Project description:Tumours in Min mice arise as a result of aberrant b-catenin activity which then acts as a transcriptional activator and repressor of certain target genes. Some of these target genes will essential to the development of tumours. By comparing expression profiles of tumour tissue with normal intestinal mucosa, it is hoped that some of these genes will be identified. By including both small and large intestine tumours (ie two different tissue types) and by testing mice in different genetic backgrounds, it should be possible to identify genes which are generically changed during tumour development. Once identified, the genes need to be tested for their role -if any- in tumourigenesis through in-vitro experiments and genetic engineering of mice.

Gene expression profiling in paired normal, apparently normal and breast tumour tissues

Project description:The main objective of the study was to identify potential diagnostic and follow up markers along with therapeutic targets for breast cancer. We performed gene expression studies using the microarray technology on 65 samples including 41 breast tumours [24 early stage, 17 locally advanced, 18 adjacent normal tissue [paired normal] and 6 apparently normal from breasts which had been operated for non-malignant conditions. All the samples had frozen section done – tumours needed to have 70% or more tumour cells; paired normal and apparently normal had to be morphologically normal with no tumour cells.

Project description:mRNA expression profiling of pancreatic cancer, comparing adjacent normal tissue, patient tumour and first generation patient derived xenograft tumours Fresh tumour samples for human pancreatic adenocarcinoma patients were implanted in SCID mice. 70% of these pancreatic ductal adenocarcinoma patients grew as PDX tumours, confirmed by histopathology. Frozen samples from F1 PDX tumours could be later successful passaged in SCID mice to F2 PDX tumours. The human origin of the PDX was confirmed using human specific antibodies; however, the stromal component was replaced by murine cells. Cell lines were successfully developed from three PDX tumours. RNA was extracted from 8 PDX tumours and where possible, corresponding primary tumour and adjacent normal tissues. mRNA profiles of tumour vs F1 PDX and normal vs tumour were compared by Affymetric microarray analysis
