Project description:Arabidopsis thaliana plants were infested i) with sucking insect herbivores (the generalist aphid Myzus persicae and the specialist aphid Brevicoryne brassicae), ii) with chewing insect herbivores (generalist caterpillars of Spodoptera exigua and specialist caterpillars of Pieris rapae) or iii) were treated by wounding. For each treatment, rosette leaves were harvested at two time points (6h and 24h) after removal of insects. For chewing herbivores and wounding both local, i.e. immediately damaged leaves, and systemic, i.e. undamaged leaves from the same plant, were collected. Control plants were uninfested, but otherwise equally treated and harvested in parallel. We tested the hypothesis that Arabidopsis can recognize and respond differentially to insect species at the transcriptional level using a genome wide microarray. Transcriptional reprogramming was characterized using co-expression analysis in damaged and undamaged leaves at two times in response to mechanical wounding and four insect species. In all, 2778 (10.6%) of annotated genes on the array were differentially expressed in at least one treatment. Responses differed mainly between aphid and caterpillar and sampling times. Responses to aphids and caterpillars shared only 10% of up-regulated and 8% of down-regulated genes. Responses to two caterpillars shared 21% and 12% of up- and down-regulated genes, whereas responses to the two aphids shared only 7% and 4% of up-regulated and down-regulated genes. Overlap in genes expressed between 6h and 24h was 3-15%, and depended on the insect species. Responses in attacked and unattacked leaves differed at 6h but converged by 24h. Genes responding to the insects are also responsive to many stressors and included primary metabolism. Aphids down-regulated amino acid catabolism; caterpillars stimulated production of amino acids involved in glucosinolate synthesis. Co-expression analysis revealed 17 response networks. Transcription factors were a major portion of differentially expressed genes throughout and responsive genes shared most of the known or postulated binding sites.

Project description:In this experiment we aimed to decipher the transcription response of rice to the different cues of an herbivory attack from the chewing caterpillar Spodoptera frugiperda. We performed a herbivory mimic procedure on seedlings and applied mechanical wounding (WOS), application of herbivore oral secretions (OS) after wounding (WOS+) and dysbiotic OS (WOS-) on rice leaves. Project description:Plants are attacked by diverse herbivores and respond with manifold defense responses. To study transcriptional and other early regulation events of these plant responses, herbivory is often mimicked to standardize the temporal and spatial dynamics that vary tremendously for natural herbivory. Yet to what extent such mimics of herbivory are able to elicit the same plant response as real herbivory remains largely undetermined. We examined the transcriptional response of a new model plant to herbivory by a lepidopteran larva and to a commonly used herbivory mimic by applying the larvae’s oral secretions to standardized wounds. We designed a microarray for Solanum dulcamara and showed that the transcriptional response to real and to simulated herbivory by Spodoptera exigua overlapped moderately by about 40%. Interestingly, certain responses were mimicked better than others; 60% of the genes up-regulated but not even a quarter of the genes down-regulated by herbivory were similarly affected by application of oral secretions to wounds. While the regulation of genes involved in signaling, defense and water stress were mirrored well by the herbivory mimic, most of the genes related to photosynthesis, carbohydrate- and lipid metabolism were exclusively regulated by real herbivory. Thus, wounding and elicitor application decently mimics herbivory-induced defense responses but likely not the re-allocation of primary metabolites induced by real herbivory.

Project description:Tomato plants are submitted to a high diversity of herbivory pests, among them the leafminer Tuta absoluta, considered as one of the most important threat on the tomato worldwide production. In spite of its susceptibility to this pest, a better understanding of the tomato plant response to T. absoluta herbivory will help defining plant resistance traits and enlarging the range of possibilities for an efficient integrated pest management strategy. We analyzed the transcriptomic response in leaves of tomato (cv. Better Bush) submitted to the herbivory of T. absoluta larvae after 5h and 24h.

Project description:Tobacco is mainly consumed in two different forms (smoking and chewing) that vary in their composition and methods of intake. Despite being the leading cause of oral cancer, the molecular mechanisms resulting in malignancy upon tobacco exposure are yet to be fully elucidated. We therefore sought to compare the molecular alterations in oral keratinocytes exposed to smoke and chewing tobacco. OKF6/TERT1 cells were exposed to cigarette smoke condensate or chewing tobacco for progressively increasing durations (2, 4, 6 and 8 months). We employed a TMT-based quantitative proteomics approach to investigate the adverse effects of chronic cigarette smoke or chewing tobacco exposure in oral keratinocytes. LC/MS3 analysis resulted in the quantification of 5,342 proteins and 2,821 proteins in cigarette smoke and chewing tobacco exposed cells, respectively. Upstream regulator analysis indicates the involvement of distinct regulators in CSC exposed cells compared to STE exposed cells. In addition, exome sequencing revealed discrete genetic alterations in cells exposed to each insult. Current analysis defines a clear distinction in the molecular dysregulation in oral cells in response to different tobacco-based insults. Some of the proteins dysregulated in cigarette smoke or chewing tobacco exposed cells may serve as potential early detection biomarkers which could aid in stratification of patients based on tobacco usage history.

Project description:The practice of using chewing tobacco is common in certain socio-economic sections of southern Asia particularly in the Indian subcontinent. The molecular mechanism of smokeless tobacco which leads to malignancy is unclear. Chewing tobacco demonstrates a carcinogenic effect through chronic and not acute exposures. Using a cell line model, we studied the chronic effects of chewing tobacco on the proteome in normal oral keratinocytes (OKF6/TERT1). We carried out iTRAQ-based quantitative proteomic analysis of the untreated and chewing tobacco treated oral keratinocytes. LC-MS/MS analysis of this cell line pair resulted in the identification of 3,638 proteins of which 408 were found to be differentially expressed.
