Project description:RNA seq project designed to understand the role of pesticide on transription of mRNA in the testis. We treated adult female mice with chlordecone with doses 0 (control), 100 Xg/kg/day and 1 mg/kg/day during gestation period E6- E15. Male born from Chlordecone- treated mice were crossed for a three generations. The testes from F3 generations were taken for mRNA extraction. For mRNA extraction we used columb. Three biological replicates used for each condition . We hypothesize that chlordecone cause changes in mRNA due to transgenerational effect of pesticides (Skinner et al, Science 2005). The changes in expression of certain genes can help us understand the effect of pesticides on many tissues. Number of sample :9 samples. We require Paired End sequencing and number of reads equal or more than 25M tags, strand specific sequencing

To test our hypothesis, we exposed mice to chlordecone during embryonic development and we analyzed F1 female progeny epigenetic mark H3K4me3 in ovaries. We performed ChIP using anti-H3K4me3 commercial antibody (Merck Millipore, 07-473). DNA-protein complexes were immunoprecipitated, eluted and then DNA were purified.

Moreover, the changes could be transferred to a subsequent generation via gametes. In this project we studied whether the pesticide chlordecone could cause the developmental effects and cause the transgenerational inheritance in outbred mouse strain. Mice were treated during development at dose 100 µg/kg/day and the progeny of exposed mice were crossed for three generations. The testes of the third generations males were used for anti-H3K4me3 ChiP-seq analysis. We used two biological replicates for each condition.

Chemical Treatments for Mobilizing Arsenic from Contaminated Aquifer Solids to Accelerate Remediation.

Project description:Arsenic is a prevalent contaminant at US Superfund sites where remediation by pump and treat systems is often complicated by slow desorption of As from Fe and Al (hydr)oxides in aquifer solids. Chemical amendments that either compete with As for sorption sites or dissolve Fe and Al (hydr)oxides can increase As mobility and improve pump and treat remediation efficiency. The goal of this work was to determine optimal amendments for improving pump and treat at As contaminated sites such as the Vineland Chemical Co. Superfund site in southern New Jersey. Extraction and column experiments were performed using As contaminated aquifer solids (81 ± 1 mg/kg), site groundwater, and either phosphate (NaH(2)PO(4)·H(2)O) or oxalic acid (C(2)H(2)O(4)·2H(2)O). In extraction experiments, phosphate mobilized between 11% and 94% of As from the aquifer solids depending on phosphate concentration and extraction time (1 mM-1 M; 1-24 h) and oxalic acid mobilized between 38 and 102% depending on oxalic acid concentration and extraction time (1-400 mM; 1-24 h). In column experiments, phosphate additions induced more As mobilization in the first few pore volumes but oxalic acid was more effective at mobilizing As overall and at lower amendment concentrations. At the end of the laboratory column experiments, 48% of As had been mobilized from the aquifer sediments with 100 mM phosphate and 88% had been mobilized with 10 mM oxalic acid compared with 5% with ambient groundwater alone. Furthermore, simple extrapolations based on pore volumes suggest that chemical treatments could lower the time necessary for clean up at the Vineland site from 600 a with ambient groundwater alone to potentially as little as 4 a with 10 mM oxalic acid.

The prostate of the first (F1) and third generation males (F3) were used for RNA-seq-analysis. We used three biological replicates for each condition.

Iron electrolysis-assisted peroxymonosulfate chemical oxidation for the remediation of chlorophenol-contaminated groundwater.

Project description:BackgroundElectrolysis with an iron anode is a novel way to provide ferrous activators for chemical oxidation. The objective of this study is to evaluate the performance of peroxymonosulfate (PMS) for chlorophenol destruction when compared with H2O2 and persulfate (PS), and to see whether the electrolysis mode facilitates the buildup of conditions that favor the activation of PMS and removal of chlorophenols.ResultsFerrous species can effectively activate the PMS over a wide pH range. In comparison with H2O2 and PS, PMS is less sensitive to the solution's pH and possesses stronger oxidation capability at alkaline pHs. The optimal molar ratio of PMS to Fe(II) activator is 1:1 for the destruction of 2,4-dichlorophenol (2,4-DCP). The column experiments show that an acidic zone developed downstream from the anode is favorable to maintain ferrous ions and subsequent activation of PMS. The reactivity of the PMS can be manipulated by varying the electrical currents, and the process demonstrates effectiveness for treating organic contaminants. 2,4-DCP contaminated groundwater shows decreased biotoxicity after the chemical oxidation process without considering the residual PMS.ConclusionsIron electrolysis-assisted peroxymonosulfate chemical oxidation can effectively treat the 2,4-dichlorophenol and mixtures of organic contaminants. This process can be engineered as an in situ chemical oxidation method for groundwater remediation.
