in the mammary gland of grazing dairy cows. Milk composition and gene expression in the mammary gland tissue were evaluated in grazing dairy cows supplemented with different unsaturated fatty acids (UFA). The UFA supplementation improves the health and nutrition quality aspects of dairy milk, but also affects the gene networks expression signature associated with cellular growth and proliferation, cell-death, signalling, nutrient metabolism, and immune response, and in turn, the mammary gland integrity and health. Project description:To investigate dairy consumption in ancient Mongolia, we analysed dental calculus samples from four Late Bronze Age (LBA, 1500-1000 BCE) individuals for proteomic evidence of milk proteins. As many archaeological sites before Mongolia's Iron Age suffer from a dearth of occupational materials, looking to biomolecular markers of dietary intake can open new investigational avenues into ancient economies. In this case, we use a previously established method of extracting proteins from calculus to explore the consumption of dairy products at LBA Khirigsuur sites in northern Mongolia's Hovsgol Aimag. Seven of nine individual's calculus contained peptides from the whey protein Beta-lactoglobulin from Ovis, Capra hircus, Bos, and general Bovidae species. Aside from proteomics, these and 16 other individuals from the site were analysed for aDNA. We found that 18 of the 20 were primarily from one genetic ancestral group, and Ancient North Eurasian (ANE). One of the outliers represents a combination of ANE and Western Steppe Herder (WSH), with the other a combination of ANE and Eastern Asian (EE). This finding, while important in its own right, evidences the earliest known dairy consumption in Mongolia, and supports a widely held assumption that pastoralism was a primary subsistence strategy in the ancient Eastern Steppes. The combined proteomic and DNA evidence suggest that Western Steppe dairy animals and technology entered Mongolia before genetic admixture.

Project description:The gut microbiota in our intestinal tract metabolizes non-digestible compounds into essential nutrients and signaling molecules (i.e. short-chain fatty acids), affecting our immune system and the development of various human diseases. Ingested environmental contaminants (xenobiotics) can disrupt the bacterial community and enzymatic activity, ultimately influencing the host. Pervasive xenobiotics include bisphenols and poly- and perfluoroalkyl substances (PFAS). Both classes of chemicals have been reported to affect the immune system and cause adverse effects on human metabolism. Since humans are exposed to a complex mixture of environmental contaminants it is critical to evaluate the effects of xenobiotics in mixtures. In our study, an in vitro bioreactor model system based on the simplified human microbiome model (SIHUMIx) was used to investigate the direct effects of either perfluorooctanoic acid (PFOA), perfluorohexanoic acid (PFHxA) and perfluorobutanoic acid (PFBA) or bisphenol S (BPS) and bisphenol F (BPF) or a combined mixture on the microbiota. We observed an increased production of the short-chain fatty acids (SCFAs) acetate and butyrate following PFAS exposure. A metaproteomics approach revealed changes in molecular pathways in all treatments, including alterations in vitamin and cofactor synthesis and an additional effect on fatty acid synthesis in BPX-treated reactors. This study highlights the need to assess the combined effects of xenobiotics and to better protect public health by considering adverse effects on the microbiome in the risk assessment of environmental chemicals.

High-performing dairy cows require a particular composition of nutritional ingredients, adapted to their individual requirements and depending on their production status. The optimal dimensioning of minerals in the diet, one being potassium, is indispensable for the prevention of imbalances. Potassium balance in cows is the result of potassium intake, distribution in the organism, and excretion, and it is closely related to glucose and electrolyte metabolism. In this paper, we present a dynamical model for potassium balance in lactating and nonlactating dairy cows based on ordinary differential equations. Parameter values were obtained from clinical trial data and from the literature. To verify the consistency of the model, we present simulation outcomes for 3 different scenarios: potassium balance in (1) nonlactating cows with varying feed intake, (2) nonlactating cows with varying potassium fraction in the diet, and (3) lactating cows with varying milk production levels. The results give insights into the short- and long-term potassium metabolism, providing an important step toward the understanding of the potassium network, the design of prophylactic feed additives, and possible treatment strategies.
