Project description:Marine ingredients can contain relatively high levels of arsenic. The main aim of this study is to assess the hepatic toxicity of arsenic species in Atlantic salmon (Salmo salar) in vitro. Primary hepatocytes were isolated from six male Atlantic salmon and exposed for 48 h to establish dose-response curves for arsenite (AsIII) (0.1-3 μM) and for arsenosugar (AsSug) (5-250 μM). Regarding arsenite, results obtained indicated that the dose between 0.1 and 1 represented a marked threshold for effect. Very few genes were differentially expressed (N=5, p-adjust < 0.1) following AsIII 0.1 µM treatment compared to AsIII 1µM (N=2338 p-adjust < 0.01). Similar was observed for AsSug exposure where exposure to 50 µM resulted in 195 DEGs (p-adjust < 0.1) vs 6899 (p-adjust < 0.01) following AsSug 250µM exposure

Transcriptional response of Atlantic salmon (Salmo salar) after primary versus secondary exposure to Infectious Salmon Anemia Virus (ISAV)

Project description:Following an infection with a specific pathogen, the acquired immune system of many teleostean fish, including salmonids, is known to retain a specific memory of the infectious agent, which protects the host against subsequent infections. For example Atlantic salmon (Salmo salar), which have survived an infection with a low-virulence infectious salmon anemia virus (ISAV) isolate are less susceptible against subsequent infections with high-virulence ISAV isolates. A greater understanding of the mechanisms and immunological components involved in this acquired protection against ISAV is fundamental for the development of efficacious vaccines and treatments against this pathogen. To better understand the immunity components involved in this observed resistance, we have used an Atlantic salmon DNA microarray and RT-qPCR assays to study the global gene expression responses of preexposed Atlantic salmon (fish having survived an infection with a low-virulence ISAV isolate) during the course of a secondary infection with a high-virulence ISAV isolate

Salmon liver proteomics - Liver proteome response of pre-harvest Atlantic salmon following exposure to elevated temperature

Project description:Atlantic salmon production in Tasmania (Southern Australia) occurs near the upper limits of the species thermal tolerance. Summer water temperatures can average over 19°C over several weeks and have negative effects on performance and health. Liver tissue exerts important metabolic functions in thermal adaptation. With the aim of identifying the mechanisms underlying liver plasticity in response to chronic elevated temperature in Atlantic salmon, label-free quantitative shotgun proteomics was used to explore quantitative protein changes after 43 days of exposure to elevated temperature. A total of 277 proteins were differentially (adjusted p-value <0.05) expressed between the control (15°C) and elevated (21°C) temperature treatments. As predicted identified by Ingenuity pathway analysis (IPA), transcription and translation mechanisms, protein degradation via the proteasome, and cytoskeletal components were down-regulated at elevated temperature. In contrast, an up-regulated response was predicted identified for NRF2-mediated oxidative stress, endoplasmic reticulum stress, and amino acid degradation. The proteome response was paralleled by reduced fish condition factor and hepato-somatic index at elevated temperature . The present study provides further evidence of the interplay among different cellular machineries in a scenario of heat-induced energy deficit and oxidative stress, and refines present understanding of how Atlantic salmon cope with chronic exposure to temperature near the upper limits of thermal tolerance.

Project description:The Atlantic salmon (Salmo salar) genome contains 10 chitinase encoding genes, but little is known about the function of these chitinases. Three of the chitinase genes have previously been shown to be expressed in the stomach tissue of Atlantic salmon. In the current study we show that the protein products of these genes, the family 18 glycoside hydrolase (GH18) chitinases, Chia.3, Chia.4 and Chia.7 are secreted into the stomach mucosa and are amongst the most abundant proteins in this matrix.
