Project description:Heterozygous mutations in HNF1B in humans result in a multi-system disorder, including pancreatic hypoplasia and diabetes mellitus. The underlying mechanisms that contribute to disease pathogenesis remain largely unknown, partially accounted by the fact that mouse models with heterozygous deletions in Hnf1b do not develop diabetes, in contrast to the phenotypes observed in MODY patients. Here we used a well-controlled human induced pluripotent stem cell pancreatic differentiation model to elucidate the molecular mechanisms underlying HNF1B-associated diabetes and pancreatic hypoplasia. Our results show that lack of HNF1B blocks specification of pancreatic fate from the foregut progenitor stage, but HNF1B haploinsufficiency allows differentiation of multipotent pancreatic progenitor cells and insulin secreting β-like cells. We further report that HNF1B happloinsuffiency impairs cell proliferation in foregut and multipotent pancreatic progenitors (MPCs). Our analyses suggest that this could be attributed to impaired induction of key pancreatic developmental genes, including FOXA1, SOX11, ROBO2, and additional TEAD1 target genes whose function could be associated with MPCs self-renewal. Taken together, these analyses have uncovered an exhaustive list of potential HNF1B gene targets during human pancreas organogenesis whose downregulation might underlie HNF1B-associated diabetes onset in humans, thus providing an important resource to understand the pathogenesis of this disease.

Project description:Patients with an HNF1B(S148L/+) mutation (MODY5) typically exhibit pancreatic hypoplasia. However, the molecular mechanisms are unknown due to inaccessibility of patient material and because mouse models do not fully recapitulate MODY5. Here, we differentiated MODY5 human-induced pluripotent stem cells (hiPSCs) into pancreatic progenitors, and show that the HNF1B(S148L/+) mutation causes a compensatory increase in several pancreatic transcription factors, and surprisingly, a decrease in PAX6 pancreatic gene expression. The lack of suppression of PDX1, PTF1A, GATA4, and GATA6 indicates that MODY5-mediated pancreatic hypoplasia is mechanistically independent. Overexpression studies demonstrate that a compensatory increase in PDX1 gene expression is due to mutant HNF1B(S148L/+) but not wild-type HNF1B or HNF1A. Furthermore, HNF1B does not appear to directly regulate PAX6 gene expression necessary for glucose tolerance. Our results demonstrate compensatory mechanisms in the pancreatic transcription factor network due to mutant HNF1B(S148L/+) protein. Thus, patients typically develop MODY5 but not neonatal diabetes despite exhibiting pancreatic hypoplasia.

Expression Data from HNF1b-knockdown RMG2-ovarian cancer cells

Project description:Ovarian clear cell carcinoma (OCCC) is a morphologically and biologically distinct subtype of ovarian carcinomas. We previously reported a gene expression profile characteristic of OCCC (OCCC signature), which contains hepatocyte nuclear facter-1b ( HNF-1b). To elucidate the biological role of HNF-1b in OCCC, we performed the suppression of the HNF-1b expression in human ovarian cancer cell line RMG2 using short hairpin RNA. We are now evaluating the functional effect using these cells. Affimetrix Human Genome U133A plus 2.0 chips was conducted for HNF1b knockdown and non-silencing cells (five replicates each for RMG2-HNF1b-sh1 and RMG2-HNF1b-sh2, ten replicates for the RMG2-control). All specimens were arrayed in parallel and used for RMA normalization.

Visceral Endoderm differentiation mediated by Hnf1B and is required for epiblast survival

Project description:E5.0 entails an intermediate state for primitive endoderm (PE) to visceral endoderm (VE) differentiation: a process required for proper embryonic development. Hnf1b is essential for primitive to visceral endoderm differentiation from E4.5 to E5.5. To understand development at E5.0 and pinpoint the role Hnf1b in proper embryonic development, embryos were harvest at E5.0 and single cells from three WT embryos and 3 Hnf1b KO embryos were isolated, dissociated, and scRNA sequenced using smartseq2. For experiments on WT embryos, CD1 females were crossed with CD1 or homozygous E-cadherin-GFP males. Hnf1b -/+22 line was derived through IVF of oocytes of Black6 mice with EMMA strain EM:07827(B6.129P2(D2)-Hnf1btm1Sce/Orl) Sperm, that was bought from the EMMA institution in Orleans.
