Project description:The lack of optimal models to evaluate novel agents is delaying the development of effective immunotherapies against human breast cancer (BC). In this prospective open label study, we applied neoadjuvant intratumoral immunotherapy with empty cowpea mosaic virus-like particles (eCPMV) to 11 companion dogs diagnosed with canine mammary cancer (CMC), a spontaneous tumor resembling human BC. We found that two neoadjuvant intratumoral eCPMV injections resulted in tumor reduction in injected tumors in all patients and in noninjected tumors located in the ipsilateral and contralateral mammary chains of injected dogs. Tumor reduction was independent of clinical stage, tumor size, histopathologic grade, and tumor molecular subtype. RNA-seq-based analysis of injected tumors indicated a decrease in DNA replication activity and an increase in activated dendritic cell infiltration in the tumor microenvironment. Immunohistochemistry analysis demonstrated significant intratumoral increases in neutrophils, T and B lymphocytes, and plasma cells. eCPMV intratumoral immunotherapy demonstrated antitumor efficacy without any adverse effects. This novel immunotherapy has the potential for improving outcomes for human BC patients.

An Abscopal Effect on Lung Metastases in Canine Mammary Cancer Patients Induced by Neoadjuvant Intratumoral Immunotherapy with Cowpea Mosaic Virus Nanoparticles and Anti-Canine PD-1.

Project description:Neoadjuvant intratumoral (IT) therapy could amplify the weak responses to checkpoint blockade therapy observed in breast cancer (BC). In this study, we administered neoadjuvant IT anti-canine PD-1 therapy (IT acPD-1) alone or combined with IT cowpea mosaic virus therapy (IT CPMV/acPD-1) to companion dogs diagnosed with canine mammary cancer (CMC), a spontaneous tumor resembling human BC. CMC patients treated weekly with acPD-1 (n = 3) or CPMV/acPD-1 (n = 3) for four weeks or with CPMV/acPD-1 (n = 3 patients not candidates for surgery) for up to 11 weeks did not experience immune-related adverse events. We found that acPD-1 and CPMV/acPD-1 injections resulted in tumor control and a reduction in injected tumors in all patients and in noninjected tumors located in the ipsilateral and contralateral mammary chains of treated dogs. In two metastatic CMC patients, CPMV/acPD-1 treatments resulted in the control and reduction of established lung metastases. CPMV/acPD-1 treatments were associated with altered gene expression related to TLR1-4 signaling and complement pathways. These novel therapies could be effective for CMC patients. Owing to the extensive similarities between CMC and human BC, IT CPMV combined with approved anti-PD-1 therapies could be a novel and effective immunotherapy to treat local BC and suppress metastatic BC.

Label-free Proteomics Reveals that Cowpea Severe Mosaic Virus Transiently Suppresses the Host Leaf Protein Accumulation During the Compatible Interaction with Cowpea (Vigna unguiculata [L.] Walp.)

Project description:Viruses are important plant pathogens that threaten diverse crops worldwide. Diseases caused by Cowpea severe mosaic virus (CPSMV) have drawn attention because of the serious damages they cause to economically important crops including cowpea. This work was undertaken to quantify and identify the responsive proteins of a susceptible cowpea genotype infected with CPSMV, in comparison with mock-inoculated controls, using label-free quantitative proteomics and databanks, aiming at providing insights on the molecular basis of this compatible interaction. Cowpea leaves were mock-inoculated or inoculated with CPSMV and 2 and 6 days later proteins were extracted and analyzed. More than 3000 proteins were identified and 75 and 55 of them differentially accumulated in response to CPSMV, at 2 and 6 DAI, respectively. At 2 DAI, 76% of the proteins were down-represented and 24% upaccumulated. However, at 6 DAI, 100% of the identified proteins were up-accumulated. Thus CPSMV transiently suppresses the synthesis of proteins involved particularly in the redox homeostasis, protein synthesis, defense, stress, RNA/DNA metabolism, signaling, and other functions, allowing viral invasion and spread in cowpea tissues. It is expected that identification of differentially accumulated proteins and their interactions advance our understanding on how a susceptible cowpea genotype responds to CPSMV infection.

Project description:Cowpea mosaic virus is a potent intratumoral immunotherapy agent that has shown promise in preclinical studies and canine cancer trials with tumor- and tissue-agnostic efficacy. As we move towards the clinic, it is imperative to investigate combination strategies that synergize to further improve the potency of the approach. Here, we combined CPMV with the clinically approved chemotherapeutic agent oxaliplatin. CPMV's ability to recruit and activate naive immune cells synergized with oxaliplatin's ability to induce immunogenic cell death in the ID8-Defb29/Vegf-A ovarian and B16F10 melanoma murine cancer models with an increase of median survival of 57.7% and 162.2%, respectively. The combination therapy outperformed the CPMV or oxaliplatin monotherapy, and achieved a percent difference in tumor burden of 26.1% and 170.6% in the ID8-Defb29/Vegf-A ovarian and B16F10 melanoma models, respectively. Immunofluorescence staining of treated tumor sections elucidated the role of damage associated molecular patterns (calreticulin and HMGB1), innate immune cells (myeloid cells - likely neutrophils, NK cells, and macrophages), and regulatory T cells (Tregs) as a function of the treatment regimen. Overall, our proposed combination therapy modulated the dormant tumor microenvironment which resulted in effective tumor cell death. This study demonstrates the potential for clinical combination of chemotherapy and CPMV intratumoral immunotherapy.
