T. atroviride is considered a necrotrophic mycoparasite and has developed the ability to kill other organisms and obtain nutrients from them. The object of this work is to explore the role of small RNAs in mycoparasitism. To this end, we obtained small RNA-Seq libraries from the interactions of T. atroviride against Alternaria alternata. The libraries were obtained from three stages during mycoparasitsm: before contact (BC), contact (C), and after contact (AC).

Project description:Fungi as animals and plants conserves the ability to respond to stressful environmental conditions as biotic an abiotic stress. A correct injury response in Eukaryotes including fungi is carried out thought the activation of signaling pathways Ca2+ related dependent of DAMPs/MAPMPs recognizing for the activation of the innate immune system. During herbivory, plants, in addition to activating pathways related to injury, activates specific responses to combat its predator. Using a transcriptional approach, here, we reported the capacity of the filamentous fungus T. atroviride to activate or not a specific response to injury and predation to different predators also, we provide evidence of how fungivory by Drosophila melanogaster larvae inhibits the reported injury response for T. atroviride, although this predation negatively affects the hyphal regeneration thought the larval salivary compounds. Also, the activation of a putative alternate injury reproduction pathway is proposed.
Project description:Mycoparasitism is a key feature of Trichoderma biocontrol agents. Recent studies of intracellular signal transduction pathways of the potent mycoparasite Trichoderma atroviride revealed the involvement of Tmk1, a mitogen-activated protein kinase, in triggering the mycoparasitic response. We previously showed that mutants missing Tmk1 exhibit reduced mycoparasitic activity against several plant pathogenic fungi. In this study we identified the most robustly regulated targets governed by Tmk1 during mycoparasitism by whole transcriptome gene expression profiling using a custom microarray.
Project description:Eight new and four known peptaibols were isolated from a strain of the fungus, Trichoderma atroviride (NF16), which was cultured from an Axinellid sponge collected from the East Mediterranean coast of Israel. The structures of the pure compounds were determined using HRMS, MS/MS and one- and two-dimensional NMR measurements. The isolated compounds belong to the trichorzianines, a family of 19-residue linear hydrophobic peptides containing a high proportion of ?-aminoisobutyric acid (Aib), an acetylated N-terminus and a C-terminal amino alcohol. These new peptaibols exhibited antimicrobial activity against environmental bacteria isolated from the Mediterranean coast of Israel.

Project description:The morphology of filamentous fungi has a close relationship with the production of many products (e.g. industrial enzymes). In this study, we deleted gul1, which encodes a putative RNA-binding protein, in cellulolytic fungus Trichoderma reesei. The mutant showed different morphologies both on agar plates and in liquid cultures compared with its parent. RNA-seq study showed that the expression levels of genes in many biological processes were affected by gul1 deletion.
