Two-dimensional standing wave total internal reflection fluorescence microscopy: superresolution imaging of single molecular and biological specimens.

Project description:The development of high resolution, high speed imaging techniques allows the study of dynamical processes in biological systems. Lateral resolution improvement of up to a factor of 2 has been achieved using structured illumination. In a total internal reflection fluorescence microscope, an evanescence excitation field is formed as light is total internally reflected at an interface between a high and a low index medium. The <100 nm penetration depth of evanescence field ensures a thin excitation region resulting in low background fluorescence. We present even higher resolution wide-field biological imaging by use of standing wave total internal reflection fluorescence (SW-TIRF). Evanescent standing wave (SW) illumination is used to generate a sinusoidal high spatial frequency fringe pattern on specimen for lateral resolution enhancement. To prevent thermal drift of the SW, novel detection and estimation of the SW phase with real-time feedback control is devised for the stabilization and control of the fringe phase. SW-TIRF is a wide-field superresolution technique with resolution better than a fifth of emission wavelength or approximately 100 nm lateral resolution. We demonstrate the performance of the SW-TIRF microscopy using one- and two-directional SW illumination with a biological sample of cellular actin cytoskeleton of mouse fibroblast cells as well as single semiconductor nanocrystal molecules. The results confirm the superior resolution of SW-TIRF in addition to the merit of a high signal/background ratio from TIRF microscopy.

On the effects of 30 GHz sinusoidal wave exposure on glioblastoma organoids

Project description:In this study, we evaluated the effects of milli-meter waves on structured Glioblastoma organoids to assess the possibility of therapeutic applications. The exposure setup was completely developed, and the dosimetry carried on based on both numerical and experimental activities. Our results showed that continuous milli-meter waves at 30.5 GHz affect cell proliferation and apoptosis, thus not affecting the differentiation status of the organoids composing of GBM cells. By applying the power level of 0.1 W (RMS), we obtain a synergistic effect with the chemotherapeutic Temozolomide in terms of GBM cell death. All these data open the way to an interventional window in which to treat GBM cells (i.e. with TMZ) exploiting 30.5 GHz CW exposure for potential therapeutic purposes thus improving GBM future management, which remain extremely difficult so far. Our investigation sheds light on the characterization of possible bio-effects of MMW (30.5 GHz CW) on GBM organoids highlighting cell and molecular responses in a relevant 3D tumor model.

Project description:On December 15, 2009, a very high wave crest was recorded by a local camera at the CNR-ISMAR oceanographic tower, 15?km offshore Venice in the Northern Adriatic Sea (Italy). The height of the estimated crest elevation appears well beyond the value (1,25·Hs) commonly used to identify a wave as freak. We document the wave event with a full description of the corresponding met-ocean conditions and related measurements, of which we provide a critical analysis.

BV6 induces an early wave of gene expression via NF-κB and AP-1 and a second wave via TNFα/TNFR1 signaling

Project description:Smac mimetics are considered as promising cancer therapeutics, but little is yet known about how they alter gene expression. In this study we used an unbiased genome-wide expression array to investigate Smac mimetic BV6-induced gene regulation in breast cancer cell lines. Kinetic analysis revealed that BV6 alters gene expression in two waves. The first wave primarily involves NF-κB- and AP-1 families of transcription factors, while the second wave largely depends on tumor necrosis factor receptor 1 (TNFR1) signaling. Interestingly, disrupting auto-/paracrine tumor necrosis factor-α (TNFα)/ (TNFR1) signaling by knockdown of TNFR1 strongly attenuates the BV6-induced second wave of gene expression and upregulation of many pathways including NF-κB signaling, apoptosis and immune signalling, but not MAPK signaling pathways. Consistently, BV6 stimulates phosphorylation of cJun, a marker of MAPK cascade activation, irrespective of the presence or absence of the TNFα blocking antibody Enbrel. We show here in a comprehensive overview that BV6-induced gene expression in breast cancer cells takes place in a time- as well as TNFR1-dependent manner.

Forced wave motion with internal and boundary damping.

Project description:A d'Alembert-based solution of forced wave motion with internal and boundary damping is presented with the specific intention of investigating the transient response. The dynamic boundary condition is a convenient method to model the absorption and reflection effects of an interface without considering coupled PDE's. Problems with boundary condition of the form [Formula: see text] are not self-adjoint which greatly complicates solution by spectral analysis. However, exact solutions are found with d'Alembert's method. Solutions are also derived for a time-harmonically forced problem with internal damping and are used to investigate the effect of ultrasound in a bioreactor, particularly the amount of energy delivered to cultured cells. The concise form of the solution simplifies the analysis of acoustic field problems.

Calibrating Evanescent-Wave Penetration Depths for Biological TIRF Microscopy.

Project description:Roughly half of a cell's proteins are located at or near the plasma membrane. In this restricted space, the cell senses its environment, signals to its neighbors, and exchanges cargo through exo- and endocytotic mechanisms. Ligands bind to receptors, ions flow across channel pores, and transmitters and metabolites are transported against concentration gradients. Receptors, ion channels, pumps, and transporters are the molecular substrates of these biological processes, and they constitute important targets for drug discovery. Total internal reflection fluorescence (TIRF) microscopy suppresses the background from the cell's deeper layers and provides contrast for selectively imaging dynamic processes near the basal membrane of live cells. The optical sectioning of TIRF is based on the excitation confinement of the evanescent wave generated at the glass/cell interface. How deep the excitation light actually penetrates the sample is difficult to know, making the quantitative interpretation of TIRF data problematic. Nevertheless, many applications like superresolution microscopy, colocalization, Förster resonance energy transfer, near-membrane fluorescence recovery after photobleaching, uncaging or photoactivation/switching as well as single-particle tracking require the quantitative interpretation of evanescent-wave-excited images. Here, we review existing techniques for characterizing evanescent fields, and we provide a roadmap for comparing TIRF data across images, experiments, and laboratories.
