Project description:The model was constructed to describe TLR4 induced NF-κB activation in native bone marrow-derived macrophages. It included processes of ligand (lipopolysaccharide) recognition, formation of dimer receptor complex and further signal transduction through TRAF6/TAK1 complex that leads to the activation of IKKα/β kinase, which in turn enables the NF-κB transcription factor phosphorylation and translocation in the cell nucleus, and induction of IkB and WIP1 (as an example of induced protein that promotes NF-κB dephosphorylation 2) gene transcription. Models were based on the current knowledge of TLR signaling framework, protein interactions within the TLR4 pathway, and up-to-date mathematical models describing Toll receptor activation.

Project description:TLR4/NF-κB signaling plays a central mediator in response to danger signals released in the muscle ischemia-reperfusion injury (IRI). This study was designed to profile TLR4/NF-κB-responsive microRNAs (miRNAs) in the skeletal muscles following IRI. Following 2 h of ischemia and subsequent reperfusion for indicated times (0 h, 4 h, 1 d, and 7 d) of the isolated thigh skeletal muscles based on femoral artery perfusion of C57BL/6, Tlr4–/–, and NF-κB–/–mice, the muscle specimens were analyzed with an miRNA array to detect the TLR4/NF-κB-responsive miRNAs.

Project description:This experiment was undertaken to document changes in gene expression in the skin of tick-resistant Brahman (Bos indicus) and tick-susceptible Holstein-Friesian (Bos taurus) cattle prior to, and following, infestation with the cattle tick Rhipicephalus (Boophilus) microplus Experiment Overall Design: RNA was extracted from skin samples of tick-naïve cattle (animals with no previous R.microplus exposure) and tick-infested cattle after a period of successive, heavy infestations with R. microplus. Skin samples taken from tick-infested animals were taken at sites where tick larvae (approximately 24 h old) were attached to the skin sample. Skin samples were of 8 mm diameter and full skin thickness (approximately 10 mm). RNA samples from 12 animals (3 tick-naive Holstein-Friesian, 3 tick-naive Brahman, 3 tick-infested Holstein-Friesian and 3 tick-infested Brahman) were processed and hybridised to individual slides.
Project description:This trial was undertaken to examine the perhipheral cellular and antibody response of cattle following infestation with the cattle tick, Rhipicephalus microplus. The information from the Affymetrix gene expression data is used to complement other measurements of immune function such as cellular subset composition and antibody response in cattle of high (Brahman) and low (Holstein-Friesian) resistance to the cattle tick. Experiment Overall Design: RNA was extracted from white blood cells during a period of successive, heavy infestations with R. microplus. RNA samples from 3 Holstein-Friesian and 3 Brahman animals were analysed on individual slides.
Project description:Bovine mastitis is an inflammatory disease of the mammary gland with serious economic implications for dairy industries worldwide. We performed total RNA sequencing using whole blood cells collected from multiparous Holstein Friesian dairy cows with naturally occurring mastitis to investigate the changes in systemic gene expression and their association with inflammatory responses. Some related sequencing data are deposited in E-MTAB-9347 and E-MTAB-9348.
