Project description:Female adult spongy moths vary in their flight capacity depending upon their origin and subspecies. Asian spongy moth (Lymantria dispar asiatica) females (such as the RM strain here) can fly whereas European spongy moth (Lymantria dispar dispar) females cannot (such as the CT strain here). We hypothesized that the genetic origins of these differences in flight capacity would be reflected in differences in gene expression during metamorphosis (the pupal stage). To test this, we compared the gene expression at days 1, 3, 5, 8, and 11 of pupal development of female pupae from CT and RM strains.

Project description:Transcriptome analysis of the spongy tissue disorder in Mangifera indica Cv. Alphonso was carried out individually at two stages of ripening (Table green and mid ripe) in the healthy and the spongy mesocarp. Sequencing was carried out using Illumina technique. Total number of raw reads generated from each stage of tissue ranged from 5-7 Gb. Importantly, 30,582 transcripts were annotated and 1,218 enzyme commission number were assigned from oxidoreductase, transferase, hydrolase, ligase, lyase and isomerase classes. Various differential transcripts (p < 0.05) were highlighted within the healthy and the spongy tissue during ripening. These were involved in biological pathways related to metabolism of fatty acids, sugars, amino acids etc. The altered expression pattern of these transcripts indicated the change in primary as well as secondary metabolism of fruit under the spongy tissue condition. Interestingly, several stress related transcripts were differentially expressed in both the stages of the spongy mesocarp which has advocated the role of abiotic stress in the spongy tissue disorder.

Project description:miRNAs play important roles in various biological processes through post-transcriptional regulation of gene expression. We previously identified 203 mature miRNAs in Diamondback moth, Plutella xylostella. This species has developed extremely high levels of resistance to chlorantraniliprole and other class of insecticides in the field. In this study, we examined the miRNA profile of P. xylostella in response to chlorantraniliprole exposure. The smRNA-seq data analyses showed that insecticide treatment caused significant changes in the abundance of some miRNAs. Increasing exposure time (6h to 24h) and insecticide concentration (0.01 to 0.1 ppm) induced more dysregulated miRNAs in DBM larvae.

Project description:Arabidopsis thaliana plants (ecotype Landsberg erecta) were grown at 20˚C constant temperature, 8 hr/16 hr Light/Dark photoperiod at 50-60% ambient humidity, for 8 to 9 weeks. Short day conditions prevented the onset of flowering and the plants were thus maintained in growth stage 1 (leaf production) with 13 to 15 rosette leaves larger than 1mm (stage 1.13 to 1.14). Diamondback moth (DBM, Plutella xylostella) larvae were maintained on cabbage (Brassica oleracea) plants in a climate-controlled room at 25ºC, 12 hr photoperiod with 50%-60% relative humidity. Two days before exposing A. thaliana plants to herbivore treatment, plants were transferred to a climate-controlled room (22ºC, 50-60% humidity, 12 hr photoperiod). For insect treatment, seven Diamondback moth (DBM, Plutella xylostella) larvae (third to fifth instars) were placed on a group of four or five plants until time of harvest, for each time point separately. All rosette leaves were harvested at 1h, 4h, 12h, and 24h after onset of continuous herbivory and flash frozen in liquid nitrogen. Project description:Polyethylene pollutions are considered inert in nature and adversely affect the entire ecosystem. Larvae of greater wax moths (Galleria mellonella) have the ability to masticate and potentially biodegrade polyethylene films at elevated rates. The wax moth has been thought to metabolize PE independently of gut flora, however, the role of the microbiome is poorly understood and degradation by the wax moth might be involved. To determine whether the salivary glands of the wax moth were potentially involved in the PE degradation, it was investigated how surface changes of polyethylene were affected by mastication and consumption. Formation of pitting and degradation intermediates, including carbonyl groups, indicated that salivary glands could assist in polyethylene metabolism. We investigated the biochemical effect of exposure to PE on the composition of the salivary gland proteome. The expression of salivary proteins was found to be affected by PE exposure. The proteins that were significantly affected by the exposure to PE revealed that the wax moth is undergoing general changes in energy levels, and that enzymatic pathways associated with fatty acid beta oxidation were induced during PE consumption.

Project description:Laser Capture Microdissection (LCM) was used for expression analysis of three contrasted fungal tissues of uredinia corresponding respectively to spores and sporogenous hyphae, fungal structures in the spongy mesophyll and fungal infection structures in the palisade mesophyll. The array probes were designed from gene models taken from the Joint Genome Institute (JGI, Department of Energy) Melampsora larici-populina genome sequence version 1. The aim of this study was to identify tissue-specific gene expression to gain insights into the genes specifically associated with the biotrophic phase and the sporulation phase of the rust fungus. We performed 9 hybridizations (NimbleGen) with samples derived from Laser Capture Microdissection (LCM) of three contrasted fungal tissues of uredinia, corresponding respectively to spores and sporogenous hyphae, fungal structures in the spongy mesophyll and fungal infection structures in the palisade mesophyll. Three replicates per tissue. All samples were labeled with Cy3.
