The effect of insecticide synergist treatment on genome-wide gene expression in a polyphagous pest

Project description:Synergists can counteract metabolic insecticide resistance by inhibiting detoxification enzymes or transporters. In this study we used Illumina RNA-sequencing to investigate genome-wide transcriptional responses in an acaricide resistant strain (JP-R) of the spider mite Tetranychus urticae upon exposure to synergists such as S,S,S-tributyl phosphorotrithioate (DEF), diethyl maleate (DEM), piperonyl butoxide (PBO) and cyclosporin A (CsA).

Cyenopyrafen resistance in the two-spotted spider mite Tetranychus urticae

Project description:Cyenopyrafen is a novel acaricide with a currently unknown mode of action. We selected for high levels of cyenopyrafen resistance in a susceptible spider mite strain and previously published patterns of cross-resistance to other well-described acaricides (Khalighi et al., 2014). In addition, we also included an independent cyenopyrafen-susceptible mite strain (referred to as Akita) in this study. We performed transcriptomic analysis to identify on the molecular level how T. urticae develops resistance to this novel acaricide. Using this dataset, we show that the selection for cyenopyrafen resistance resulted in massive transcriptomic responses in T. urticae. The multi-gene family of cytochrome P450 monooxygenases (CYPs) exhibited a drastic differential expression with a significant transcriptional induction in the acaricide resistant strain. This transcriptomic study highlights the potential importance of CYPs in mite resistance to the cyenopyrafen acaricide.

Next Generation Sequencing Facilitates Quantitative Analysis of Tetranychus urticae and Its Sibling Species Tetranychus cinnabarinus Transcriptomes

Project description:Purpose: Next-generation sequencing (NGS) has revolutionized systems-based analysis of differentially expressed genes (DEG) between the treated samples of mites.The goal of the study is to elucidate the invasion mechanism that results in the continuous expansion of Tetranychus urticae that has occurred in China. Methods:Transcriptome profiling by RNA sequencing is becoming an attractive method as it facilitates rapid generation, identification and quantification of large number of transcripts, even in the species where no prior genome sequence information is available. Results:the expression of 10 P450(cytochrome P450 monooxygenases), 6 GST(glutathione S-transferases), 13 CCE(carboxy/cholinesterases) and other novel candidate genes was higher (>2-fold) in T. urticae than in T. cinnabarinus as detected by using the digital gene expression (DGE) method.

Hopeful monsters: Unintended sequencing of famously malformed mite mitochondrial tRNAs reveals widespread expression and processing of sense-antisense pairs

Project description:While performing mitochondrial isolations and recently developed tRNA-seq methods (AlkB treatment and YAMAT-Seq) in plant tissue, we inadvertently sequenced the mitochondrial tRNAs from a common plant pest, the acariform mite Tetranychus urticae, to a high enough coverage to detect all previously annotated T. urticae tRNA regions. The results not only confirm expression, CCA-tailing and post-transcriptional base modification of these highly divergent tRNAs, but also revealed paired sense and antisense expression of multiple T. urticae mitochondrial tRNAs.

Cyflumetofen resistance in the two-spotted spider mite Tetranychus urticae

Project description:Cyflumetofen is a novel acaricide on the international market with an unknown mode of action. Under laboratory conditions, we selected for high levels of cyflumetofen resistance in a susceptible mite strain and performed genome-wide gene-expression analysis. Differential expression between the resistant and susceptible strain was identified to study on the molecular level how mites develop resistance to this novel acaricide. Our results show that in T. urticae, the selection for cyflumetofen resistance resulted in a differential expression in only a limited number of genes. This transcriptomic study provides an unbiased look how cyflumetofen resistance triggers selection on the transcriptional level in T. urticae.

Whole genome tomato transcriptional response to Tetranychus urticae herbivory [Heinz1706]

Project description:The two-spotted spider mite, Tetranychus urticae, is one of the most significant mite pests in agriculture that can feed on more than 1,100 plant hosts, including model plants Arabidopsis thaliana and tomato, Solanum lycopersicum. Here, we described tomato transcriptional responses to spider mite feeding and compared them to Arabidopsis in order to determine conserved and divergent responses to this pest. 2,133 differentially expressed genes (DEGs) were detected at 1, 3, 6, 12 or 24 hours post spider mite infestation (hpi) relative to non-infested control plants. Based on Biological Process Gene Ontology annotations, improved in the course of our analysis, DEGs were grouped in 60 significantly enriched gene sets that highlighted perception of the spider mite attack (1 hpi), metabolic reprogramming (3-6 hpi), and establishment and maintenance of the defense responses (6-24 hpi). We used microarray to assess global gene expression in Solanum lycopersicum cv. Heinz 1706 upon Tetranychus urticae attack. 1 month old tomato plants were subjected to Tetranychus urticae attack through application of 100 adult mites on a terminal leaflet of leaf 3 for various periods of time (timecourse scenario) or hundreds of mites for 1 hour (feeding site scenario).
