Project description:The role of PPARβ/δ in maintaining skin homeostasis during skin injury or inflammation has been widely studied. However, the majority of these reports based their studies in PPARβ/δ found in keratinocytes, which is the major cell type of the skin epidermis. The skin is mainy made up of the epidermis and dermis, and skin homeostasis is tightly regulated by complex crosstalks between epidermis and dermis. The dermis is predominantly made up of fibroblasts, and the predominant PPAR subtype in dermal fibroblasts is PPARβ/δ. Knowledge in the role of fibroblasts PPARβ/δ in skin homeostasis is lacking. To identify gene changes leading to phenotypical and biological functions alterations upon deletion of fibroblasts PPARβ/δ, we performed a comparative microarray gene expression analysis between Pparb/d^fl/fl and FSPCre-Pparb/d^fl/fl mice.
Project description:To elucidate the mechanisms underlying the perturbation of tumor vascularization in Pparb/ mice we performed matrigel plug assays23. Matrigel containing prostaglandin E2 (PGE2) and basic fibroblast growth factor (FGF-2) was injected subcutaneously into both wild-type and Pparb/ mice where it formed semi-solid plugs. These plugs became rapidly invaded by AQP-1 positive cells. To analyze differences between Pparb+/+ and Pparb/ samples we compare the gene expression profile of both samples by microarray analysis.

Project description:Autologous skin grafting is a standard treatment for skin defects such as burns. No artificial skin substitutes are functionally equivalent to autologous skin grafts. The cultured epidermis lacks the dermis and does not engraft deep wounds. Although reconstituted skin, which consists of cultured epidermal cells on a synthetic dermal substitute, can engraft deep wounds, it requires the wound bed to be well-vascularized and lacks skin appendages. In this study, we successfully generate complete skin grafts with PSC-derived epidermis with appendages on p63 knockout embryos' dermis. Donor PSC-derived keratinocytes encroach the embryos' dermis by eliminating p63 knockout keratinocytes based on cell-extracellular matrix adhesion mediated cell competition. Although the chimeric skin contains allogenic dermis, it is engraftable as long as autologous grafts. Furthermore, we could generate semi-humanized skin segments by human keratinocytes injection into the amnionic cavity of p63 knockout mice embryos. Niche encroachment opens the possibility of human skin graft production in livestock animals.

Datasets were used to identify glycogen synthase 2 as novel PPAR target. Experiment Overall Design: Three month old male PPARb/d knock-out mice were on a mixed background (Sv129/C57BL/6). Wild-type littermates served as control animals. Animals received standard chow. Epididymal white adipose tissue (WAT) was removed and total RNA was isolated and pooled afterwards (five animals per group). Pooled RNA was hybridized to Affymetrix mouse genome 430 2.0 arrays arrays. Five microgram total RNA was labeled according to the ENZO-protocol, fragmented and hybridized according to Affymetrix protocols.
