Project description:This study was designed to address key questions concerning the use of alternative protein sources for animal feeds and addresses aspects such as their nutrient composition and impact on gut function. We used casein (CAS), spray dried porcine plasma (SDPP), soybean meal (SBM), and yellow meal worm (YMW) as protein sources. We have investigated the use of intestinal organoids as a model to test the effects of different protein sources on the intestinal epithelium. Mouse enteroids were exposed to different undigested protein sources (4% w/v, viz. soybean meal, SBM; casein, CAS; spray dried plasma protein, SDPP; and yellow meal worm, YMW) or DMEM as a control. Microarrays were used to detail the global gene expression.

Male C57BL/6J mice were stratified according to bodyweight and litter of origin into six dietary treatment groups. These mice were fed for four weeks with semi-synthetic diets containing one of the following protein sources. At the end of four weeks, mice were sacrificed by euthenesia and ileal tissue samples (scrapings) were collected for RNA extraction and hybridization on Affymetrix microarrays. Soybean meal (SBM) diet group served as reference to make comparisons with other experimental diets.

The transcriptional response of intestinal mucosal tissue (jejunum and ileum) served as parameters for the local response. Growing pigs (BW 35 kg/approx. 10 weeks) were fed with experimental diets containing a single, common or new protein sources viz. soybean meal (SBM), black soldier fly larvae (BSF), spray dried blood plasma (SDPP), rapeseed meal (RSM), and wheat gluten meal (WGM) over a period of 4 weeks.

Project description:Peptides and protein hydrolysates are promising alternatives to substitute chemical additives as functional food ingredients. In this study, we present a novel approach for producing a potato protein hydrolysate with improved emulsifying and foaming properties by data-driven, targeted hydrolysis. Based on previous studies, we selected 15 emulsifier peptides derived from abundant potato proteins, which were clustered based on sequence identity. Through in silico analysis, we determined that from a range of industrial proteases (Neutrase (Neut), Alcalase (Alc), Flavorzyme (Flav) and Trypsin (Tryp)), Tryp was found more likely to release peptides resembling the target peptides. After applying all proteases individually, hydrolysates were assayed for in vitro emulsifying and foaming properties. No direct correlation between degree of hydrolysis and interfacial properties was found. Tryp produced a hydrolysate (DH=5.4%) with the highest (P<0.05) emulsifying and foaming abilities, good stabilities, and high aqueous solubility. Using LC-MS/MS, we identified >10,000 peptides in each hydrolysate. Through peptide mapping, we show that random overlapping with known peptide emulsifiers is not sufficient to quantitatively describe hydrolysate functionality. While Neut hydrolysates had the highest proportion of peptides with target overlap, they showed inferior interfacial activity. In contrast, Tryp was able to release specifically targeted peptides, explaining the high surface activity observed. While modest yields and residual unhydrolyzed protein indicate room for process improvement, this work shows that data-driven, targeted hydrolysis is a viable, interdisciplinary approach to facilitate hydrolysis design for production of functional hydrolysates from alternative protein sources.

Project description:Seaweeds are attracting substantial interest as novel sources of sustainable food protein, as they are established sources of industrial hydrocolloids and have reasonable protein content. In this study we investigate the protein composition and quality of a seaweed protein extract (SPE) from Gigartina radula obtained as a side-stream from industrial carrageenan production. The SPE displayed low (<2%), but pH-dependent, aqueous solubility due to the harsh (heat and extreme pH) employed during extraction. Solubility was improved using buffer and detergent to facilitate proteomic characterization by quantitative LC-MS/MS. Proteomics revealed that the SPE was dominated by proteins related to light harvest and particularly rich in phycobiliproteins (44% relative molar abundance), where phycoerythrin was the most abundant (28%). Based on subcellular localization of identified proteins, the extraction method was evaluated as good for release of cellular protein. The SPE was found to be rich in essential amino acids (36-41%) and particularly in branched chain amino acids (22-24%) and thereby a potential source of nutritional food protein. Using bioinformatic prediction and structural modelling, we found that abundant proteins in the SPE contained high potential, novel emulsifier peptides with amphiphilic properties required to stabilize an oil/water interface. Based on this study, Gigartina could serve as a good candidate for extraction of sustainable and nutritious food protein and possibly be further processed to obtain a hydrolysate with good emulsifying properties for use in foods.
