Project description:The biological functions of circadian clock on growth and development have been well elucidated in model plants, while its regulatory roles in crop species, especially the roles on yield-related traits are poorly understood. Here, we characterize the core clock gene CCA1 homoeologs in wheat and studied their biological functions in seedling growth and spike development. TaCCA1 homoeologs exhibit typical diurnal expression patterns which are positively regulated by rhythmic histone modifications (H3K4me3, H3K9ac and H3k36me3). TaCCA1s are preferentially located in the nucleus and tend to form both homo- and heterodimers. TaCCA1 overexpression (TaCCA1-OE) transgenic wheat plants show disrupted circadian rhythmicity coupling with reduced chlorophyll and starch content, as well as biomass at seedling stage, also decreased spike length, grain number per spike and grain size at the ripening stage. Further studies using DNA affinity purification followed by deep sequencing (DAP-seq) indicates that TaCCA1 preferentially binds to sequences similar to “evening elements” (EE) motif in the wheat genome, particularly genes associated with photosynthesis, carbon utilization and auxin homeostasis, and decreased transcriptional levels of these target genes are observed in TaCCA1-OE transgenic wheat plants. Collectively, our study provides novel insights into a circadian-mediated mechanism of gene regulation to coordinate photo synthetic and metabolic activities in wheat, which is important for optimal plant growth and crop yield formation.

Project description:The developmental switch from skotomorphogenesis to photomorphogenesis is critical for the survival and growth of plants, but its regulatory mechanism remains unclear. Here, we report that the steroid hormone brassinosteroids (BRs) play crucial roles in the transition from skotomorphogenesis to photomorphogenesis by regulating chlorophyll biosynthesis to promote the greening of etiolated seedlings upon light exposure. Seedlings of BR-deficient mutant det2-1 accumulated excess protochlorophyllide when grown in darkness, resulting in photo-oxidative damage upon exposure to light. Conversely, the gain-of-function mutant bzr1-1D suppressed the protochlorophyllide-accumulated phenotype of det2-1, thereby promoting greening of etiolated seedlings. Genetic analysis indicated that phytochrome-interacting factors (PIFs) were required for BZR1-promoted seedlings greening. Furthermore, we revealed that the GROWTH REGULATING FACTOR 7 (GRF7) and GRF8 were induced by BZR1 and PIF4 to repress the chlorophyll biosynthesis and promote seedling greening. Suppression the functions of GRFs by overexpressing microRNA396a (miR396a) caused the high-accumulated photochlorophyllide in darkness and more serious photobleach upon light exposure. Additionally, BZR1, PIF4 and GRF7 interact with each other and precisely regulate the expression of chlorophyll biosynthetic genes. Our findings revealed an essential role of brassinosteroid in promoting seedling development and survival during the critical initial emergence of seedlings from subterranean darkness to sunlight.

Project description:Indica rice seedlings of IR64 variety were grown hydroponically for 7-days in a culture room with a daily photoperiodic cycle of 14h light and 10h dark. Seedlings were incubated in 0.1% dimethyl sulfoxide (control) or 50 micromolar solutions of indole-3-acetic acid (IAA treatment) and benzyl aminopurine (BAP treatment) for 1h and 3h. Equal amounts of 1h and 3h samoles were pooled for each treatment before RNA isolation. Project description:The japonica rice variety Nipponbare was grown under hydroponic condition for 10-days (untreated seedling sample). Seedlings were exposed to once stress conditions for 24 hours (stress treated sample). Seedlings were exposed to one of the following for 24 h: cold stress, incubation at 10 °C; salt stress by adding 150 mM sodium chloride to the planter box; osmotic stress by adding 260 mM mannitol to the planter box; and drought stress by adding 25% polyethylene glycol 6000 to the planter box. Total RNAs were prepared from each sample using an RNeasy Midi Kit (Qiagen, Tokyo, Japan) and the mRNAs were purified with an Oligotex-dT30 Kit (Takara, Shiga, Japan). Purified mRNA was amplified and labeled and we hybridized the 1-mg fluorescent liner amplified Cy3- and Cy5-labeled cRNAs (each cRNA was 500 ng) to the NIAS RICE 22K oligonucleotide array ver1 according to the manufacturer. Project description:In Arabidopsis mature seeds, the onset of the embryo-to-seedling transition is nonautonomously controlled, being blocked by endospermic abscisic acid (ABA) release under unfavorable conditions. Mature embryos lack an impermeable cuticle, unlike seedlings, consistent with their endospermic ABA uptake capability. Seedling cuticle formation occurs after germination rather than during embryogenesis. Mature endosperm removal prevents seedling cuticle formation and seed reconstitution by endosperm grafting onto embryos shows that the endosperm promotes seedling cuticle development. Grafting different endosperm and embryo mutant combinations, together with biochemical, microscopy and mass spectrometry approaches, reveals that endospermic release of Tyrosyl Sulfate Transferase (TPST)-sulfated CIF2 and PSY1 peptides promotes seedling cuticle development. Endosperm-deprived embryos produced nonviable seedlings bearing numerous developmental defects, in a manner unrelated to embryo nourishment, all restored by exogenously provided endosperm. Hence, seedling establishment is nonautonomous, requiring the mature endosperm.
