Project description:Full understanding of the pathophysiology of COVID-19 is critical for adequate treatment and development of vaccine and therapeutics. Although Golden hamster has been emerged as animal model of COVID-19, it is unknown how SARS-CoV-2 enters and infects targeted epithelial cells at molecular and cellular levels. Here, by applying single cell RNA sequencing in the upper respiratory tract, lung, kidney and intestine of golden hamster, we show that the expression profiles of host factors for SARS-CoV-2 infection in specific cell types are similar to that of human. These data can be applied to a larger investigation (data not provided here) into the expression patterns of host cell entry factors of SARS-CoV-2 in golden hamster organs.

Lipoprotein profiles of human serum and golden hamster plasma

Project description:The lipoproteins of human serum and golden hamster plasma were deeper profiled by label-free proteomics and the shared proteins in lipoprotein particles from humans and golden hamsters were highlighted. The dynamic shared protein compositions of VLDL, LDL, and HDL from golden hamster plasma under normal and hyperlipidemic states were confirmed and compared by parallel reaction monitoring (PRM)-based targeted proteomics. This study was aimed to determine the relationship between the protein heterogeneity and functions of the three lipoproteins under normal conditions and explore the functional implication of protein dynamic change during the hyperlipidemic state.

Project description:Cumulative research demonstrated that heart muscles in the dilated cardiomyopathy (DCM) patients are marked by down-regulated expression of α-myosin heavy chain (α-MHC) coding by MYH6 while it remains unknown whether this reduction contributes to DCM. In our work, we proved that the expression level of α-MHC was decreased in the heart of DCM patients compared to that of non-failure donors. We established two Myh6 down-expressed Syrian Hamster models: one with α-MHC deficiency across all systems, the other with α-MHC knockdown specifically in the heart by using adeno-associatedvirus (AAV). Besides the animal models, we also programmed MYH6 knockout human-induced pluripotent stem cells (hiPSCs) which differentiated later into cardiomyocytes (hiPS-CMs) in vitro. We found α-MHC deficiency in vivo or in vitro both resulted in DCM phenotypes, meanwhile we demonstrated α-MHC deficiency is a causative factor of DCM. We also proved that the DCM-associated α-MHC deficiency was mediated by the autophagy flux. At the same time, the DCM phenotypes were attenuated by rapamycin, an autophagy activator, and exacerbated by inhibiting autophagic activities. Transcriptomic and proteomics results revealed that cardiomyocytes with α-MHC deficiency apparently exhibited actin cytoskeleton dysfunction. Our data demonstrated that α-MHC deficiency influenced the actin cytoskeleton network to regulate the autophagy activity suggesting that overexpress α-MHC or regulating autophagy would be a promising therapeutic candidate for DCM.

piRNA pathway is essential for generating functional oocytes in golden hamster

Project description:Piwi-interacting RNAs (piRNAs) are small RNAs predominantly expressed in germ cells that function in gametogenesis in various species. Notably, PIWI-deficient female mice are fertile and mouse oocytes express a panel of small RNAs that do not appear widely representative of mammals. Thus, the function of piRNAs in mammalian oogenesis remains largely elusive. Here, we generated PIWIL1- and MOV10L1-deficient golden hamsters and found that all female and male mutants were sterile, with severe defects in embryogenesis and spermatogenesis, respectively. PIWIL1-deficient oocytes displayed aberrant transposon accumulation and broad transcriptomic dysregulation, and maternal PIWIL1-deficient embryos had impaired zygotic genome activation (ZGA). Moreover, our results support that PIWIL1-piRNAs exert a non-redundant function in silencing endogenous retroviruses (ERVs) in oocytes. Together, our findings demonstrate that piRNAs are indispensable for generating functional germ cells in golden hamsters and show the informative value of this model species for functional and mechanistic investigations of piRNAs in gametogenesis, especially those related to female infertility.

Andrographis paniculata standardized extract attenuates transcriptome, proteome, and metabolome changes caused by SARS-CoV-2 infection in golden hamster kidney

Project description:SARS-CoV-2 was the cause of the COVID-19 global pandemic that resulted in millions of deaths worldwide. SARS-CoV-2 infection is often accompanied by renal manifestations such as acute kidney injury and kidney fibrosis, however there is a lack of effective treatment options that also address renal involvement. In this study, golden hamsters were inoculated with Delta variant SARS- CoV-2, and given either vehicle or Andrographis paniculata (AP) standardized extract as treatment. Hamster kidney tissues were then subjected to transcriptomics, proteomics, and metabolomics analysis. Results showed that AP potentially ameliorates kidney fibrosis mediated by ECHS1-induced mTOR activation and LHX1-HDAC8 promotion. This was supported by observations of lipid metabolism alterations that may have been a result of mTOR activation in metabolomics study. Transcriptomics analysis revealed less enrichment of viral infection pathways in AP-treated groups. Overall, AP appears to attenuate SARS-CoV-2 infection and development of kidney fibrosis.

The non-redundant functions of PIWI family proteins in gametogenesis in golden hamsters

Project description:The piRNA pathway is essential for female fertility in golden hamsters and likely humans, but not in mice. However, the role of individual PIWIs in reproduction remains poorly understood outside of mice. Here, using golden hamsters, we establish dynamic expression profiles and subcellular localization for all four PIWIs and characterize their associated reproductive defects in knockout mutants. In female golden hamsters, PIWIL1 and PIWIL3 are highly expressed throughout oogenesis and early embryogenesis, while PIWIL1 knockout leads to sterility, and PIWIL3 deficiency results in subfertility with lagging zygotic development. PIWIL1 can partially compensate for TE silencing and transcriptional regulation in PIWIL3 knockout females, but not vice versa. PIWIL1 and PIWIL4 are the predominant PIWIs expressed in adult or postnatal testes, respectively, while PIWIL2 is present in both stages. Notably, in golden hamsters, none of the differences were found between pre-pachytene and pachytene piRNAs characteristic of mice. Loss of any PIWI expressed in testes leads to sterility and severe but distinct spermatogenesis disorders, which are markedly less severe in mice. These findings expand our understanding of the non-redundant PIWI-piRNA regulatory functions in gametogenesis and early embryogenesis in golden hamster, increasing the value of this model for studying human fertility.
