The response of the melanized yeast Exophiala dermatitidis to gamma radiation exposure

Project description:The melanized yeast Exophiala dermatitidis is resistant to many environmental stresses, and is used as a model for understanding the diverse roles of melanin in fungi. To further our understanding resistance mechanism of E. dermatitidis to acute γ-radiation exposure and whether melanin plays a role in this process, we performed RNA-seq on WT and the melanin deficient mutant Δpks1 cultures exposed to γ-radiation at the dose of 1000 Gy, which resulted in ~50% cell deaths in both strains. We observed a dramatic transcriptomic response to γ-radiation that mobilizes pathways involved in morphological development, protein degradation, and DNA repair, and is unaffected by the presence of melanin.

DIA-based quantitative proteomics of Staphylococcus aureus exposed to gamma radiation

Project description:S. aureus was exposed to gamma radiation at an irradiation dose of 100 Gy, and its descendant were cultured under normal conditions. Protein identification and quantification of unirradiated, irradiated, and descendant of irradiated S. aureus were performed using DIA proteomic technology to obtain protein abundance profiles for each group. Combined with biological experiments, further functional enrichment analysis and protein-protein interaction network analysis were performed to explore the effect of gamma radiation on the pathogenicity of S. aureus.

Tardigrades dramatically upregulate DNA repair pathway genes in response to ionizing radiation

Project description:Tardigrades can survive remarkable doses of ionizing radiation, up to about 1000 times the lethal dose for humans. How they do so is incompletely understood. We found that the tardigrade Hypsibius exemplaris suffers DNA damage upon gamma irradiation, but damage is repaired. We show that tardigrades have a specific and robust response to ionizing radiation: irradiation induces a rapid, dramatic upregulation of many DNA repair genes. By expressing tardigrade genes in bacteria, we validate that increased expression of some repair genes can suffice to increase radiation tolerance. We show that at least one such gene is necessary for tardigrade radiation tolerance. Tardigrades’ ability to sense ionizing radiation and massively upregulate specific DNA repair pathway genes may represent an evolved solution for maintaining DNA integrity.

Gamma irradiation does not induce detectable changes in DNA methylation in exposed human cells

Project description:Ionizing radiation, released during accidents at nuclear power plants or after atomic bomb explosions, is a potentially serious health threat for the exposed human population. This type of high-energy radiation causes DNA damage including single- and double-strand breaks and induces chromosomal rearrangements and mutations, but it is not known if ionizing radiation directly induces changes in the epigenome of irradiated cells. We treated normal human fibroblasts and normal human bronchial epithelial cells with different doses of gamma-radiation emitted from a cesium 137 (137Cs) radiation source. After a recovery period, we analyzed global DNA methylation patterns in the irradiated and control cells using the methylated-CpG island recovery assay (MIRA) method in combination with high-resolution microarrays. Bioinformatics analysis revealed only a small number of potential methylation changes with low fold-difference ratios in the irradiated cells. These minor methylation differences seen on the microarrays could not be verified by COBRA (combined bisulfite restriction analysis) or bisulfite sequencing of selected target loci. Our study shows that acute gamma-radiation treatment of two types of human cells had no appreciable effect on DNA cytosine methylation patterns in exposed cells. Global gene expression analysis in PBMCs of individuals from normal and high level natural radiation areas of Kerala Coast, India

Project description:Gene expression changes in human populations exposed to chronic low level radiation have important implications. There are few areas around the world where human population is exposed to elevated levels of natural background radiation. The high level natural radiation areas (HLNRAs) of Kerala coast in south west India is unique for its wide variation in the background radiation dose (<1.0mGy to 45mGy/year). The areas with a background radiation dose of ≤ 1.5mGy/year are considered as normal level natural radiation areas (NLNRA), whereas areas with > 1.5mGy/year are considered as HLNRA. We used microarray analysis to find out global changes in gene expression in peripheral blood mononuclear cells (PBMCs) of individuals belonging to different HLNRA groups as compared to individuals from normal level natural radiation areas (NLNRA).
