Project description:The thermophilic Aquificales inhabit and play important biogeochemical roles in the geothermal environments globally. Although intensive studies on physiology, microbial ecology, biochemistry, metagenomics and metatranscriptomics of the Aquificales¬ species and Aquificales-containing environmental samples have been conducted, comprehensive understandings about their ecophysiology, especially in the natural niches have been limited. In the present study, an integrated suite of metagenomic, metatranscriptomic and metaproteomic analyses, for the first time, were conducted on a filamentous microbial community from the Apron and Channel Facies (ACF) of CaCO3 (travertine) deposition at Narrow Gauge, Mammoth Hot Springs, Yellowstone National Park.

Project description:Protein stable isotope fingerprinting (P-SIF) is a method to measure the carbon isotope ratios of whole proteins separated from complex mixtures, including cultures and environmental samples. The goal of P-SIF is to expose the links between identity and function in microbial ecosystems by (i) determining the values of δ13C for different taxonomic divisions, and (ii) using those values as clues to the metabolic pathways employed by the respective organisms. This project measures >200 protein fractions and δ13C values for a sample of Cyanobacteria + Chloroflexi dominated microbial mat from Yellowstone National Park, USA.

Community-wide interactions sustain life in geothermal spring habitats

Project description:We investigated an alga-dominated geothermal spring community in Yellowstone National Park (YNP), USA to determine how the biota cope with abiotic stressors. Microbes showed a community level response to toxic metal resistance and energy cycling that spans the three domains of life. Arsenic detoxification is accomplished via complementary expression of genes by different lineages. Photosynthetic primary production is dominated by the obligate photoautotrophic alga Cyanidioschyzon, with the mixotroph, Galdieria, largely relegated to nighttime heterotrophy. Many key functions, including the cell cycle, are strongly regulated by diurnal fluctuations in light and nutrients. These results demonstrate that biotic interactions are highly structured and constrained in extreme habitats. We suggest this was also the case on the early Earth when geothermal springs were cradles of microbial life. Temperature and livestock grazing trigger transcriptome responses in bumblebees along an elevational gradient

Project description:It is assumed that climate and land-use changes cause increasing stress to pollinators, which play pivotal roles in almost all terrestrial ecosystems, with consequences on population growth, diversity and ecosystem functions. While these responses to global change drivers are well located, the molecular pathways triggering the response are poorly understood. We analysed the transcriptomic response of Bombus lucorum workers in their systematic responses to temperature and livestock grazing, sampled along an elevational gradient from 650 – 1930 m.a.s.l., and from differently managed grassland sites (livestock grazing vs. unmanaged) in and around the National Park Berchtesgaden (German Alps).
