Lipid droplet-loaded macrophages as a targetable pro-tumorigenic immune cell entity in human glioblastoma

Project description:Glioblastoma (GBM) presents a formidable clinical challenge due to its complex microenvironment. Here, we introduce tumor-associated foam cells (TAFs), a previously unidentified immune cell entity of lipid droplet (LD)-loaded macrophages, in GBM. Through extensive analyses of patient tumors, together with in vitro and in vivo investigations, we reveal that TAFs exhibit distinct pro-tumorigenic characteristics related to hypoxia, mesenchymal transition, angiogenesis, and impaired phagocytosis. Moreover, TAF presence correlates with worse patient outcome. Our mechanistic investigations demonstrate that TAF formation is facilitated by lipid cargo transfer from extracellular vesicles released by GBM cells. Importantly, we demonstrate that targeting key enzymes involved in LD formation, such as DGAT1 or ACSL, effectively disrupts TAF functionality. Project description:To investigate global changes in the structure of the bone marrow niche with age, we performed droplet-based scRNAseq (10X Genomics) of unfractionated endosteal and central marrow stromal cells (Ter119-/CD45-). 8,735 cells passed QC, and hematopoietic clusters of cells were excluded based on marker gene expression, leaving 2359 cells distributed across 9 distinct clusters for final presentation.

Transcriptomic changes induced by Curcumin loaded solid nanoparticles and radiation in breast cells

Project description:In breast cancer (BC) care, radiotherapy is considered an efficient treatment. However, approximately 90% of BC-related deaths are due to the metastatic tumor progression. Then, it is strongly desirable to improve tumors radiosensitivity using molecules with synergistic action. In this study, we developed curcumin-loaded solid nanoparticles (Cur-SLN) in order to increase curcumin bioavailability and evaluated their radiosensitizing ability in comparison to curcumin free (Cur), by using an in vitro approach on BC models. Precisely, here we analyzed the transcriptomic profiles induced by Cur-SLN treatments, in non tumorigenic MCF10A and tumorigenic MCF7 and MDA-MB-231 BC cell lines, highlighting the networks responsible of its radiosensitization ability. Curcumin loaded- SLN can be suggested in future preclinical and clinical studies to test its concomitant use during radiotherapy treatments with the double implications of being a radiosensitizing molecule against cancer cells, with a protective role against IR side effects.
