Estimating the ecological water levels of shallow lakes: a case study in Tangxun Lake, China.

Project description:Water level management is an effective tool for the ecological restoration of shallow lakes. In this study, we developed an ecologically-based approach to estimate the monthly suitable ecological water levels (EWLs). This approach took both the lake topographic features and aquatic plants' growth characteristics into account. The aquatic vegetation coverage was used to characterize the degree of the lake ecological restoration. The relationship between water level and vegetation coverage was established. We chose the Tangxun Lake as a testbed, and the recommended lowest EWL was 16.6?m, as the minimum threshold for water level regulations. The results revealed that the predicted vegetation coverage decreased with the rise of water level during the germination period (February and March). To achieve the vegetation coverage goal of 30% and 50%, the lake's water levels must be lowered to 17.1?m and 16.8?m respectively during germination. The EWLs were recommended to be low in spring and high in summer, which was matched with the natural water level regimes. The proposed approach can provide a reliable reference for water level regulation of shallow lakes especially the lakes with insufficient data.

Benthic-pelagic coupling drives non-seasonal zooplankton blooms and restructures energy flows in shallow tropical lakes.

Project description:Zooplankton blooms are a frequent phenomenon in tropical systems. However, drivers of bloom formation and the contribution of emerging resting eggs are largely unexplored. We investigated the dynamics and the triggers of rotifer blooms in African soda-lakes and assessed their impact on other trophic levels. A meta-analysis of rotifer peak densities including abundances of up to 6 × 105 individuals L-1 demonstrated that rotifer bloom formation was uncoupled from the food environment and the seasonality of climatic conditions. A time series with weekly sampling intervals from Lake Nakuru (Kenya) revealed that intrinsic growth factors (food quality and the physicochemical environment) significantly affected rotifer population fluctuations, but were of minor importance for bloom formation. Instead, rotifer bloom formation was linked to sediment resuspension, a prerequisite for hatching of resting-eggs. Population growth rates exceed pelagic birth rates and simulations of rotifer dynamics confirmed the quantitative importance of rotifer emergence from the sediment egg-bank and signifying a decoupling of bloom formation from pelagic reproduction. Rotifer blooms led to a top-down control of small-sized algae and facilitated a switch to more grazing-resistant, filamentous cyanobacteria. This shift in phytoplankton composition cascaded up the food chain and triggered the return of filter-feeding flamingos. Calculations of consequent changes in the lake's energy budget and export of aquatic primary production to terrestrial ecosystems demonstrated the large potential impact of nonseasonal disturbances on the functioning of shallow tropical lakes.

Project description:Longitudinal bone growth depends upon the execution of an intricate series of cellular activities by epiphyseal growth plate chondrocytes. In order to better understand these coordinated events, microarray analysis was used to compare gene expression in chondrocytes isolated from the proliferative and hypertrophic zones of the avian growth plate. In this experiment we compared pooled samples of proliferative and hypertrophic chondrocytes isolated from the chick growth plate. The expression of 745 genes was found to differ 3-fold or greater at the 0.05 level of probability. Experiment Overall Design: We examined 8 samples using arrays: 4 from proliferative and 4 from hypertrophic chondrocytes.

Epigenome analysis of normal skin and hypertrophic scar samples

Project description:Hypertrophic scarring (HS) is characterized by excessive extracellular matrix deposition, matrix metalloprotein gene activation, and fibroblast invasive growth. However, the methylation level of hypertrophic scarring is poorly understood. Genome wide DNA methylation profiling of normal skin and hypertrophic scar. The Illumina Infinium Methylation EPIC BeadChip (850K) was used to obtain DNA methylation profiles across approximately 853,307 CpGs in liquid based scar samples. Samples included 6 normal skin, and 6 hypertrophic scar.
