Project description:Temperature has fundamental influences on fitness and distribution of insects. There have been very few studies on effects of mild cold conditions on insects. We found Plutella xylostella had reduced fitness and females produced few viable eggs when reared at 10°C, a temperature at which they can complete development. Male moths reared at 10°C were not able to fertilize eggs, but the eggs produced by female moths reared at this temperature were viable and could be fertilisied by males moths reared at a warmer temperature (25°C). Subsequently, we examined the transcriptomic changes in mid-fourth instar female and male larvae reared at 10°C and 25°C to investigate sex-dependent developmental and physiological responses of P. xylostella to the mild cold stress. We found 624 differentially expressed genes (DEGs) in females, the majority of which were down-regulated. In males 3239 genes were dysregulated and the majority were up-regulated. Only 280 DEGs were common to both sexes. In females, no DEGs encoded heat shock or cold shock proteins, but some of the DEGs in males did encode these proteins. These differences might suggest that female and male adopt some different strategies in coping with cold stress and/or they were passively affected by coldness to different degrees and in different ways. In addition, DEGs encoding antimicrobial peptides, cytochrome P450 monooxygenases, fatty acid-related enzymes, cuticle proteins, myofilament, and hormone-related proteins were found in both sexes under cold stress. The transcriptome study reveals unexpected sex-dependent thermal responses and provides much new information of how insects that do not diapause cope with a decreased temperature

Project description:Arabidopsis thaliana plants (ecotype Landsberg erecta) were grown at 20˚C constant temperature, 8 hr/16 hr Light/Dark photoperiod at 50-60% ambient humidity, for 8 to 9 weeks. Short day conditions prevented the onset of flowering and the plants were thus maintained in growth stage 1 (leaf production) with 13 to 15 rosette leaves larger than 1mm (stage 1.13 to 1.14). Diamondback moth (DBM, Plutella xylostella) larvae were maintained on cabbage (Brassica oleracea) plants in a climate-controlled room at 25ºC, 12 hr photoperiod with 50%-60% relative humidity. Two days before exposing A. thaliana plants to herbivore treatment, plants were transferred to a climate-controlled room (22ºC, 50-60% humidity, 12 hr photoperiod). For insect treatment, seven Diamondback moth (DBM, Plutella xylostella) larvae (third to fifth instars) were placed on a group of four or five plants until time of harvest, for each time point separately. All rosette leaves were harvested at 1h, 4h, 12h, and 24h after onset of continuous herbivory and flash frozen in liquid nitrogen. Project description:RNA-seq to analyse differential expression between moths (Helicoverpa armigera) flown on tethered flight mills. Insects were split into two phenotypes based on the distance flown during the course of a single night. Two separate comparisons were performed. The first compared long-distance fliers from Dafeng (DF) with short-distance fliers from Anyang (AY). These are two separate populations approximately 650km apart in China. The second comparison looked at two flight phenotypes from the same population originating from Northern Greece (GR).

Control plants were harvested in parallel. Total RNA from each sample was used for two labelling reactions using dye-flips (Cy5 or Cy3) and labelled cDNA from treatment and control plants were co-hybridized. Thus, for each time-point four replicates were analyzed each comparing treatment with the corresponding control (rep1 and rep2 are dye-flips of biological replicate 1, and rep3 and rep4 are dye-flips of biological replicate 2). For background correction, we defined the mean of the lowest 10% of spot intensities from a particular subgrid as the background for that subgrid. This mean was subtracted from each spot in the subgrid. Signal intensities that did not exceed the background plus 3 standard deviations thereof were defined as not detectable and were excluded from further analyses. We normalized using loess curves thus generating log2 transformed expression ratios comparing DBM treatment with control. These ratios are given in the VALUE columns of each array.

Project description:Transcriptional regulation by Store-operated Calcium Entry (SOCE) is well studied in non-excitable cells. However, the role of SOCE has been poorly documented in neuronal cells with more complicated calcium dynamics. Previous reports demonstrated a requirement of neuronal SOCE for Drosophila flight. We identified the early pupal stage to be critical and used RNA-sequencing to identify SOCE mediated gene expression changes in the developing Drosophila pupal nervous system. We down-regulated dStim, the endoplasmic reticular calcium sensor and a principal component of SOCE in the nervous system for a 24h period during pupal development, and compared wild type and knockdown transcriptional profiles, immediately after knockdown as well as after a 36h recovery period. We found that dStim knockdown altered the expression of a number of genes. We also characterized one of the down-regulated genes, Ral for its role in flight. Thus, we identify neuronal SOCE as a mechanism that regulates expression of a number of genes during the development of the pupal nervous system. These genes can be further studied in the context of pupal nervous system development.

Project description:Polyethylene pollutions are considered inert in nature and adversely affect the entire ecosystem. Larvae of greater wax moths (Galleria mellonella) have the ability to masticate and potentially biodegrade polyethylene films at elevated rates. The wax moth has been thought to metabolize PE independently of gut flora, however, the role of the microbiome is poorly understood and degradation by the wax moth might be involved. To determine whether the salivary glands of the wax moth were potentially involved in the PE degradation, it was investigated how surface changes of polyethylene were affected by mastication and consumption. Formation of pitting and degradation intermediates, including carbonyl groups, indicated that salivary glands could assist in polyethylene metabolism. We investigated the biochemical effect of exposure to PE on the composition of the salivary gland proteome. The expression of salivary proteins was found to be affected by PE exposure. The proteins that were significantly affected by the exposure to PE revealed that the wax moth is undergoing general changes in energy levels, and that enzymatic pathways associated with fatty acid beta oxidation were induced during PE consumption.

Project description:In insects, male accessory gland proteins (ACPs) are important reproductive proteins secreted by male accessory glands (MAGs) of the internal male reproductive system. During mating, ACPs were transferred along with sperms inside female bodies and have a significant impact on the physiology of female reproduction. Under sexual selection pressures, the ACPs exhibit remarkably rapid and divergent evolution and varies from species to species. The diamondback moth, Plutella xylostella (L.) (Lepidoptera: Plutellidae), is a major insect pest of cruciferous vegetables worldwide. The reproductive physiology on post-mating state of this species is still largely unknown, which is important for management of this pest. In this study, the ACPs transferred into females during mating were identified by using a tandem mass tags quantitative proteomic analysis. The MAGs were compared before and after mating immediately. The proteomes of copulatory bursas (CB) in mated females shortly after mating were also analyzed by shotgun LC-MS/MS technique. In total, we identified 123 putative secreted ACPs, including most important physiological: regulators of proteolysis, transporters and protein export machinery, signal transduction and immunity. Comparing P. xylostella with other four insect ACPs, trypsins were the only ACPs detected in all insect species. We also identified some new insect ACPs, including protein with chitin binding Peritrophin-A domain, PMP-22/EMP/MP20/Claudin tight junction domain-containing protein, netrin-1, type II inositol 1,4,5-trisphosphate 5-phosphatase, two spaetzles, allatostatin-CC and cuticular protein. This was the first time to identify and analyze ACPs in P. xylostella. Our results have provided an important list of putative secreted ACPs, and have set the stage for further explore functions of these putative proteins in P. xylostella reproduction.
