Project description:Bumblebees (Hymenoptera: Apidae) are important pollinating insects that play pivotal roles in crop production and natural ecosystem services. To achieve a comprehensive profile of accessible chromatin regions and provide clues for all possible regulatory elements in the bumblebee genome, we did ATAC-seq for Bombus terrestris samples derived from its four developmental stages: egg, larva, pupa, and adult, respectively. The sequencing reads of ATAC-seq were mapped to B. terrestris reference genome, and its accessible chromatin regions were identified and characterized using bioinformatic methods. Our study will provide important resources not only for uncovering regulatory elements in the bumblebee genome, but also for expanding our understanding of bumblebee biology.

Project description:The goal of this study is to determine the feasibility of CEA assessments at home using (automated) capillary sampling in patients in the follow-up after treatment for colorectal cancer.
The main questions it aims to answer are:
* To determine the success rate of capillary sampling at home by the patient
* To assess reliability and satisfaction of (automated) capillary CEA measurements Participants will be asked to perform automated capillary sampling and lancet capillary sampling at home twice after regular check-up visits in the hospital, with an interval of 3-6 months in between. During this hospital visit, a CEA measurement in blood sampled by venipuncture will be performed to act as a reference for the CEA measurements in (automated) capillary blood to be sampled at home.
Reliability of CEA measurements will be assessed for automated capillary and lancet capillary sampling compared to venipuncture.
Satisfaction in terms of patient reported outcomes (pain, burden, ease of use, and preference) will be evaluated.

Project description:In this series of experiments, we wanted to study the transcriptional responses of plants to different levels of water limitation. For mild drought stress, we controlled water potential (a scientific concept for dryness of soil) by using an automated watering system. This system adds water to soil based on the pF values reported by a pF sensor in soil. pF is a classical and widely used index of water potential that was first defined by Schofield (1935). For severe stress conditions, we withheld watering or even dried plants on a lab bench. Project description:Is there a correlation between miRNA diversity and levels of organismic complexity? Exhibiting extraordinary levels of morphological and developmental complexity, insects are the most diverse animal class on earth. Their evolutionary success was in particular shaped by the innovation of holometabolan metamorphosis in endopterygotes. Previously, miRNA evolution had been linked to morphological complexity, but astonishing variation in the currently available miRNA complements of insects made this link unclear. To address this issue, we sequenced the miRNA complement of the hemimetabolan Blattella germanica and reannotated that of two other hemimetabolan species, Locusta migratoria and Acyrthosiphon pisum, and of four holometabolan species, Apis mellifera, Tribolium castaneum, Bombyx mori and Drosophila melanogaster. Our analyses show that the variation of insect miRNAs is an artefact mainly resulting from poor sampling and inaccurate miRNA annotation, and that insects share a conserved microRNA toolkit of 65 families exhibiting very low variation. For example, the evolutionary shift toward a complete metamorphosis was accompanied only by the acquisition of three and the loss of one miRNA families.

Project description:In flowers of Asteraceae, yellow flavonols bearing an additional hydroxyl group at positions 6 or 8 (ring A) contribute to petal UV-absorbing pigmentation patterns, which play a crucial role in attracting pollinating insects. Understanding the biogenesis of these special flavonols requires the identification of any specific enzyme involved in these incorporations of extra hydroxyl groups on the quercetin ring A. To this aim, flavonol-bearing biotinylated probes have been designed and synthesized to explore their ability to selectively capture target proteins or biosynthetic enzymes under oxidative activation. These probes demonstrate the ability to capture several flavonoid enzymes from Rudbeckia and Tagetes microsomes and allow the identification of uncharacterized candidates for novel flavonoid enzymes.

Project description:Female whiteflies are used to inoculate the susceptible and resistant tomato genotypes as follows. Whiteflies are caged with TYLCV-infected tomato plants for a 48 h acquisition access period. Then, the viruliferous insects are collected and used to inoculate 20 plants of each genotype. The plants at their 4 leaf-stage are inoculated by caging ten viruliferous whiteflies with the youngest true leaf (more than 1 cm) of each plant, using leaf cages. Mock inoculations are conducted using the same number of non-viruliferous insects. After a 12 h inoculation access period (which ensures 100% infection), the insect cages are removed and the plants are treated with insecticide (imidacloprid). Project description:The Asian citrus psyllid (Diaphorina citri) is a pest of citrus and the primary insect vector of the bacterial pathogen, ‘Candidatus Liberibacter asiaticus’ (CLas), which is associated with citrus greening disease. Variability in CLas titer in insects collected from infected plants has been attributed in part to the host plant from which the insects were collected. CLas accumulates to high titers in infected Citrus macrophylla, and in D. citri feeding on the infected plants of this species. In contrast, in the citrus relative Murraya paniculata, CLas titers remain low in infected plants and in D. citri exposed to infected plants. In this study, top-down and bottom-up proteomics methods were used to investigate the impact of these different host plants on D. citri protein expression. Difference in gel electrophoresis (DIGE) was used to identify protein spots on two-dimensional gels that were larger in one of three insect sample classes compared to the other two: D. citri continuously reared on C. macrophylla, D. citri reared continuously on M. paniculata, and D. citri transferred to M. paniculata for five days feeding after continuous rearing on C. macrophylla. Peptide mass spectrometry was used to identify and quantify proteins in target spots upregulated in each sample class. Shotgun proteomics was used to identify and quantify proteins from analysis of tryptic peptide samples prepared from whole insects from four sample classes: the reciprocal host switch condition (D. citri transferred to C. macrophylla for five days feeding after continuous rearing on M. paniculata) in addition to the three sample classes used in DIGE analysis. Integration of the results of both analyses reveals proteins identified by separate experimental workflows to be upregulated in insects adapted to each host plant, and in insects adapting to a novel host plant. A peptidoglycan-degrading protein involved in the immune response to bacterial pathogens was found to be upregulated in M. paniculata-reared D. citri. In the absence of CLas infection, host plant factors specific to M. paniculata may prime the antibacterial immune response in D. citri. Understanding the insect proteins involved in the adaptation of D. citri to host plants with variation in their susceptibility to CLas will inform the development of control strategies aimed at stopping the spread of citrus greening disease.
