Project description:Phosphate-solubilizing bacteria (PSB) have the ability to dissolve insoluble phosphate and enhance soil fertility. However, the growth and mineral phosphate solubilization of PSB could be affected by exogenous soluble phosphate and the mechanism has not been fully understood. In the present study, the growth and mineral phosphate-solubilizing characteristics of PSB strain Burkholderia multivorans WS-FJ9 were investigated at six levels of exogenous soluble phosphate (0, 0.5, 1, 5, 10 and 20 mM). The WS-FJ9 strain showed better growth at high levels of soluble phosphate. The phosphate-solubilizing activity of WS-FJ9 reduced as the soluble phosphate concentration increased, as well as the production of pyruvic acid. Transcriptome profiling of WS-FJ9 at three levels of exogenous soluble phosphate (0, 5 and 20 mM) identified 446 differentially expressed genes, among which 44 genes were continuously up-regulated when soluble phosphate concentration increased and 81 genes were continuously down-regulated. Some genes related to cell growth were continuously up-regulated which would account for the better growth of WS-FJ9 at high levels of soluble phosphate. Genes involved in glucose metabolism, including glycerate kinase, 2-oxoglutarate dehydrogenase, and sugar ABC-type transporter were continuously down-regulated which indicates that metabolic channeling of glucose towards phosphorylative pathway was negatively regulated by soluble phosphate.

Project description:Phosphorus (P) is essential for metabolism and homeostasis of living organisms. However, a major source of P in animal feed comes in a form of inositol phosphate (InsPx) which is inefficiently used by monogastrics animals due to an intrinsic minimal activity of endogenous phytase. This study aims to understand molecular mechanisms of P utilization in quails using an mRNA-Seq analysis of high- and low-P utilization quails selected from discordant sib pairs from different families. The results of differentially expressed genes and pathways related to P utility should serve as an important knowledge for field applications aiming to improve P utility in animal production and to reduce source of P pollution into environment.

Project description:Intracellular signaling is conserved in eukaryotes to allow for response to extracellular signals and to regulate development and cellular functions. In fungi, inositol phosphate signaling has been shown to be involved in growth, sexual reproduction, and metabolic adaptation lacking, however, reports of mushroom forming fungi so far. In Schizophyllum commune, an inositol monophosphatase has been found up-regulated during sexual development. The enzyme is crucial for inositol cycling where it catalyzes the last step of inositol phosphate metabolism restoring the inositol pool from the monophosphorylated inositol monophosphate. We overexpressed the gene in this model basidiomycete and verified its involvement in cell wall integrity and intracellular trafficking. Strong phenotypes in mushroom formation and cell metabolism were evidenced by proteome analyses. In addition, altered inositol signaling was shown to be involved in tolerance towards cesium and zinc, and increased metal tolerance towards cadmium, associated with induced expression of kinases and repression of phosphatases within the inositol cycle. The presence of the heavy metals Sr, Cs, Cd, and Zn lowered intracellular calcium levels. We could develop a model integrating inositol signaling in the known signal transduction pathways governed by Ras, G-protein coupled receptors, cAMP and elucidate different roles in development.

Project description:The pathogenesis of primary hyperparathyroidism (I-HPT) and secondary hyperparathyroidism (II-PTH) remains to be elucidated. To characterize their pathophysiology, we investigated the effects of calcium and phosphate on cell proliferation and PTH release in an organ culture of parathyroid tissues. Dissected parathyroid tissues obtained from patients with I-HPT (adenoma) or II-PTH (nodular hyperplasia) were precultured on a collagen-coated membrane for 1-4 week. After exchanging the medium for one containing various concentrations of phosphate, PTH release and [3H]thymidine incorporation were studied. In contrast to dispersed parathyroid cells cultured in a monolayer, calcium decreased PTH release in a concentration-dependent manner in parathyroid tissues. Furthermore, when parathyroid tissues obtained from II-PTH were precultured for 1-4 weeks, PTH release and parathyroid cell proliferation were significantly increased in high-phosphate medium. These phosphate effects were also observed to a lesser extent in parathyroid tissues obtained from I-HPT, but there was no significant difference between I-HPT and II-HPT. Microarray analyses revealed that mRNA levels of PTH, CaSR, and VDR were well preserved, and several growth factors (e.g. TGF-beta1-induced protein) were abundantly expressed in II-PTH. Using organ cultures of hyperparathyroid tissues, in which PTH release and CaSR are well preserved for a prolonged period, we have demonstrated that phosphate stimulates parathyroid cell proliferation not only in II-PTH but also in I-HPT. Although the mechanism responsible for phosphate-induced cell proliferation remains to be elucidated, our in vitro findings suggest that both parathyroid tissues preserve to some extent a physiological response system to hyperphosphatemia as observed in normal parathyroid cells. Project description:The filamentous fungus Mucor circinelloides VI 04473 (indicated by MC1 in these data) was exposed to treatments known to trigger the accumulation of lipids in fungal cells, in order to investigate the genomic basis of this trait. Treatments consisted of different levels of calcium and inorganic phosphorus substrate in the growth media used for cultivation: For calcium, the two levels were calcium present in a concentration of 0.1 g/L (Ca1), and absence of calcium (Ca0). For phosphorus, there were three different levels: The reference concentration P1 (phosphate salts in a concentration of 7 g/L KH2PO4 and 2 g/L Na2HPO4), four times the reference concentration (P4), and half the reference concentration (P0.5). The combination of the different levels of calcium and phosphorus resulted in six different growth media, indicated in these data by numbers _1 - _6: 1 = Ca1P1, 2 = Ca1P4, 3 = Ca1P0.5, 4 = Ca0P1, 5 = Ca0P4, 6 = Ca0P0.5. There were 3-4 replicates per unique growth media, indicated by R1-R4 . The biomass was washed and harvested after 5 days of cultivation. RNA was extracted from the biomass and sequenced, followed by analysis of the transcriptome (genes differentially expressed between treatments).
