RNA samples from independently induced cultures, plus fibroblast and human dermal microvascular endothelial cultures were converted into individual cDNA libraries using Illumina TruSeq methods and subjected to single-end 50 base-sequence analysis at 20-30 million read depths.

Project description:In order to understand the mechanisms underlying the effects of itraconazole in infantile hemangioma endothelial cells, we further explored the potential targeting genes of itraconazole in infantile hemangioma endothelial cells in vitro by using a genome-wide gene expression array. In total 172 mRNAs were up-regulated (fold change > 2) and 819 mRNAs were down-regulated (fold change > 2) upon itraconazole treatment in infantile hemangioma endothelial cells. The top signaling pathway closely related to differentially expressed genes upon itraconazole treatment is the cytokine and cytokine receptor pathway. Within this pathway, the mRNA expression of PDGF-D, an important growth factor belonging to the PDGF family, was the most significantly reduced (30 folds) among 991 differentially expressed genes after itraconazole treatment. qRT-PCR and western blotting confirmed the inhibitory effects on PDGF-D levels.

Project description:As the most common vascular tumor during infancy, infantile hemangioma (IH) is clinically featured by a rapid proliferation phase of disorganized blood vessels and a subsequent spontaneous involution phase. Infantile hemangioma arises from a unique type of multipotent stem cells called hemangioma stem cells (HemSCs), which could differentiate into endothelial cells, pericytes and adipocytes in IH. However, the underlying mechanisms that regulate the cell fate determination of HemSCs are not well elucidated. Here, we identified KLF2 as a candidate transcription factor involved in the control of HemSCs differentiation. KLF2 was expressed in endothelial cells in proliferating IH and its expression diminished in adipocytes in involuting IH. KLF2 regualtes the proliferation, apopotosis and cell cycle progression in HemSCs. Moreover, KLF2 is a critical regulator in HemSCs that control their differentiation direction between endothelial cells and adipocytes. Knockdown of KLF2 inhibited the formation of blood vessels in vivo while accelerated the progress of adipogenesis. RNA-seq analysis suggested an induction of pro-adipogenic transcriptome in HemSCs upon KLF2 knockdown. Our data showed that KLF2 exhibited pleiotropic effects in regulating the biological behaviours of HemSCs, and was involved in the progression and involution of IH via determining the cell fate of HemSCs.

This study sought to identify endothelial BMP9-regulated proteins that could affect the HHT phenotype. Gene ontology analysis of cDNA microarray data obtained following BMP9 treatment of primary human endothelial cells indicated regulation of chemokine, adhesion, and inflammation pathways.
