Project description:Plants are attacked by diverse herbivores and respond with manifold defense responses. To study transcriptional and other early regulation events of these plant responses, herbivory is often mimicked to standardize the temporal and spatial dynamics that vary tremendously for natural herbivory. Yet to what extent such mimics of herbivory are able to elicit the same plant response as real herbivory remains largely undetermined. We examined the transcriptional response of a new model plant to herbivory by a lepidopteran larva and to a commonly used herbivory mimic by applying the larvae’s oral secretions to standardized wounds. We designed a microarray for Solanum dulcamara and showed that the transcriptional response to real and to simulated herbivory by Spodoptera exigua overlapped moderately by about 40%. Interestingly, certain responses were mimicked better than others; 60% of the genes up-regulated but not even a quarter of the genes down-regulated by herbivory were similarly affected by application of oral secretions to wounds. While the regulation of genes involved in signaling, defense and water stress were mirrored well by the herbivory mimic, most of the genes related to photosynthesis, carbohydrate- and lipid metabolism were exclusively regulated by real herbivory. Thus, wounding and elicitor application decently mimics herbivory-induced defense responses but likely not the re-allocation of primary metabolites induced by real herbivory.

Project description:Permafrost soil in high latitude tundra is one of the largest terrestrial carbon (C) stocks and is highly sensitive to climate warming. Understanding microbial responses to warming induced environmental changes is critical to evaluating their influence on soil biogeochemical cycles. In this study, a functional gene array (i.e. GeoChip 4.2) was used to analyze the functional capacities of soil microbial communities collected from a naturally degrading permafrost region in Central Alaska. Varied thaw history was reported to be the main driver of soil and plant differences across a gradient of minimally, moderately and extensively thawed sites. Compared with the minimally thawed site, the number of detected functional gene probes across the 15-65 cm depth profile at the moderately and extensively thawed sites decreased by 25 % and 5 %, while the community functional gene beta-diversity increased by 34% and 45%, respectively, revealing decreased functional gene richness but increased community heterogeneity along the thaw progression. Particularly, the moderately thawed site contained microbial communities with the highest abundances of many genes involved in prokaryotic C degradation, ammonification, and nitrification processes, but lower abundances of fungal C decomposition and anaerobic-related genes. Significant correlations were observed between functional gene abundance and vascular plant primary productivity, suggesting that plant growth and species composition could be co-evolving traits together with microbial community composition. Altogether, this study reveals the complex responses of microbial functional potentials to thaw related soil and plant changes, and provides information on potential microbially mediated biogeochemical cycles in tundra ecosystems.

Project description:Climate change can influence ecosystems via both direct effects on individual organisms and indirect effects mediated by species interactions. However, we understand little about how these changes will ripple through ecosystems or whether there are particular ecological characteristics that might make ecosystems more susceptible-or more resistant-to warming. By combining in situ experimental warming with herbivore manipulations in a natural rocky intertidal community for over 16 months, we show that herbivory regulates the capacity of marine communities to resist warming. We found that limpet herbivores helped to preserve trophic and competitive interactions under experimental warming, dampening the impact of warming on overall community composition. The presence of limpets facilitated the survival of the main habitat modifier (barnacles) under warmer conditions, which, in turn, facilitated the presence of a consumer guild. When limpets were removed, environmental warming altered trophic, competitive, and facilitative interactions, with cascading impacts on community succession and stability. We conclude that conserving trophic structure and the integrity of interaction networks is vitally important as Earth continues to warm.

Differentially expressed genes were determined using three criteria: fold-change between treated and untreated control leaves > 1.5-fold, P value < 0.05 and Q value < 0.05. This study identified > 3,000 differentially expressed genes in response to simulated insect feeding damage, which possess distinct source/sink and treated/systemic patterns.

Project description:Plant-emitted volatile organic compounds (VOCs) play fundamental roles in atmospheric chemistry and ecological processes by contributing to aerosol formation1 and mediating species interactions2. Rising temperatures and the associated shifts in vegetation composition have been shown to be the primary drivers of plant VOC emissions in Arctic ecosystems3. Although herbivorous insects also strongly alter plant VOC emissions2, no studies have addressed the impact of herbivory on plant VOC emissions in the Arctic. Here we show that warming dramatically increases the amount, and alters the blend, of VOCs released in response to herbivory. We observed that a tundra ecosystem subjected to warming, by open-top chambers, for 8 or 18 years showed a fourfold increase in leaf area eaten by insect herbivores. Herbivory by autumnal moth (Epirrita autumnata) larvae, and herbivory-mimicking methyl jasmonate application, on the widespread circumpolar dwarf birch (Betula nana) both substantially increased emissions of terpenoids. The long-term warming treatments and mimicked herbivory caused, on average, a two- and fourfold increase in monoterpene emissions, respectively. When combined, emissions increased 11-fold, revealing a strong synergy between warming and herbivory. The synergistic effect was even more pronounced for homoterpene emissions. These findings suggest that, in the rapidly warming Arctic, insect herbivory may be a primary determinant of VOC emissions during periods of active herbivore feeding.
