Project description:Type 2 diabetes (T2D) and obesity are strongly associated with low natriuretic peptide (NP) plasma levels and impaired NP signaling with a down-regulation of NP guanylyl cyclase receptor-A (GCA) in skeletal muscle and adipose tissue. However, no study has so far provided evidence for a causal link between ANP/GCA deficiency and T2D development. Here, we show that both ANP and GCA deficiency in mice promotes metabolic disturbances and prediabetes. In GCA haploinsufficient mice, skeletal muscle insulin resistance is associated with altered mitochondrial function and impaired endurance running capacity. Muscle RNA-seq analyses reveal down-regulation of various gene sets related to mitochondrial protein complexes and translation. Despite normal mitochondrial content, GCA haploinsufficient mice exhibit increased proton leak and reduced content of mitochondrial oxidative phosphorylation (OXPHOS) proteins. Using various clinical studies and cohorts, we further show that GCA is related to various metabolic traits in T2D and positively correlates with markers of oxidative capacity in human skeletal muscle. Altogether, these results indicate that ANP/GCA signaling controls muscle mitochondrial integrity and oxidative capacity in vivo and plays a causal role in the development of T2D.

Impairment of cardiac natriuretic peptide signaling develops the excessive cardiac hypertrophy during lactation period

Project description:Two natriuretic peptides, atrial natriuretic peptide (ANP) and brain natriuretic peptide (BNP) act through the common receptor, guanylyl cyclase-A (GC-A) to lower blood pressure, induce diuresis/natriuresis and dilate blood vessels. Recently, we discovered that the excessive cardiac hypertrophy accompanied with cardiac dysfunction was induced in the lactating natriuretic peptide receptor 1 (Npr1, which encodes GC-A)-deficient mice. To clarify the cause of lactation-induced cardic hypertrophy in Npr1-/-, we performed the gene expressions analysis using nulliparous (NP) or postpartum lactating wild-type (Npr1+/+) and Npr1-/- mice. Numerous genes were altered in the postpartum lactating period both in Npr1+/+ and Npr1-/-. Additionally, the involvement of inflammatory responce in the cardiac hypertrophy in lactating-Npr1-/- mice was clarified bythe gene ontology analysis.

Atrial Fibrillation After Surgery for Colorectal Cancer

Project description:This study will report the incidence of atrial fibrillation after elective colorectal cancer resection in the over 65 age group. This will be used to validate a risk model for the development of post-operative atrial fibrillation. Eligible patients will undergo electrocardiogram based screening for atrial fibrillation, as well as brain natriuretic peptide tests prior to surgery. They will undergo 24 hour holter monitor prior to surgery, and at 30 and 90 days following surgery. The primary outcome will be occurrence of atrial fibrillation within 90 days of surgery. Secondary outcomes include quality of life change, use of hospital services for atrial fibrillation, and complications of atrial fibrillation. This will be used to validate the pre-existing model for prediction of atrial fibrillation.

Cardiac specific deletion of natriuretic peptide receptor A induces different myocardial expression of circular RNA and mRNA involved with metabolism in mice

Project description:Atrial natriuretic peptide (ANP) and brain natriuretic peptide (BNP) are not only important biological markers, but also regulators of cardiac functions. The natriuretic peptide A receptor (NPRA), also called NPR1 or guanylyl cyclase A (GC-A), binds with ANP or BNP ligand and fulfils transmembrane signalling transduction by elevating the intracellular levels of cGMP. However, the comprehensive effects and mechanisms downstream to NPRA are still largely to be elucidated. Here, the cardiac expressing profiles of mRNA in the mice with myocardium-specific deletion of NPRA were analyzed. It was found that differently expressed mRNAs were detected and proved by Gene Ontology (GO) and pathway analysis to be mainly related to the metabolic process. Moreover, circular RNAs (circRNAs) were scrutinized, and subsequently a possible regulatory network consisting of circRNAs- MicroRNAs (miRNAs) -mRNAs was predicted and constructed by ceRNA (competing endogenous RNA) analysis. In conclusion, NPRA plays possible roles in cardiac metabolism, which might be mediated by circRNAs via endogenous competition mechanisms.

We will measure this by collecting tissue, blood and urine samples, energy expenditure measurements using the metabolic cart, and a DXA to assess the amount of bone, fat and muscle in the body from a single study visit.

Effects of supervised physical activity on endurance capacity in colorectal cancer patients undergoing ambulatory Chemotherapy

Project description:Interventions: Group 1: With beginning of the chemotherapy a standardized endurance training of 15/30 min will be accomplished within 24 weeks. 2 sessions per week will be passed on a bicycle ergometer (30min) and 3 sessions will be passed by a walking or Nordic walking program (15min). The first 10 sessions are supervised to ensure controlled environment. From week 3 patients exercise only the 2 sessions on bicycle ergometer under supervision. At the beginning the intensity is defined as 50% of the maximum endurance capacity and increases during the next 10 sessions on 70%. The exercise program accords to an intensive endurance training. Group 2: patients receive general recommendations for regular physical activity, including 3 times subjectively moderate endurance training per week. primary outcome is the physical fitness, measured by spiroergometry before and after intervention (24 weeks after baseline), and at the 3 year follow-up. Identification of natriuretic peptide receptor A (NPR-A) expression signatures in podocytes in vivo reveals baseline control of protective pathways including Protein S

Project description:Natriuretic peptide receptor-A (NPR-A) is the principal receptor for the natriuretic peptides ANP and BNP. Targeted deletion of NPR-A in mouse glomerular podocytes significantly enhances renal injury in vivo in the DOCA-salt experimental model. It was therefore hypothesized that natriuretic peptides exert a direct protective effect on glomerular barrier integrity through activation of NPR-A and modulation of gene expression patterns in podocytes. RNA-seq analysis revealed a total of 158 DEGs with 81 downregulated DEGs and 77 upregulated DEGs in Npr1 deficient podocytes. Among the downregulated genes were protein S and semaphorin 3G, which are known to have a protective effect in podocytes. Protein S was also expressed in and secreted from isolated human glomeruli. GO enrichment analysis revealed that the upregulated DEGs in NPR-A deficient podocytes were associated with cell migration and motility. In line, BNP significantly decreased podocyte outgrowth from cultured glomeruli.
