Sex differences in the molecular signature of the developing mouse hippocampus

Project description:A variety of neurological disorders, including Alzheimer’s disease, Parkinson’s disease, major depressive disorder, dyslexia and autism, are differentially prevalent between females and males. To better understand the possible molecular basis for the sex-biased nature of neurological disorders, we measured both mRNA and protein in the hippocampus of female and male mice at 1, 2, and 4 months of age with RNA-sequencing and mass-spectrometry respectively. Differential expression analyses identify 2699 genes that are differentially expressed between animals of different ages. 198 transcripts are differentially expressed between females and males at one or more ages. The number of transcripts that are differentially expressed between females and males is greater in adult animals than in younger animals. Additionally, we identify 69 transcripts that show complex and sex-specific patterns of temporal regulation across all ages, 8 of which are heat-shock proteins. We also find a modest correlation between levels of mRNA and protein in the mouse hippocampus (Rho = 0.53). This study adds to the substantial body of evidence for transcriptomic regulation in the hippocampus during postnatal development. Additionally, this analysis reveals sex differences in the transcriptome of the developing mouse hippocampus, and further clarifies the need to include both female and male mice in longitudinal studies involving molecular changes in the hippocampus.

MICRO-RNA AS BIOMARKERS IN POMPE DISEASE

Project description:We studied microRNAs as potential biomarkers for Pompe disease (PD). We analyzed microRNA expression by small RNA-seq in tissues from the PD mouse model at two different ages (3 and 9 months), and in plasma from 6 PD patients. In the PD mouse we found 211 microRNAs that were differentially expressed in gastrocnemii and 66 in heart, with a different pattern of expression at different ages. In a preliminary analysis in plasma from 6 PD patients 55 microRNAs were differentially expressed.

Next generation sequencing contributes to the quantitative analysis of AGEs stimulated HK-2 cell transcriptome

Project description:We sequenced the HK-2 gene expression profile stimulated by ages by high-throughput RNA analysis. Select Ctrl group and ages group, Differentially expressed genes (DEGs) were analyzed. Under the screening condition of fold change greater than or equal to 2, P < 0.05. We obtained 284 up-regulated genes and 193 down-regulated genes. It can be seen that there are obvious gene differences between Ctrl group and ages group (Fig. 3a). Gene ontology analysis (go) was completed. Through goseq software, the biological process of quantitative distribution of rich differential genes was intuitively reflected (BP), molecular function (MF) and cell composition (CC). In addition, in order to explore the impact of ages on cell pathways, we used the Kyoto Encyclopedia of biology to analyze the significantly rich metabolic pathways. There are 221 differentially expressed genes in Ctrl group and ages group. A total of 276 pathways have been established, of which 46 pathways have p < 0.05, and the first 20 pathways are shown in Figure 2C.
