Project description:Chemical investigation of Cordia millenii, Baker resulted in the isolation of a new depsidone, cordidepsine (1), along with twelve known compounds including cyclooctasulfur (2), lup-20(29)-en-3-triacontanoate (3), 1-(26-hydroxyhexacosanoyl)glycerol (4), glyceryl-1-hexacosanoate (5) betulinic acid (6), lupenone (7), ?-amyrone (8), lupeol (9), ?-amyrin (10), allantoin (11), 2'-(4-hydroxyphenyl)ethylpropanoate (12) and stigmasterol glycoside (13). Hemi-synthetic reactions were carried out on two isolated compounds (5 and 6) to afford two new derivatives, that is, cordicerol A (14) and cordicerol B (15), respectively. The chemical structures of all the compounds were established based on analysis and interpretation of spectroscopic data such as electron ionization mass spectrometry (EI-MS), high resolution electrospray ionization mass spectrometry (HR-ESI-MS), fast atom bombardment mass spectrometry (FAB-MS), one dimension and two dimension nuclear magnetic resonance (1D and 2D-NMR) spectral data as well as X-ray crystallography (XRC). Lupeol ester derivatives [Lup-20(29)-en-3-triacontanoate (3)], monoglycerol derivatives [1-(26-hydroxyhexacosanoyl)glycerol (4) and glyceryl-1 hexacosanoate (5)] were isolated for the first time from Cordia genus while sulfur allotrope [cyclooctasulfur (2)] was isolated for the first time from plant origin. Biological assays cordidepsine (1) exhibited significant anti-HIV integrase activity with IC50 = 4.65 ?M; EtOAc extract of stem barks, EtOAc fraction of roots and leaves were not toxic against 3T3 cells.

Project description:Firensia Scop. was based on Cordia flavescens Aubl., a species described and illustrated from a mixed collection that Scopoli never transferred to Firensia. The genus included three additional species formally named by Rafinesque. Currently the four species are placed in three different families and none retained the epithet accepted by Scopoli or given by Rafinesque for reason of priority. A lectotype is designated for Cordia flavescens that places Firensia in the synonymy of Ocotea (Lauraceae).

Flower, fruit phenology and flower traits in Cordia boissieri (Boraginaceae) from northeastern Mexico.

Project description:We characterized variations in Cordia boissieri flowers and established if these variations occur between plants or between flowering events. Flowering and fruiting was measured for 256 plants. A GLM test was used to determine the relationship between flowering and fruit set processes and rainfall. We performed measurements of floral traits to detect variations within the population and between flowering events. The position of the anthers with respect to the ovary was determined in 1,500 flowers. Three out of four flowering events of >80% C. boissieri plants occurred after rainfall events. Only one flowering event occurred in a drought. Most plants flowered at least twice a year. The overlapping of flowering and fruiting only occurred after rainfall. Anthesis lasted three-to-five days, and there were two flower morphs. Half of the plants had longistylus and half had brevistylus flowers. Anacahuita flower in our study had 1-4 styles; 2-9 stamens; 6.5-41.5 mm long corolla; sepals from 4.5-29.5 mm in length; a total length from 15.5-59 mm; a corolla diameter from 10.5-77 mm. The nectar guide had a diameter from 5-30.5 mm; 4-9 lobes; and 5 distinguishable nectar guide colors. The highest variation of phenotypic expression was observed between plants.

Phytochemical Investigation of <i>Cordia africana</i> Lam. Stem Bark: Molecular Simulation Approach.

Project description:BackgroundThe current work planned to evaluate Cordia africana Lam. stem bark, a traditionally used herb in curing of different ailments in Africa such as gastritis and wound infections, based on phytochemical and antibacterial studies of two pathogenic microorganisms: methicillin-resistant Staphylococcus aureus (MRSA) and Helicobacter pylori.MethodsHigh performance liquid chromatography (HPLC) profiling was used for qualitative and quantitative investigation of the ethanol extract. The minimum inhibitory concentration (MIC) of the ethanolic extract and isolated compounds was estimated using the broth microdilution method and evidenced by molecular dynamics simulations.ResultsFour compounds were isolated and identified for the first time: α-amyrin, β-sitosterol, rosmarinic acid (RA) and methyl rosmarinate (MR). HPLC analysis illustrated that MR was the dominant phenolic acid. MR showed the best bacterial inhibitory activity against MRSA and H. pylori with MIC 7.81 ± 1.7 μg/mL and 31.25 ± 0.6, respectively, when compared to clarithromycin and vancomycin, respectively.ConclusionThe antibacterial activity of the stem bark of Cordia africana Lam. was evidenced against MRSA and H. pylori. Computational modeling of the studied enzyme-ligands systems reveals that RA and MR can potentially inhibit both MRSA peptidoglycan transpeptidases and H. pylori urease, thereby creating a pathway via the use of a double target approach in antibacterial treatment.
