Project description:The aim of this study was to identify differentially expressed genes among male, female and hermaphrodite flowers in papaya during early (pre-meiosis) and later (post-meiosis) stages of flower development. We identified a Male Sterility 1 gene (CpMS1) highly up-regulated in male and hermaphrodite flower buds compared to female flower buds. Besides important transcription factors related to flower organ development and flowering time regulation, we identified differential expression of genes that are known to participate in ABA, ROS and auxin signaling pathways.

Project description:Heat stress is a major challenge for crop production. During the reproduction stage, pollen development is the most sensitive process to abnormal environmental conditions. In this project, we have developed a heat treatment system for tomato where plants produce a significantly lower number of pollens and the germination rate of those pollens was also very low. The size of the flower buds and pollen developmental stages were examined under confocal microscope. Pollen cells at different developmental stages were collected from flower buds at corresponding sizes using LCM. The single cell population proteomics analysis was used to identify proteome changes in a distinct group of cells. This information was used to determine the function of proteins that are associated with heat stress on cellular processes/functions during pollen development.

Project description:Phalaenopsis amabilis, one of the most important flowers in the current international flower market, is a plant that undergoes vernalization and requires low temperature treatment for flowering. There have been few reports on the proteomic analysis of the development of flower buds. In this study, by using isobaric tags for relative and absolute quantification (iTRAQ), 4096 differentially expressed proteins were identified in P. amabilis under low temperature treatment, of which 42 were associated with early floral induction, and 18 were verified by mass spectrometry multi-reaction monitoring (MRM). Among the proteins associated with the vernalization pathway, PEQU_11434 (glycine-rich RNA-binding protein GRP1A-like) and PEQU_11045 (UDP-N-acetylglucosamine diphosphorylase) were upregulated compared to their expression in control flower buds. It was therefore inferred that O-GlcNAc glycosylation was involved in the posttranscriptional modification of VRN1 (API homolog) and that the GRP2 protein (glycine-rich RNA-binding protein) was glycosylated to relieve binding to the VRN1 mRNA precursor to promote the expression of VRN1, which initiates floral development. Furthermore, phytochromes A (PEQU_13449, PEQU_35378), B (PEQU_09249) and C (PEQU_41401) were downregulated under low temperature treatment compared to their expression in control flower buds, suggesting that they could repress the expression of the VRN2 gene and thus release its repression of VRN3 to enable the high-level expression of FTPEQU_19304 (FT, VRN3 homolog), which promotes VRN1 expression and then stimulates flowering.

ABAP1 is a negative regulator of the cell cycle that binds to transcription factors and represses their target gene expression, including pre-replication complex genes. Project description:Watermelon (Citrullus lanatus (Thunb.) Matsum. & Nakai) is an important cucurbit crop grown worldwide. Watermelon fruit quality, fertility, and the seed setting rate are closely related to male flower development. In this study, the different developmental stages of flower buds of the watermelon variety Xinteda Zhengkang 9 were distinguished by cytological observation, and transcriptome sequencing analysis was performed subsequently. Acetocarmine staining of anthers was performed and the longitudinal and transverse diameters of the unopened male flower buds were measured. Cytological observations of anthers at different developmental stages showed that the anther grew from the tetrad to the mature stage, and the longitudinal and transverse diameters of the flower buds increased. The length of the male flower buds also changed significantly during development. Transcriptome sequencing analysis at four periods, the tetrad (A group), mononuclear (B group), dikaryophase (C group), and mature stages (D group). Total 16288 differentially expressed genes (DEGs) were detected in the four stages, with the prolongation of developmental stages the number of DEGs increased gradually in the comparison groups, such as there was 2014, 3259, 4628, 1490, 3495 and 1132 DEGs were revealed in six comparison groups (A-VS-B, A-VS-C, A-VS-D, B-VS-C, B-VS-D, and C-VS-D), respectively. Gene ontology (GO) and KEGG enrichment analysis showed that the DEGs were mainly enriched in cellular component and starch and sucrose metabolism, phenylpropanoid biosynthesis and pentose sugar, etc. Finally, we totally screened 59 DEGs were in the six comparison groups, and most of them were up-regulated, interestingly, we find one pollen-specific protein (Cla001608) was significantly up-regulated (the value of log2Fold Change up to 17.32), which indicating that it may play important role in the development of male flowers. This work provides insight into the molecular basis of the developmental stages of male flowers in watermelon and may aid in dominant cross breeding.
