Project description:In this study we analyzed the effect of overexpression of an HA-tagged version of the ERF RAP2.12 on the transcriptome levels in aerobic and hypoxic-treated (O2 21% and 1%, respectively) Arabidopsis thaliana rosettes. We also analyzed the effect of a RAP2.12 and RAP2.2 simultaneous silencing in aerobic and hypoxic-treated (O2 21% and 1%, respectively) Arabidopsis thaliana rosettes. We treated Arabidopsis Col-0 (wt) rosettes and transgenic HA::RAP2.12 and amiRAP2.2-12 , 5-week old, grown in 8/16 light/dark photoperiod with: -Control (22°C, dark, 21% O2, 1.5h). -Hypoxia (22°C, dark, 1% O2, 1.5h).

In this work we analyzed the effect of ectopic expression of HRE1 and HRE2 on the arabidopsis transcriptome in aerobic and hypoxic (1% O2) conditions. While HRE1 has a moderate effect on the expression of anaerobic genes under hypoxia, HRE2 does not affect them either under aerobic or hypoxic conditions. Experiment Overall Design: We treated Arabidopsis seedling Col-0 (wt) and transgenic 35S::HRE1 and 35S::HRE2 , 7-day old, germinated in 12/12 light/dark photoperiod with: Experiment Overall Design: -Control (23°C, dark, 21% O2, 4h). Experiment Overall Design: -Hypoxia (23°C, dark, 1% O2, 4h). Experiment Overall Design: Two biological replicates for each condition.

Project description:The present study reports the gene expression data of Mycobacterium tuberculosis H37Rv and H37RvΔdosSΔdosT (DKO) grown on 0.2 % acetate/glucose under aerobic/hypoxic conditions. Acetate was reported to be present in granulomas of Mycobacterium tuberculosis infected guinea pigs which are also hypoxic. By exposing Mycobacterium tuberculosis H37Rv and H37RvΔdosSΔdosT to different combinations of granulomatous stresses (acetate/glucose and aerobic/hypoxic conditions) alongwith other experimental data, we were able to delineate a new signaling pathway that activates DevR (DosR) regulon through Acetyl phosphate. The presence of two pathways highlights the importance of targeting DevR and not DevS/DosT for intercepting DevRST signalling cascade.
