Project description:72 plants have been grown in 2 phytochambers for 30 days under control temperature (21Â°C/19Â°C) and short day conditions (8 hours light/16 hours darkness). After 30 days the plants were switched to long day conditions (16 hours light/8 hours darkness) and the temperature of one phytochamber was increased to 29Â°C/27Â° (heat chamber). Moreover,a heatplate was located in the control chamber and a coldplate was located in the heat chamber. 18 plants have been grown on the heatplate with a constant temperature of 29Â°C. 18 plants have been grown on the coldplate with a constant temperature of 22Â°C. After 9 days of the heat period leaf samples (end of night) were taken for micorarray analysis. After 10 days of heat, tuber samples were taken for microarray analysis.

Transcription profiling of rice seedlings after heat shock

Project description:In this study, we have analyzed the expression profiles of rice genes under control and heat shock conditions using microarray technology to identify the genes differentially expressed. Experiment Overall Design: 14-day-old light-grown rice seedlings grown under controlled conditions and those subjected to heat shock were used for RNA extraction and hybridization on Affymetrix microarrays. Three technical replicates of each sample were used for microarray analysis. For heat shock treatment, the seedlings were kept in a chamber at 42 degree C. The seedlings kept at 28-30 degree C, served as control (Seedling).

Project description:Wild type Golden Promise barley and the frost tolerant TaCBF14 and TaCBF15 transgenic barley lines were grown in plant growth chamber in the Phytotron of ELKH ATK Agricultural Institute, Martonvasar, Hungary. After three weeks in control conditions, plants were treated by short-term cold-treatment. Samples were collected under control condition and after the short-term cold-treatment too.

Transcriptional profiling of S. tuberosum cv. Agria leaf and tuber samples after mild heat stress

Project description:The consequences on tuber transcriptome of a short heat period during tuber development was investigated in this study with special regard to the development of secondary tuber growth. Plants were grown for 47 days in the greenhouse under ambient conditions (21Â°C/ 19Â°C, 16h light, 8h dark) before application of mild heat stress temperatures (29Â°C/27Â°C) to one group of plants for 7 days and a stress release period on control temperature for 2 more weeks until harvest. Leaves were sampled before the heat period, at the end of the heat period and at harvest, two weeks after stress release. Tuber samples were taken at harvest. Tubers grown at normal temperatures and exhibiting a normal growth phenotype were used as control. Tubers subjected to the heat treatment and exhibiting a second-growth phenotype (chain tubers) were grouped into primary (attached to stolon from plant) and secondary tubers (attached to stolon from primary tuber).

Cryptochrome 1 attenuates plant heat stress response by light-heat stress response signal pathways crosstalk in Arabidopsis

Project description:Plants regulate growth and development in different ambient environments through light signaling pathways and heat stress signaling pathways, both of which are very important for plant adaptation to the environment, but whether there is an interaction between the two is still unclear. In this article, we reported that the blue light receptor Cryptochrome CRY1 phosphorylation will be inhibited under high temperature stress, and CRY1 activity will decrease; at this time, CRY1 could not interact with E3 ubiquitin ligase COP1(CONSTITUTIVELY PHOTOMORPHOGENIC 1) and COP1 activity increased. Its substrate HY5(ELONGATED HYPOCOTYL 5）is continuously degraded under heat stress. Through further transcriptome analysis, we found that the substrates of HY5 have a class of HSFAs（HEAT SHOCK FACTOR）HSFA2/HSFA7As/HSFA7Bs transcription factors, which are rapidly induced under heat stress conditions and activate downstream HSPs (HEAT SHOCK PROTEIN)expression.Under normal conditions, HY5 protein inhibits HSFA2/HSFA7As/HSFA7Bs. Under heat stress, HY5 protein is degraded, which releases the expression of HSFAs, thereby enhancing plant heat tolerance. In this paper, we put forward the hypothesis that the phosphorylation of CRY1 seems to be a switch in the ambient environment , and through the regulation of phosphorylation level, the light and heat stress signal are transmitted downward, forming an light-heat signal interactive regulation pathway to regulate the growth and development of plants.
