Project description:Overall, the study aims at obtaining a comprehensive picture of the African malaria mosquito, Anopheles gambiae, transcriptome using high-coverage RNA-seq of sexed whole-insect samples. This experiment focuses on transcriptomes of females subjected to physiological stress. The samples include adult females collected at different time intervals subject to sub-optimal relative humidity, after exposure to sublethal dose of deltamethrin, or following blood-feeding. Females kept at standard laboratory conditions without exposure to stressors, were collected at the same time intervals as control samples.

Transcriptome analysis of virgin and mated Anopheles gambiae and Anopheles albimanus

Project description:We custom-built a bioinformatics pipeline to search for 20E-modifying enzymes in the accessory glands of Anopheles gambiae males, searching for ecdysteroid kinases (EcK), ecdysone oxidases (EO), and ecdysteroid-phosphate phosphatases (EPP). To this end, we generated RNAseq datasets of different An. gambiae tissues dissected from virgin and mated females and males, and produced similar datasets for Anopheles albimanus, a South American species that does not synthetize and transfer ecdysteroids during mating. These analyses led to the identification of one candidate EPP and two potential EcKs (EcK1 and EcK2), which we demonstrated are involved in the activity of a male-specific oxidized ecdysteroid (3D20E). We further determined that 3D20E is specifically produced by the An. gambiae male accessory glands and is transferred to females during copulation, where it triggers a series of post-mating responses.

Transcriptomic analysis of tissues and embryos from control and triglyceride lipase 2 (TL2)-deficient Anopheles gambiae females.

Project description:We explored the role of triglyceride (TAG) mobilization in the reproductive success of female Anopheles gambiae after a blood meal. Silencing triglyceride lipase 2 (TL2) in An. gambiae reduced egg production and abrogates fertility. Embryos from AgTL2-depleted females develop normally but fail to hatch and are characterized by reduced levels of lipids and key metabolites, including carbohydrates, Tricarboxylic acid (TCA) intermediates, paralleled by increased levels of purine and pyrimidine breakdown products, signifying possible cellular death. To obtain a better understanding of the mechanism underlying embryonic lethality in AgTL2-deficient females, we generated RNA-Seq datasets of tissues and embryos from control and TL2-knockdown females at different time points after blood feeding (for tissues) and oviposition. These analyses revealed that while at earlier time points there is almost no difference, transcriptomic profiles at the later time point of 38 hours after laying were profoundly distinct between both groups. Specifically, pathways involved in neuropeptide signaling, G-protein coupled receptor signaling, and eclosion hormone synthesis were repressed in TL2-knockdown condition while hallmarks of cellular stress were significantly up-regulated. More importantly, pathways involved in de novo lipid synthesis and transport were significantly up-regulated, possibly to compensate for reduced levels of maternally inherited lipids.

A Microarray-based Analysis of Transcriptional Compartmentalization in the Alimentary Canal of Anopheles gambiae

Project description:Although the basic anatomical sub-divisions of the larval mosquito gut were established several decades ago, information regarding their exact physiological roles is rather scarce. Several studies have reported differences between larval gut compartments in various morphological and physiological aspects. Unfortunately, the fragmentary and incomplete nature of this information makes it hard to establish clear links to the specific and/or unique physiological roles of each gut region. In this study, we attempted to identify transcripts significantly enriched in each gut section of the 4th instar Anopheles gambiae larva. We took advantage of a commercially-available microarray platform containing ~14,000 different An. gambiae transcripts (Affymetrix GeneChip® Plasmodium/Anopheles array, Affymetrix Inc., Santa Clara, CA). Based on the information obtained, we discuss the putative physiological roles of each major compartment of the larval gut, and identify potential new targets for larval control strategies. Experiment Overall Design: We attempted to identify transcripts significantly enriched in each gut section of the 4th instar Anopheles gambiae larva. Signal intensities of transcripts from each gut section were compared to those of a matching group of whole 4th instar larvae using t-tests. Transcripts showing an intensity value two-fold or higher (p < 0.05) in each gut section as compared to whole larvae were considered to be significantly enriched.
