Project description:A one of general treatments of patient with acute promyelocytic leukemia (APL) is performed a high dose rate of total-body irradiation therapy. However, the appearance of radioresistant cell that is rarely expressed induces a harmful outcome even performing radiotherapy. We have been established a radioresistant APL cell model (Resistant type), and some characteristics was clarified. But little is known as to the micro RNAs (miRs) expression profile in this cell. In the present study, we analyzed on the expression profile of miRs included in small RNAs using miRNA microarray, The focused miRs that is enabled to investigate by normalization was 1187 molecules. Among these miRs, 27 miRNAs (10 up-regulated miRs and 17 down-regulated miRs) in Res-HL60 showed a p-value < 0.05, Fold change > 1.5 or Fold change < 0.66 in comparison to Wt-HL60. In addition, five miRs were reproducible in RT-qPCR (miR-146a-5p, miR-30c-1-3p, miR-671-5p, miR-610, miR-3675-5p). miRNA-target gene regulation networks were looked for the five miRNAs using OmicsNet 2.0. Interestingly, the five miRNAs were Superoxide Dismutase 2 (SOD2) mRNA. These results were suggested that radioresistant leukemia cell acquires antioxidant system through from specific miRs endogenously.

Transcription profiling of human leukemia HL60-NG cell and HL60-RG cell

Genomic Analysis of effect of bufalin on HL60 Cells

Project description:Bufalin, a natural toxin isolated from the Chinese toad venom preparation Chan ’su, has been used in leukemia therapy in the mainland China by inducing human leukemia cells to differentiation and apoptosis. In the present study, oligonucleotide microarrays were used to assess profiles of target gene regulation at several points at a 48h period by bufalin at an appropriate concentration in the human promyelocytic leukemia cell line HL60. Data from the gene expression profiles indicated that bufalin altered the expression of 1206 genes which are distributed in multiple profiles of regulation. Results are consistent with bufalin regulating HL60 cells proliferation and differentiation as well as apoptosis. Keywords: time course

Transcriptional Regulation of HL60 Neutrophil Differentiation: Promyelocyte trajectory vs. Neutrophil trajectory

Project description:Using the HL60 multipotent promyelocytic leukemia cell line, we performed experiments that lead to two different cell fate attractors by treatments of varying dosage and duration of the differentiation agent all-trans-retinoic acid (ATRA). The dosage and duration combinations corresponding to the two treatments were chosen by means of of cytometric measurements of CD11b, a well-known early differentiation marker, such that the two populations are poised at the same stage of differentiation. Using the selected treatment combinations, one population proceeds toward the terminally differentiated neutrophil attractor while the other population reverts back toward the undifferentiated promyelocytic attractor. We monitored the gene expression changes in the two populations after their respective treatments over a period of five days and identified a set of genes that diverged in their expression; a subset of which promotes neutrophil differentiation while the other represses cell cycle progression. By employing promoter based transcription factor binding site analysis, we found enrichment of transcription factors functionally linked to tumor progression, cell cycle, and development. Keywords: Time course, dose response Untreated HL60 cells are hybrized on the channel 1 with Cy3, ATRA-Treated HL60 cells are hybridized on channel 2 with Cy5. HK60 cells were treated with high dosage/short duration and low dosage/long duration to achieve 55% of CD11b+ cells. These positive cells were then isolated and culture in fresh media and total RNA were isolated for comparison. Cells treated with high dosage/short duration treatment proceed toward the neutrophil attractor while cells treated with the low dosage/long duration treatment revert back toward the promyelocyte attractor.
