Project description:Rationale Microplastics are a pressing global concern and inhalation of microplastic fibers has been associated with interstitial and bronchial inflammation in flock workers. However, how microplastic fibers affect the lungs is unknown. Objectives Our aim was to assess the effects of 12x31 µm nylon 6,6 (nylon) and 15x52 µm polyethylene terephthalate (polyester) textile microplastic fibers on lung epithelial growth and differentiation. Methods We used human and murine alveolar and airway-type organoids as well as air-liquid interface cultures derived from primary lung epithelial progenitor cells and incubated these with either nylon or polyester fibers or nylon leachate. In addition, mice received one dose of nylon fibers or nylon leachate and 7 days later organoid-forming capacity of isolated epithelial cells was investigated. Results We observed that nylon microfibers, more than polyester, inhibited developing airway organoids and not established ones. This effect was mediated by components leaching from nylon. Epithelial cells isolated from mice exposed to nylon fibers or leachate, also formed fewer airway organoids, suggesting long-lasting effects of nylon components on epithelial cells. Part of these effects were recapitulated in human air-liquid interface cultures. Transcriptome analysis revealed upregulation of Hoxa5 post-exposure to nylon fibers. Inhibiting Hoxa5 protein during nylon exposure restored airway organoid formation, confirming Hoxa5's pivotal role in the effects of nylon. Conclusions These results suggest that components leaching from nylon 6,6 may especially harm developing airways and/or airways undergoing repair and we strongly encourage to characterize both hazard of and exposure to microplastic fibers in more detail.

Project description:Purpose: To analyze the toxicity effect of sheepshead minnow larvae exposed to PET microfibers through RNA seq. The expression of specific and sensitive genes on PET microfibers was confirmed and verified through qRT-PCR. Method: RNA was isolated from a sample exposed to PET microfibers (concentration of 10, 100 mg/L) for 10 days using a 72 hpf sheepshead minnow larvae. mRNA was produced with a TruSeq Standed mRNA library kit, and sequencing using a NovaSeq6000 equipment. qRT–PCR validation was performed using SYBR Green assays Results and Conclusions: Our study represents a detailed analysis of the transcriptome of sheepshead minnow larvae exposed to PET microfibers by RNA-seq technology. Through the 10 and 100 mg/L PET exposure, 670 differentially expressed genes confirmed. These genes that are specific and sensitive to PET microfibers are concentrated in Dendrite morphogenesis, heart development, intracellular protein kinase cascade, cell migration, and cardiac muscle cell development through GO analysis. In addition, the changes in cancer genesis, cell reaction, energy, and neuronal related genes were confirmed.

Project description:Mytilus coruscus were injected with microbes into the adductor muscle. The microbes are Staphylococcus aureus, Vibrio alginolyticus, and Candida albicans. After microbial challenge, hemolymph was collected from the adductor muscle of M. coruscus at 2h post bacterial challenge. The hemolymph was isolated by 12% SDS-PAGE and the differential bands were collected for LC-MS/MS proteomic analysis.

Project description:Opioids such as morphine have many beneficial properties as analgesics, however, opioids may induce multiple adverse gastrointestinal symptoms. We have recently demonstrated that morphine treatment results in significant disruption in gut barrier function leading to increased translocation of gut commensal bacteria. However, it is unclear how opioids modulate the gut homeostasis. By using a mouse model of morphine treatment, we studied effects of morphine treatment on gut microbiome. We characterized phylogenetic profiles of gut microbes, and found a significant shift in the gut microbiome and increase of pathogenic bacteria following morphine treatment when compared to placebo. In the present study, wild type mice (C57BL/6J) were implanted with placebo, morphine pellets subcutaneously. Fecal matter were taken for bacterial 16s rDNA sequencing analysis at day 3 post treatment. A scatter plot based on an unweighted UniFrac distance matrics obtained from the sequences at OTU level with 97% similarity showed a distinct clustering of the community composition between the morphine and placebo treated groups. By using the chao1 index to evaluate alpha diversity (that is diversity within a group) and using unweighted UniFrac distance to evaluate beta diversity (that is diversity between groups, comparing microbial community based on compositional structures), we found that morphine treatment results in a significant decrease in alpha diversity and shift in fecal microbiome at day 3 post treatment compared to placebo treatment. Taxonomical analysis showed that morphine treatment results in a significant increase of potential pathogenic bacteria. Our study shed light on effects of morphine on the gut microbiome, and its role in the gut homeostasis.

Project description:Mussels are a group of sessile and filter-feeding invertebrates, and are key structural elements of many marine and freshwater ecosystems. The mussel haemolymph is the first defense line to pathogen infection and inflammation with an innate immune response. Despite the importance of this body fluid in determining the physiological condition of the animal, little is known about the molecular mechanisms underlying the cellular and humoral responses. Proteomic characterization of this body fluid has been hampered by the scarce Mytilus sp genomic data available. To systematic characterize the marine mussel Mytilus edulis hemolymph proteome, we have applied a mass spectrometry (nanoLC-MS/MS) strategy integrating genomics and transcriptomics data for proteins identification. After sample analysis and first identification based on MS/MS data comparison, proteins with unknown functions were annotated with blast using public database (nrNCBI) information. Overall 654 haemolymph proteins were identified with high confidence. The gene ontology analysis revealed that the majority of haemolymph proteins participate in primary cellular metabolic processes: energy production and metabolism of biomolecules. Nevertheless it also revealed a protein complement whose functions could be related to oxidative stress defense, xenobiotic detoxification, drug metabolism and immune response.
