Project description:In this experiment we measured the transcriptional response of tomato plants (cv “Money maker”) when attacked belowground by the nematode M. inognita and, subsequently, aboveground by different (and common for this crop) biotic agents. Three weeks-old plants were exposed to nematodes for 5 days. At the fifth day the terminal leaflet of one of the first two true leaves was infected with the Cauliflower Mosaic Virus, or with the pathogen, or with the potato aphid Macrosiphum euphorbiae, or with the CMV-infected M. euphorbiae. For each belowground/aboveground combination treatment a set of control plants that received only the aboveground treatment was prepared. The infected leaflets of 5 biological replicates, each consisting of 1 plant, were collected 1, 2, 3, 4, 5 and 6 days after the onset of the aboveground treatment and flash frozen in liquid nitrogen. A different set of plants was used for every time point. Corresponding leaves from plants that did not receive any aboveground treatment (control) were selected and sampled as described above. Three biological replicates were selected among the five for RNA isolation. Total RNA was sent for sequencing to BGI Hong Kong.

Project description:Background and Aims Plants modulate defence signalling networks in response to various biotic stresses via inter-organ communications. The root-mediated transmission of systemic acquired resistance (SAR) against soil-borne and air-borne plant pathogens from SAR-induced plants to neighbouring plants subjected to local chemical and pathogen treatments was evaluated. Methods The first two plants out of ten Nicotiana benthamiana seedlings were pre-treated with the SAR-triggering chemical benzothiadiazole (BTH). All ten seedlings were then challenged with two pathogenic bacteria, i.e. the root (bacterial wilt) pathogen Ralstonia solanacearum and the leaf (wildfire) pathogen Pseudomonas syringae pv. tabaci, at 7 d after SAR induction. Key Results Disease severity was noticeably lower in BTH-pre-treated plants than in the control. Surprisingly, two plants located next to BTH-treated plants exhibited reduced disease symptoms indicating that SAR signal transmission occurred through the root system. Determinant(s) secreted from the root system were search for and it was found that salicylic acid (SA) is a major molecule involved in SAR transmission through the root. Analysis of the expression of the defence-related genes N. benthamiana pathogenesis-related gene 1a (NbPR1a) and NbPR2 confirmed that BTH treatment elicited SAR via root-root transmission between plants. Plants with knock-down of the multiple resistance component SGT1 and SA biosynthesis-related gene ICS1 by Tobacco rattle virus-mediated virus-induced gene silencing exhibited a lack of root-mediated SAR transmission. The biological relevance of this finding was validated by challenge with the SAR-inducing avirulent pathogen P. syringae pv. syringae instead of BTH, which produced similar results. Conclusions Our findings demonstrated that SAR is transmissible through the root system from SAR-triggered plants to neighbouring plants.

Project description:Assessing the degree to which climate explains the spatial distributions of different taxonomic and functional groups is essential for anticipating the effects of climate change on ecosystems. Most effort so far has focused on above-ground organisms, which offer only a partial view on the response of biodiversity to environmental gradients. Here including both above- and below-ground organisms, we quantified the degree of topoclimatic control on the occurrence patterns of >1,500 taxa and phylotypes along a c. 3,000 m elevation gradient, by fitting species distribution models. Higher model performances for animals and plants than for soil microbes (fungi, bacteria and protists) suggest that the direct influence of topoclimate is stronger on above-ground species than on below-ground microorganisms. Accordingly, direct climate change effects are predicted to be stronger for above-ground than for below-ground taxa, whereas factors expressing local soil microclimate and geochemistry are likely more important to explain and forecast the occurrence patterns of soil microbiota. Detailed mapping and future scenarios of soil microclimate and microhabitats, together with comparative studies of interacting and ecologically dependent above- and below-ground biota, are thus needed to understand and realistically forecast the future distribution of ecosystems.

Project description:Background and aimsModern lettuce cultivars underperform under conditions of variable temporal and spatial resource availability, common in organic or low-input production systems. Information is scarce on the impact of below-ground traits on such resource acquisition and performance of field-grown lettuce; exploring genetic variation in such traits might contribute to strategies to select for robust cultivars, i.e., cultivars that perform well in the field, even under stress.MethodsTo investigate the impact of below-ground (root development and resource capture) on above-ground (shoot weight, leaf area) traits, different combinations of shoot and root growth were created using transplants of different sizes in three field experiments. Genetic variation in morphological and physiological below- and above-ground responses to different types of transplant shocks was assessed using four cultivars.ResultsTransplanting over-developed seedlings did not affect final yield of any of the four cultivars. Small transplant size persistently impacted growth and delayed maturity. The cultivars with overall larger root weights and rooting depth, "Matilda" and "Pronto," displayed a slightly higher growth rate in the linear phase leading to better yields than "Mariska" which had a smaller root system and a slower linear growth despite a higher maximal exponential growth rate. "Nadine," which had the highest physiological nitrogen-use efficiency (g dry matter produced per g N accumulated in the head) among the four cultivars used in these trials, gave most stable yields over seasons and trial locations.ConclusionsRobustness was conferred by a large root system exploring deep soil layers. Additional root proliferation generally correlates with improved nitrate capture in a soil layer and cultivars with a larger root system may therefore perform better in harsh environmental conditions; increased nitrogen use efficiency can also confer robustness at low cost for the plant, and secure stable yields under a wide range of growing conditions.

Project description:Herbivorous insects can influence grassland ecosystem functions in several ways, notably by altering primary production and nutrient turnover. Interactions between above- and belowground herbivory could affect these functions; an effect that might be modified by nitrogen (N) addition, an important global change driver. To explore this, we added above- (grasshoppers) and belowground (wireworms) insect herbivores and N into enclosed, equally composed, grassland plant communities in a fully factorial field experiment. N addition substantially altered the impact of above- and belowground herbivory on ecosystem functioning. Herbivory and N interacted such that biomass was reduced under above ground herbivory and high N input, while plant biomass remained stable under simultaneous above- and belowground herbivory. Aboveground herbivory lowered nutrient turnover rate in the soil, while belowground herbivory mitigated the effect of aboveground herbivory. Soil decomposition potential and N mineralization rate were faster under belowground herbivory at ambient N, but at elevated N this effect was only observed when aboveground herbivores were also present. We found that N addition does not only influence productivity directly (repeatedly shown by others), but also appears to influence productivity by herbivory mediated effects on nutrient dynamics, which highlights the importance of a better understanding of complex biotic interactions.

Project description:72 plants have been grown in 2 phytochambers for 30 days under control temperature (21°C/19°C) and short day conditions (8 hours light/16 hours darkness). After 30 days the plants were switched to long day conditions (16 hours light/8 hours darkness) and the temperature of one phytochamber was increased to 29°C/27° (heat chamber). Moreover,a heatplate was located in the control chamber and a coldplate was located in the heat chamber. 18 plants have been grown on the heatplate with a constant temperature of 29°C. 18 plants have been grown on the coldplate with a constant temperature of 22°C. After 9 days of the heat period leaf samples (end of night) were taken for micorarray analysis. After 10 days of heat, tuber samples were taken for microarray analysis.
