Project description:In the present study, we investigated whether the acetylation of H3K56, known as relevant for C. albicans virulence, might affect the Candida-macrophages interaction. Therefore, by the mean of the non-specific Hst3 inhibitor nicotinamide (NAM), we evaluated whether the alteration of H3K56 acetylation would affect the secretion of soluble factors by C. albicans. In particular, we tested the effect of the two different Candida growth-conditioned media (with and without NAM) on murine macrophages at the morphological and functional levels. In addition, we analyzed transcriptome modifications and genome enrichment of H3K56ac in the same experimental conditions.

Project description:Candida albicans is a normally commensal yeast which becomes pathogenic to human hosts under some conditions and specifically in immune compromised individuals. This contributes to a global health crisis of fungal disease. This yeast can morphologically adapt to the environment by reversibly developing hyphae which allows for traversing of tissue. Histone deacetylase 1 (Hda1) is involved in this transition as it is established that the deletion of the associated gene stunts this morphological switch. In Saccharomyces cerevisiae this protein complexes with two similarly named proteins: Hda2 and Hda3. Here we evaluate Candida albicans strains with mutations in the genes that code for these proteins. The strains are grown in yeast (YPD 30 °C) and hyphae (RPMI 37 °C) -inducing conditions for 90 minutes prior to RNA extraction. After alignment by HISAT2, the homozygous mutants are then individually compared to wildtype (matching growth conditions) using DESeq2.

Transcriptional profiling of Candida albicans SC5314, hgc1Δ and cph1Δ/efg1Δ in yeast-and hyphae-inducing conditions at two time points.

Project description:Goal of this study was to determine metabolic adaptation processes in C. albicans associated to hyphal morphogenesis. Accessory to the metabolic profiling the corresponding transcriptome was investigated. To identify media-specific and general adaptation three different hyphae stimuli were used (M199 pH 7.4, Human serum and N-Aectylglucosamine) were used and compared again two respective yeast conditions (SD and M199 pH 4). Two filamentation-affected mutant strains were included - lacking either two central regulators of filamentation (cph1Δ/efg1Δ) or a downstream effector (hgc1Δ) - for the identification of specfic morphotype-dependent metabolic adaptation in repsonse to hyphae stimuli. Two different time points (90 min and 240 min after transition to test medium) were investigated to define the progression of the examined adaptation processes.

H3K56ac profiling in Candida albicans by high throughput sequencing

Project description:In C. albicans the sirtuin Hst3p is responsible for removing the acetyl group of the Lys 56 of the H3 histone which is particularly abundant in yeasts and contributes to fungal genome integrity and virulence. Here we identified the H3K56ac patterns across Candida albicans genome in cells maintained for 28 h in YPD at 25°C in the presence or absence of 10 mM nicotinamide (used in this study as Hst3p inhibitor). Chromatin immunoprecipitation was carried out using an anti-H3K56ac antibody, whereas input samples were taken before addition the antibody. Two biological replicates were performed for each condition.
