Project description:OBJECTIVE: External environmental stressors and internal factors significantly impact the skin by causing inflammation, aging, reduced immunity, and other adverse responses. Dead Sea Water (DSW) is well known for its dermatological benefits and has been widely used in dermatological therapy and skincare (e.g. psoriasis, atopic dermatitis and photoaging). However, the anti-aging and rejuvenating effects of DSW and the related biological pathways involved, which have attracted increasing attention, remain to be fully understood. The aim of this study is to investigate the anti-aging and rejuvenating effects and to probe to explore the related potential biological mechanisms of DSW under different environmental conditions. METHODS: Using in vitro human dermal cells and reconstructed skin models, the effects of DSW treatment on the extracellular matrix (ECM) components and the morphological changes at the dermal-epidermal junction (DEJ) were investigated. RNA sequencing (RNA-seq) analysis of human dermal fibroblast models after DSW treatment was performed to explore the potential mechanisms of action of DSW under normal and UV stress conditions. RESULTS: The novel findings in this work present the biological functions of DSW, including procollagen-1 and elastin secretion, hemidesmosome increase, and the epidermal basal cell regeneration. In addition, GO, KEGG, and Reactome analysis results reveal the activation of pathways related to ion transmembrane transporter activity, ECM component biosynthesis, senescence-associated secretory phenotype (SASP), DNA repair, and autophagy, which are associated with the anti-aging activities of DSW. CONCLUSION: Our work provides new perspectives for understanding the anti-aging and rejuvenating effects and mechanisms of DSW. The new findings also provide a theoretical basis for further development of age-related strategies.

Project description:Skeletal aging and disease are associated with a misbalance in the opposing actions of osteoblasts and osteoclasts that are responsible for maintaining the integrity of bone tissues. Here, we show through detailed functional and single-cell genomic studies that intrinsic aging of bona fide mouse skeletal stem cells (SSCs) alters bone marrow niche signaling and skews bone and blood lineage differentiation leading to fragile bones that regenerate poorly. Aged SSCs have diminished bone and cartilage forming potential but produce higher frequencies of stromal lineages that express high levels of pro-inflammatory and pro-resorptive cytokines. Single-cell transcriptomic studies reveal a distinct population of SSCs in aged mice that gradually outcompete their younger counterparts in the bone marrow niche. While systemic exposure to a youthful circulation through heterochronic parabiosis reduced local expression of inflammatory cytokines, it did not reverse the diminished osteochondrogenic activity of aged SSCs and was insufficient to improve bone mass and skeletal-healing parameters in aged mice. Hematopoietic reconstitution of aged mice with young hematopoietic stem cells (HSC) also did not improve bone integrity and repair. We find that deficient bone regeneration in aged mice could only be reversed by the local application of a combinatorial treatment that re-activates aged SSCs and simultaneously abates crosstalk to hematopoietic cells favoring an inflammatory milieu. This treatment expanded aged SSC pools, reduced osteoclast activity, and enhanced bone healing to youthful levels. Our findings provide mechanistic insight into the complex, multifactorial mechanisms underlying skeletal aging and offer new prospects for rejuvenating the aged skeletal system.

Project description:The styrene-butadiene-styrene (SBS)-modified asphalt pavement has been in growing demand in the road construction field owing to its workable mechanical property and temperature durability. This paper prepared a penetrative rejuvenator (PR) with waste cooking oil (WCO) and emulsified asphalt, then applied PR on SBS copolymers to investigate its aging and rejuvenating effects in an asphalt binder. After a thin film oven test (TFOT) and ultraviolet (UV) aging of SBS copolymers, Fourier transform infrared (FTIR) spectra were used to analyse the aged copolymers' chemical structure. Moreover, both aged and rejuvenated SBS copolymers were added into a fresh asphalt binder to get two kinds of modified asphalt binders, namely, MAAC (modified by aged copolymer) and MARC (modified by rejuvenated copolymer). Aiming to analyse the monomer effect of SBS copolymers in the asphalt binder, the rheological characteristic with dynamic shear rheometer (DSR), chemical structure with FTIR and physical properties with penetration, soft point and ductility tests were investigated using MAAC and MAAC samples. The results showed that rejuvenated SBS copolymer could improve MAAC's viscoelasticity, but from FTIR spectral analysis, PR resulted in no chemical changes to SBS copolymers. A tough coat which made MAAC of higher stiffness was observed on the copolymer surface after thermal treatment. UV caused evidently negative effects on SBS copolymer because of accelerating oxidation by ozone, which brought about high possibility of cracks during servicing periods of asphalt pavement. In addition, MAAC was inferior in both rheological and physical properties, which reflected the significance and necessity in consideration of alleviating SBS copolymer aging in field.
