Targeting Cyclin Y Inhibits STAT3 Activation to Enhance Radiosensitivity via Down-regulation of RRM2 in Lung Cancer

Project description:Purpose: Radioresistance is a main reason for the recurrence and metastasis of lung cancer. Cyclin Y is involved in normal physiological processes such as cell proliferation, growth and metabolism, as well as in the development of certain cancers. However, the function and mechanism of Cyclin Y in lung cancer are still unclear.Methods: Lung cancer cell lines were subjected to Cyclin Y knockdown using small interfering RNA (siRNA), and their growth and proliferation were assessed in vitro. Comet assay, γH2AX/Rad51 foci formation assay and clonogenic cell survival assay were performed to assess DNA damage and the DNA repair capacity. SgRNA transfection was applied to construct cell lines with stable Cyclin Y knockdown. Xenograft mouse models were used to validate the findings in vivo. RNA sequencing (RNA-seq) analysis was conducted to clarify the downstream target genes of Cyclin Y. Western blotting was employed to measure protein expression.Results: Cyclin Y was overexpressed in lung cancer, correlating with poor prognosis. Cyclin Y knockdown inhibited cell growth and proliferation, increased DNA damage, inhibited DNA damage repair and increased radiosensitivity in vitro and in vivo. RNA-seq analysis was used to reveal RRM2 as a downstream target gene of Cyclin Y. Cyclin Y depletion down-regulated the protein and mRNA levels of RRM2, leading to reduced STAT3 phosphorylation. Rescue experiments demonstrated that the effect of Cyclin Y on lung cancer was mediated partially by RRM2.Conclusions: We revealed for the first time that targeting Cyclin Y inhibits tumorigenesis and promotes radiosensitivity in lung cancer partially through the RRM2-STAT3 signaling pathway.

Cyclin K interacts with β-catenin to induce Cyclin D1 expression and facilitate tumorigenesis and radioresistance in lung cancer

Project description:Rationale: Radioresistance remains the major cause of local relapse and distant metastasis in lung cancer. However, the underlying molecular mechanisms remain poorly defined. This study aimed to investigate the role and regulatory mechanism of Cyclin K in lung cancer radioresistance. Methods: Expression levels of Cyclin K were measured by immunohistochemistry in human lung cancer tissues and adjacent normal lung tissues. Cell growth and proliferation, neutral comet and foci formation assays, G2/M checkpoint and a xenograft mouse model were used for functional analyses. Gene expression was examined by RNA sequencing and quantitative real-time PCR. Protein-protein interaction was assessed by immunoprecipitation and GST pull-down assays. Results: We report that Cyclin K is frequently overexpressed and correlates with poor prognosis in lung cancer patients. Functionally, we demonstrate that Cyclin K depletion results in reduced proliferation, defective G2/M checkpoint and enhanced radiosensitivity in lung cancer. Mechanistically, we reveal that Cyclin K interacts with and promotes the stabilization of β-catenin, thereby upregulating the expression of Cyclin D1. More importantly, we show that Cyclin D1 is the major effector that mediates the biological functions of Cyclin K in lung cancer. Conclusions: These findings suggest that Cyclin K positively modulates the β-catenin/Cyclin D1 axis to promote tumorigenesis and radioresistance in lung cancer, indicating that Cyclin K may represent a novel attractive biomarker for lung cancer radiotherapy.

Chimeric RNA RRM2-C2orf48 plays an oncogenic role in the development of NNK-induced lung cancer

Project description:Chimeric RNAs have been used as biomarkers and therapeutic targets for multiple types of cancers. However, less attention has been paid to their mechanism of action in neoplasia. Here, we report that chimeric RNA RRM2-C2orf48 promotes 4-(methyl nitrosamine)-1-(3-pyridinyl)-1-butanone (NNK)-induced lung cancer. We found that RRM2-C2orf48 had higher expression levels in lung cancer cell lines and in lung cancer tissues from 74 patients than in normal cells and tissues, respectively. And its expression level was correlated with smoking. Next, we demonstrate that the expression level of RRM2-C2orf48 could increase after treatment with 200 mg/L NNK for 24 h. Overexpressing RRM2-C2orf48 accelerated the process of NNK-induced lung cancer and promoted cell growth. Meanwhile, suppressing RRM2-C2orf48 inhibited cell growth. Finally, we identified miR-219a-2-3p as a potential target of RRM2-C2orf48. In summary, RRM2-C2orf48 accelerated the process of NNK-induced lung cancer and miR-219a-2-3p may be involved in this process.
