Project description:β-cell dysfunction in pancreatic islets is the leading cause of progression to type I diabetes (T1D), but the reason for the loss of β-cell mass in T1D is still not fully understood. Using in vitro long-term cultured (LTC) human islets, we have investigated gene profiling and subtype cells changes in the leftover cells and compared them with residual cells in samples of T1D or T2D. While most of the dysregulated genes were downregulated during LTC, we found there are specific groups of mRNA or miRNA that were upregulated, and they are involved in specific pathways. In general, α-cells and β-cells of LTC-islets have elevated expressions of MAFB and MAFA, respectively. We also found exocrine cells were eliminated faster than endocrine cells, and β-cells were lost at a higher rate than α-cells. Interestingly, a specific group of cells that were classified as α-cells but have immature β-cells-like characteristics, were enriched in LTC, revealed the possibility of transdifferentiating of α-cells to β-cells under in vitro culture. Our results suggested that there are intrinsic cellular and molecular mechanisms in pancreatic cells that are associated with their maturity and correlated with their survival ability under unfavorable living conditions.

Project description:In this study we show, for the first time, that whole islets can be cultured and significantly expanded long-term in vitro on specific basement membrane (BM) laminin LN-521 which is a normal component of islet BMs. Isolated islets adhere and flatten to form 2-3 cell layer entities with ~10 % of all endocrine cell types proliferating after 2-5 weeks in culture. Time-lapse imaging of freshly isolated islets in suspension labeled with hypoxia and DNA degradation indicators reveals that spherical islets on LN-111 rapidly develop hypoxia and central necrosis, in contrast to islets attached to LN-521, which normal b-cell glucose responsiveness. RNA sequencing analyses reveal that freshly isolated islets express LN-521 binding integrins. Compared to other matrices, islets grown on LN-521 develop a unique expression signature, including expression of specific integrins, involved in cell adhesion and matrix production as well as proliferation. The results reveal specific LN-521-mediated effects into islet cells and suggest that use of in vitro expanded islets may significantly enhance the efficacy of islet transplantation treatment of T1D.

Project description:Pancreatic islets are 3-dimensional micro-organs that maintain β-cell functionality via cell-cell and cell-matrix communication. Isolated primary islets are the gold standard for in vitro models. However, native islets present experimental challenges for long-term mechanistic studies owing to their short culture life (approximately 1 week). We developed a novel long-term protocol to study the function of primary islets. The protocol employed reformed islets following dispersion and a fine-tuned culture environment. Reformed islets are highly similar to their primary counterparts across various physiological characteristics. Long-term culture of reformed islets enables high-resolution imaging, repeated functional assessment, and the study of cell-cell communication. Unlike other platforms such as stem cell-derived organoids, reformed islets retain their resident immune populations, making them ideal for studying both resident and infiltrating immune cells and their interactions with hormone-producing islet cells. Qualitative and quantitative analyses revealed that the composition and cytoarchitecture of the reformed islets mimicked those found in primary islets, including the presence of macrophages and CD4+ and CD8+ T cells, which are the key resident immune cell types. Reformed islets secrete insulin and are glucose-responsive, and their β-cells can be stimulated to proliferate using GLP-1 receptor agonism. Furthermore, a comparison of the transcriptomic landscape of isolated human islets and reformed islets generated from the same donor demonstrated a high degree of similarity. Our reformed islets provide an ideal platform to study diabetes pathology. We recapitulated both the T1DM and T2DM disease milieu and validated our model for studying islet immune trafficking and invasion using activated macrophages and T cells. Our data illustrates that reformed islets are an anatomical and functional alternative to native human and mouse islets. Moreover, reformed islets have an advantage over mouse and human β-cell lines, including MIN6 and EndoC-βH1cells, that lack the signalling input of non-β-endocrine cells and immune cell crosstalk. In this study, we showed that reformed islets are a durable paradigm (cell-based model) for islet-based exploration and a means of target discovery/validation for diabetes research.

Project description:Chimeric antigen receptor (CAR) T cells are known to be potent in recognizing and eliminating tumor cells. They induce marked responses in patients with refractory leukemia and lymphoma, but exhaustion is an inevitable obstacle important barrier for to persistant curative effect. CD19-targeted CAR incorporating the 4-1BB costimulation domain showed a lower tendency toward exhaustion during ex vivo expansion, and downstream differentiation was still observed with a prolonged culture time. We explored the transcriptional changes during the culture process using 4-1BB CD19-targeted CAR-T cells . In contrast to T cells (cultured 5 days in vitro) and short-term cultured (5 days) CAR-T cells, the pathway for regulation of the cytosolic calcium ion concentration was significantly upregulated in the long-term cultured (11 days) group.
