Project description:Purpose: To disclose the molecular mechanisms of aging-related functional impairments of the rat bladder, and to determine whether long-term caloric restriction (CR) may have preventive effects on these mechanisms. Materials and Methods: Male Fischer 344 rats were divided into three groups: young (6 months-old) fed ad libitum (Y, N = 16), old (25-28 months-old) fed ad libitum (O+AL, N = 15), and old (25-28 months-old) fed restrictedly three days a week since 6 weeks-old (O+CR, N = 16). cDNA microarray analysis and real-time polymerase chain reaction (RT-PCR) of the bladder and L6 dorsal root ganglia (L6DRG) were performed. The bladder was subjected to oxidative stress measurement and immunohistochemistry. Results: In the O+AL group, 83 genes in the bladder and 48 genes in the L6DRG were up-regulated than compared with those of the Y and O+CR groups (fold change > 2). These genes were mostly related to immune- and inflammatory-responses. Immunohistochemistry showed that Granzyme B, a cytotoxic T-lymphocyte-associated serine protease, and matrix metalloproteinase 13 (Mmp13), an interstitial collagenase, were more strongly expressed in the bladder of the O+AL than the Y and O+CR groups. The level of Malondialdehyde (MDA), an oxidative stress marker, was higher in the O+AL than the Y group, whereas there were no significant differences between the O+CR and Y groups. Conclusions: In the rat, aging is associated with up-regulation of immune and inflammatory genes in the bladder and DRG as well as with oxidative and fibrotic changes of the bladder. Long-time CR reduced these aging-related changes.

Project description:Astrocytes are key cells in brain aging, helping neurons to undertake healthy aging or otherwise letting them enter into a spiral of neurodegeneration. We aimed to characterize astrocytes cultured from senescence-accelerated prone 8 (SAMP8) mice, a mouse model of brain pathological aging, along with the effects of caloric restriction, the most effective rejuvenating treatment known so far. Analysis of the transcriptomic profiles of SAMP8 astrocytes cultured in control conditions and treated with caloric restriction serum was performed using mRNA microarrays. A decrease in mitochondrial and ribosome mRNA, which was restored by caloric restriction, confirmed the age-related profile of SAMP8 astrocytes and the benefits of caloric restriction. An amelioration of antioxidant and neurodegeneration-related path- ways confirmed the brain benefits of caloric restriction. Studies of oxidative stress and mitochondrial function demonstrated a reduction of oxidative damage and partial improvement of mito- chondria after caloric restriction. In summary, caloric restriction showed a significant tendency to normalize pathologically aged astrocytes through the activation of pathways that are protective against the age-related deterioration of brain physiology. Primary cultures enriched in astrocytes were obtained from cerebral cortical tissue from 2-day-old SAMP8 and SAMR1 mice. Astrocyte cultures were established and experiments were routinely carried out after 21 days in culture. Established astrocyte cultures of both SAMR1 and SAMP8 consisted of 85-90% astrocytes, 10-15% microglia and 0.1-1% oligodendroglia. Sera from rats subjected to ad libitum (AL) diet and to CR were obtained as described for the establishment of the CR in vitro model (de Cabo et al., 2003). Serum was heat inactivated at 56°C prior to use in astrocyte culture experiments. Treatment in vitro was performed by adding 10% volume CR or AL serum onto the astrocyte culture medium for 48 h, the cells were harvested and RNA was extracted for the microarray studies. Three biological replicates for each condition were done and RNA was extracted for the microarray studies. Please note that SAM models were developed from AKR/J by Kyoto University. Five litters with severe senescence were selected to further propagate and examine these characteristics. Litters that showed normal aging were selected as a senescence-resistant series (R-series). The genetic background of the SAM mice became suspect after the pathological findings were different from the AKR/J mouse. Each SAM model is genetically different. Each SAM colony was acquired by Harlan by Takeda Chemical Ltd. in 2002. Project description:Benign prostatic hyperplasia and related lower urinary tract symptoms remain common, costly, and impactful issues for aging males. Etiology and pathogenesis are multifactorial and include steroid hormone changes and inflammation. Noninvasive markers could one day inform personalized medicine, but interindividual variation and lack of healthy age-matched controls hamper research. Experimental models are appealing for insight into disease mechanisms. Here, we present a spatiotemporal proteomics study in a mouse model of hormone-induced urinary dysfunction. Urine samples were collected noninvasively across time: before, during, and after disease onset. Microcomputed tomography analysis implicated the prostate as a spatially relevant contributor to bladder outlet obstruction. Prostates were collected after disease onset and compared with control mice. Notable changes in urine include proteins representing oxidative stress defense and acute phase inflammatory response processes. In the prostate, hormone treatment led to perturbations related to oxidative stress response and H2O2 metabolism. Several protein changes coincided in both urine and prostate tissue, including Ctsb, Qsox1, and Gpx3. This study supports the concept of noninvasive urinary biomarkers for prostate disease diagnostics. Oxidative stress and acute phase inflammatory processes were identified as key consequences of hormone-induced bladder outlet obstruction. Future research into antioxidants and anti-inflammatories in prostate disease appears promising.
