Examine strain and gender associated gene expression in Schistosoma japonicum

Project description:We utilised a 22,575 feature custom oligonucleotide DNA microarray designed from public domain databases of schistosome expressed sequence tags to explore differential gene expression between the Philippine (SJP) and Chinese (SJC) strains of S. japonicum, and between male and female S. japonicum. We found that 593, 664 and 426 probes were differentially expressed between the two geographical strains when we compared mix sexed adults, male worms and female worms. Additionally, the study revealed that 1,163 male- and 1,016 femaleassociated probes were differentially expressed in SJP whereas 1,047 male- and 897 female-associated probes were differentially expressed in SJC. Sex-dependent and strain-dependent gene liver gene expression

Project description:A series of dual-channel gene expression profiles obtained using Agilent Mouse TOE 75k microarrays was used to examine the sex-dependent differences in gene expression across three outbred mouse strains, 129J x Black Swiss, 129J x BALB/c, and ICR. This series is comprised of samples obtained from 3 pools of randomly chosen independent cDNA samples of male and female 129Jx BALB/c mice and 3 randomly chosen independent cDNA samples of male and female 129J x Black Swiss mice and 5 randomly chosen independent cDNA samples of male and female ICR mice. The ICR mice were randomly distributed into three and two member pools for each sex and two of the 5 samples for each sex were independently hybridized separately as well. Comparison of the sex-specific genes for the three outbred strains generated lists of strain-dependent and strain-independent sex-specific genes. Keywords: sex response and strain response

Project description:In this study we examined the genomic transcriptional signature of mating costs in female fruitflies in which mating rate was elevated 3-4 fold. We conducted 4-fold biological replication and randomly sampled cohorts of females before the major mortality effects of the elevated mating appeared (to avoid the potentially biasing effects of lineage selection). We then assayed differential gene expression, in two different body parts (abdomen versus head+thorax), using genome wide microarrays.

Transcriptional responses in various normal mouse tissues after 24 h following 4 MV photon beam irradiation using three partial body irradiation set-ups

Project description:Female BALB/c nude mice (n=3/group) were subjected to partial body irradiation under anesthesia using 2 Gy external photon (4 MV, nominal) irradiation. In group A, the collum (i.e. the thyroid) was irradiated; in group B, thorax+abdomen were irradiated, and in group C, collum+thorax+abdomen were irradiated. The control group (n=5) was anesthetized but not irradiated. At 24 h after treatment, the kidneys, liver, lungs, spleen, and thyroid were excised, flash-frozen, and stored at -80°C. Total RNA was extracted from homogenized tissue samples (kidney cortex and kidney medulla were treated separately) and subjected to expression analysis using RNA microarray technology.

Gene expression analysis comparing a selected strain of rainbow trout with superior growth and immune performance to a commercial reference strain during infection with infectious hematopoietic necrosis virus

Project description:Through 8 generations of selection, our group has developed a strain of rainbow trout that exhibits high growth rates on an economically and environmentally sustainable all plant protein, high-soy diet. The selected strain also shows superior performance in bacterial and viral disease challenges compared to commercial trout strains, and even a strain specifically selected over many generations for viral and bacterial disease resistance. The selection criteria was strictly focused on performance on plant-based diets, and therefore the physiological mechanisms responsible for the strain’s superior disease resistance remain unresolved. To better characterize the physiological mechanism behind the superior performance of the selected strain we compared the intestinal gene expression of the select strain to that of a commercial control line of trout during an experimental viral infection using the CSF 220-90 isolate of Infectious Hematopoietic Necrosis virus (IHNv). At 65 days post hatch, all female rainbow trout from the select and commercial strain were stocked separately into four 150L tanks each, at a density of 45 fish per tank. For both strains of trout, three tanks of fish were experimentally infected with IHNv by static bath and one tank was mock challenged by static bath (Sham). Sampling was conducted at 4 days post challenge (dpc) (Early Infection) and 20 dpc (Late/Recovered Infection). Two intestinal samples from each tank were pooled and two pools from each tank were utilized for RNAseq library preparation. Infections were mild with mortality rates near 50% over the 21 day trial and no significant difference in mortality rates between the two strains of trout. Reads from the RNAseq samples were quantified at the transcript level prior to evaluating differential transcript usage and differential gene expression between the strains of trout, infection time points, and disease status.
