Project description:Coffee, after petroleum, is the most valuable commodity globally in terms of total value (harvest to coffee cup). Here, our bioeconomic analysis considers the multitude of factors that influence coffee production. The system model used in the analysis incorporates realistic field models based on considerable new field data and models for coffee plant growth and development, the coffee/coffee berry borer (CBB) dynamics in response to coffee berry production and the role of the CBB parasitoids and their interactions in control of CBB. Cultural control of CBB by harvesting, cleanup of abscised fruits, and chemical sprays previously considered are reexamined here to include biopesticides for control of CBB such as entomopathogenic fungi (Beauveria bassiana, Metarhizium anisopliae) and entomopathogenic nematodes (Steinernema sp., Heterorhabditis). The bioeconomic analysis estimates the potential of each control tactic singly and in combination for control of CBB. The analysis explains why frequent intensive harvesting of coffee is by far the most effective and economically viable control practice for reducing CBB infestations in Colombia and Brazil.
Project description:Traditionally, the study of anatomy in insects has been based on dissection techniques. Micro-computed tomography (micro-CT) is an X-ray based technique that allows visualization of the internal anatomy of insects in situ and does not require dissections. We report on the use of micro-CT scans to study, in detail, the internal structures and organs of the coffee berry borer (Hypothenemus hampei), the most damaging insect pest of coffee worldwide. Detailed images and videos allowed us to make the first description of the aedeagus and the first report of differences between the sexes based on internal anatomy (flight musculature, midgut shape, hindgut convolutions, brain shape and size) and external morphology (lateral outline of the pronotum and number of abdominal tergites). This study is the first complete micro-CT reconstruction of the anatomy of an insect and is also the smallest insect to have been evaluated in this way. High quality rendered images, and additional supplementary videos and 3D models are suitable for use with mobile devices and are useful tools for future research and as teaching aids.
Project description:The coffee industry loses millions of dollars annually worldwide due to the Coffee Berry Borer (CBB); these losses imply a decrease in quality and production. Traps are used to monitor their flight and for pest control. The main objective of this study was to determine the capture pattern and trap capture percentages of the CBB population over time using column traps (CTs) in two independent field experiments. CTs were composed of four traps installed at four different heights 0.5, 1.5, 2.5, and 3.5 m above ground. Our results demonstrated a significant difference in CBB capture by traps placed at different heights above the ground. The CT capture maintained a pattern throughout this study's lag: the lower the height, the greater the percentage of CBBs captured. The study was conducted in two independent experiments (A and B). In Experiment A and B, the traps placed at 0.5 m caught 67% and 85% of the CBBs captured, respectively. Furthermore, the trap set at 1.5 m above the ground in the multi-level CT showed a higher capture percentage than the single placed trap (ST, also at 1.5 m about ground). The pattern of the capture and proportion of the CBB in the CTs was maintained throughout the study despite the season, changes in temperature, and relative air humidity. We suggest that CTs could be explored as a useful tool for capturing the CBB, considering its monitoring and management.

Project description:Microbiota of the Coffee Berry Borer (Hypothenemus hampei)
Project description:BACKGROUND: The coffee berry borer, Hypothenemus hampei, reproduces and feeds exclusively on the mature endosperm of the coffee seed, which has a cell wall composed mainly of a heterogeneous mixture of hemicellulose polysaccharides, including arabinoxylans. Xylanases are digestive enzymes responsible for the degradation of xylan based polymers, hydrolyzing them into smaller molecules that are easier to assimilate by insects. We report the cloning, expression and enzymatic characterization of a xylanase gene that was identified in the digestive tract of the coffee berry borer. METHODS: The complete DNA sequence encoding a H. hampei xylanase (HhXyl) was obtained using a genome walking technique in a cDNA library derived from the borer digestive tract. The XIP-I gene was amplified from wheat (Triticum aestivum variety Soisson). A Pichia pastoris expression system was used to express the recombinant form of these enzymes. The xylanase activity and XIP-I inhibitory activity was quantified by the 3,5-dinitrosalicylic (DNS). The biological effects of XIP-I on borer individuals were evaluated by providing an artificial diet enriched with the recombinant XIP-I protein to the insects. RESULTS: The borer xylanase sequence contains a 951 bp open reading frame that is predicted to encode a 317-amino acid protein, with an estimated molecular weight of 34.92 kDa and a pI of 4.84. Bioinformatic analysis revealed that HhXyl exhibits high sequence homology with endo-?-D-xylanases of Streptomyces bingchenggensis from glycosyl hydrolase 10 (GH10). The recombinant xylanase showed maximal activity at pH 5.5 and 37°C. XIP-I expressed as a recombinant protein inhibited HhXyl activity in vitro and caused individual H. hampei mortality in bioassays when included as a supplement in artificial diets. CONCLUSION: A xylanase from the digestive tract of the coffee berry borer was identified and functionally characterized. A xylanase inhibitor protein, XIP-I, from wheat was shown to be a potent inhibitor of this xylanase, suggesting that its deployment has potential as a strategy to control coffee berry borer colonization of coffee plants.
