Project description:This study investigates the role of nucleos(t)ide metabolism, transport, and adenosine signaling in placental development and spontaneous preterm birth (PTB). We analyzed gene expression in human placentas, primary trophoblasts, BeWo cells and rat placentas across different gestational stages to understand metabolic adaptations and stress responses To understand the role of nucleos(t)ide metabolism (de novo synthesis and salvage patways), transport, and adenosine signaling in placental development and the adaptive response during spontaneous preterm birth (PTB), we analyzed gene expression in first-trimester and term human placentas, PTB placentas, primary human trophoblasts, and BeWo cells. Additionally, we examined rat placentas at different gestation days (GD12, GD15, and GD20) to provide developmental context and validate findings from human samples. Our results highlight the upregulation of nucleos(t)ide metabolism and adenosine signaling during placental growth and trophoblast differentiation (cytotrophoblast to syncytiotrophoblast), with further metabolic shifts in PTB placentas, particularly in pyrimidine de novo synthesis, purine salvage, and adenosine metabolism, suggesting an adaptive stress response. Additionally enhanced and also adenosine signaling and nucleoside transport was observed. This integrated approach provides novel insights into the metabolic regulation of placental function under both normal and pathological conditions

Project description:Preterm birth, defined as birth <37 weeks of gestation, is a leading cause of infant morbidity and mortality. In the United States, approximately 12% of all births are preterm.1 Despite decades of research, there has been little progress in developing effective interventions to prevent preterm birth. In fact, the rate of preterm birth has increased slightly over the last several decades.2 The ultimate goal of the Genomic and Proteomic Network for Preterm Birth Research (GPN-PBR) is to identify possible biomarkers that could predict the susceptibility to spontaneous preterm birth (SPTB) as well as to shed light on the molecular mechanisms involved in its etiologies. Understanding those mechanisms will help us predict SPTB and may facilitate the introduction of more effective prevention and treatment strategies.

Project description:The use of proteasome inhibitors (PIs) is the backbone of multiple myeloma (MM) treatment. However, almost all MM patients who initially respond to PIs eventually develop resistance to these drugs. The discovery of a pharmacological intervention that increase the potency of PIs in order to reduce their dose and/or restore the sensitivity of MM cells to PIs is an important avenue in MM research. Insulin Degrading Enzyme (IDE) is a druggable protease known to modulate proteasome. We show here that, in two independent cohorts, a high expression of IDE in cells from MM patients is associated with shorter overall survival. Likewise, we designed specific, cell permeable, IDE inihibitors and showed that full pharmacological engagement of IDE is associated with enhanced sensitivity to PIs of MM cell lines, as well as of primary patient MM cells. Furthermore, treatment of PI-resistant MM cells with the inhibitor overcomes their resistance to PI. Finally we designed an improved IDE inhibitor suitable for in vivo pharmacology. This inhibitor which develops multiple interactions with IDE at the catalytic site and display good pharmacocinetic properties, boosts the anti-myeloma potency of bortezomib in a syngenic mouse model of MM. In vitro, it appears that IDE inhibition reduces the residual proteasome activty in PI-treated cells and increases the apoptotic potency of carfilzomib through induction of an Integrated Stress Response associated with strongly reduced IGF1 and IGF1R expressions. The pharmacological profile of this new combination appears highly desirable for the treatment of MM.

Project description:Preterm birth is the major cause of newborn and infant mortality affecting nearly one in every ten live births. This study was designed to develop an epigenetic biomarker for susceptibility of preterm birth using buccal cells from the mother, father, and child (triads). MeDIP-seq was used to identify differential DNA methylation regions (DMRs) using a comparison of control term birth versus preterm birth triads. Epigenetic DMR associations with preterm birth were identified for both the mother and father that were distinct and suggest potential epigenetic contributions from both parents. The mother (165 DMRs) and female child (136 DMRs) at p<1e-04 had the highest number of DMRs and were highly similar suggesting potential epigenetic inheritance of the epimutations. The male child had negligible DMR associations. The DMR associated genes for each group involve previously identified preterm birth associated genes.

Project description:Preterm birth has dramatically increased within the population (i.e., >10%) and preeclampsia is one of the most unanticipated sub-categories of preterm birth. No clinical parameter or biomarkers exists for preeclampsia preterm birth, except for high blood pressure toward the end of pregnancy. The current study investigates the potential use of epigenetic (DNA methylation) alterations as a preeclampsia biomarker. Non-preeclampsia term births were compared to preeclampsia preterm births to identify methylation differences. Buccal cell cheek swabs were used as a marker cell for systemic epigenetic alterations in the individuals. A total of 389 differential DNA methylation regions (DMRs) were identified and associated with the presence of preeclampsia. The DMRs were genome-wide and were predominantly low CpG density (<2 CpG/100 bp). In comparison with a previous preterm birth epigenetic biomarker there was a 15% (60 DMR) overlap, indicating the majority of the DMRs are unique for preeclampsia.
