Project description:Skin aging is a complex biological process that is influenced by multiple factors, including environmental and genetic factors. Ultraviolet (UV) irradiation can cause short-term sun damage, such as erythema and edema, as well as chronic damage, which accelerates skin aging and produces skin wrinkles and skin laxity. Salvianolic acid B (Sal-B) is the most abundant and bioactive water-soluble extractant from the well-known herbal medicine--Salvia Miltiorrhiza in Chinese traditional medicine. Many studies have found that Sal-B exerts powerful antioxidant and anti-inflammatory effects. Therefore, we speculate that salvianolic acid B may have potential therapeutic value in the aspect of anti-photoaging.

Expression data in human oral mucosa fibroblasts (hOFs), dermal fibroblasts, and hOFs-derived iPS cells

Project description:To define the characteristics of human oral mucosa fibroblasts (hOFs), we analyzed the gene expression of hOFs compared with that of human dermal fibroblasts (hDFs), and that of hOF-derived induced pluripotent stem cells (hOF-iPSCs). In this dataset, we found that 5,738 probes and 5672 probes were differently expressed in hOFs and hDFs respectively, compared with those in hOF-iPSCs at more than 2-fold levels. In contrast, only 434 probes are differently expressed between hOFs and hDFs.

Project description:Salvianolic acid B (Sal-B) is the most abundant and bioactive water-soluble compound in Salvia miltiorrhiza. Studies have shown that salvianolic acid B increases the mRNA expression of adipogenic transcription factors, including PPARγ, C/EBPα and PPARα, in 3T3-L1 preadipocytes to increase glucose uptake and mitochondrial respiration, reduce glycerol release and promote adipocyte differentiation. However, the effect of Sal-B on graft fat survival and its specific mechanism have not been investigated. So we dicided using h-ADSCs to see its transcriptomes changing after Sal-B addition.

Differentially expressed genes induced with or without UVB stimulation in mice

Project description:Skin cancer is the most prevalent cancer in humans, especially in the United States, Australia, and New Zealand. Australia and New Zealand are the two countries with the highest rates of skin cancer in the world, about four times higher than the United States, the United Kingdom and Canada. According to statistics, one American dies of skin cancer every hour. Studies have shown that ultraviolet radiation is the main cause of skin cancer, and ultraviolet rays are mainly divided into UVA, UVB and UVC according to wavelength, UVA and UVB can cause DNA damage, and UVB is the main factor that induces skin cancer. UVB is primarily a direct damage to cellular DNA and generally includes the formation of pyrimidine dimers (CPDs) and 6-4 photoproducts (6-4PPs). UVB can also cause mutations in tumor suppressor genes such as p53, ptch, and ras. These bases the mutation will promote the activation of related signaling pathways, thereby inducing the production of tumors.In this study, we will use gene chip technology to screen out UVB-sensitive genes, and then select the genes of the UVB-sensitive GPCR family from these genes, and further use PCR for verification, so as to identify UVB-sensitive GPCRs, which will provide a basis for further experimental research.

ER71/ETV2-mediated direct reprogramming of human dermal fibroblasts (HDFs) into endothelial-like cells

Project description:Samples 1-6: HDFs transduced with ER71/ETV2 were sorted on KDR expression at day 7. The isolated KDR+ cells together with human umbilical venous endothelial cells (HUVECs) and untransduced HDFs were subjected to genomic gene expression profiling. A significant number of genes related with vessel development and angiogenesis was significantly upregulated in KDR+ cells, compared to control HDFs. These findings strongly argue that ER71/ETV2 directly reprograms human fibroblasts to functional endothelial-like cells, which could be useful for disease investigation as well as autologous cell therapy. Samples 7-12: HDFs transduced with ER71/ETV2 were sorted on KDR expression at day 7. The isolated KDR+ cells were further cultured up to day 93. The further cultured cells together with human umbilical venous endothelial cells (HUVECs) and untransduced HDFs were subjected to genomic gene expression profiling.
