Project description:In this study we applied RNA-sequencing to a set of larvae-challenged elm trees that had either been untreated before or been exposed to prior egg deposition by elm leaf beetles. This dataset allowed us to characterize the global transcriptional response of egg-primed and non-primed elm trees at different time points after the priming stimulus itself and after the onset of larval feeding.

RNA-Seq of Arabidopsis thaliana and Arabidopsis lyrata plants primed by insect eggs from Pieris brassicae and triggered by insect herbivory

Project description:In Arabidopsis thaliana it is known that plants that harboured eggs of the White cabbage butterfly (Pieris brassicae) before larval feeding can defend better against the herbivore stress. As Arabidopsis thaliana is an annual plant we were interested in the plant defense and possible priming reaction of the closest biannual/perennial relative Arabidopsis lyrata. The main aim of the experiment was to compare the effect induced by insect egg deposition of Pieris brassicae between A. thaliana and A. lyrata plants grown in parallel on the transcriptional level. We used a full factorial setup consisting of a) untreated control plants , b) plants which experienced eggs for 6 days without larval feeding after that period, c) plants which experienced no eggs before larval feeding for 24 hours d) plants which experienced eggs for 6 days and larval herbivory for 24 hours. This setup was conducted with 7 week old vegetative plants For all treatments leaf tissue from the leaves that experienced egg oviposition and/or larval feeding were collected.

Gene expression in Arabidopsis leaves after P. brassicae egg deposition and/or herbivory by P. brassicae larvae

Project description:Plants can prepare their defense of impending herbivory, when they previously perceived eggs deposited by herbivores insect which reliably indicates that larvae will soon hatch and feed on the plants. Using Arabidopsis thaliana and Pieris brassicae we addressed the question whether prior egg deposition affects transcriptional changes in feeding-damaged plants. We determined the transcriptomes of Arabidopsis leaves that (i) were for six days deposited with eggs and suffered larval feeding for two days, (ii) suffered larval feeding for two days without prior egg deposition or (iii) were left untreated.

Solanum dulcamara's response to insect eggs comprises ovicidal hydrogen peroxide production

Project description:Plants can respond to insect oviposition but little is known about which responses directly target the insect eggs and how. Here, we reveal a mechanism by which the bittersweet nightshade, Solanum dulcamara, kills the eggs of a generalist noctuid herbivore. The plant responded at the site of oviposition by Spodoptera exigua with neoplasm and chlorotic tissue formation, accumulation of reactive oxygen species and induction of defence genes and proteins. Transcriptome analysis revealed that these responses were reflected in the transcriptional reprogramming of the egg-laden leaf. The plant-mediated egg mortality on S. dulcamara was not present on a genotype lacking chlorotic leaf tissue at the oviposition sites on which the eggs are exposed to less hydrogen peroxide. As exposure to hydrogen peroxide increased egg mortality, while catalase supplementation prevented the plants from killing the eggs, our results suggest that ROS formation directly acts as an ovicidal plant response of S. dulcamara.

Project description:Plants can cope with stress better if they experience a mild form of the stress before the actual \\"real\\" stress event. In Arabidopsis thaliana it is known that plants that harboured eggs of the White cabbage butterfly (Pieris brassicae) before larval feeding can defend better against the herbivore stress. The main aim of the experiment was to compare the priming effect induced by insect egg deposition of Pieris brassicae between vegetative and reproductive (first open flowers) Arabidopsis thaliana plants on the transcriptional level. We used a full factorial setup consisting of a) untreated control plants , b) plants which experienced eggs for 6 days without larval feeding after that period, c) plants which experienced no eggs before larval feeding for 24 hours d) plants which experienced eggs for 6 days and larval herbivory for 24 hours. This setup was conducted with 6 week old vegetative plants and 10 week old reproductive plants were the first flowers were open. For all treatments leaf tissue from the leaves that experienced egg oviposition and/or larval feeding were collected. From reproductive plants flower buds were collected as well.

Cellulose Nanofibers from a Dutch Elm Disease-Resistant Ulmus minor Clone.

Project description:The potential use of elm wood in lignocellulosic industries has been hindered by the Dutch elm disease (DED) pandemics, which have ravaged European and North American elm groves in the last century. However, the selection of DED-resistant cultivars paves the way for their use as feedstock in lignocellulosic biorefineries. Here, the production of cellulose nanofibers from the resistant Ulmus minor clone Ademuz was evaluated for the first time. Both mechanical (PFI refining) and chemical (TEMPO (2,2,6,6-tetramethylpiperidine-1-oxyl radical)-mediated oxidation) pretreatments were assessed prior to microfluidization, observing not only easier fibrillation but also better optical and barrier properties for elm nanopapers compared to eucalyptus ones (used as reference). Furthermore, mechanically pretreated samples showed higher strength for elm nanopapers. Although lower nanofibrillation yields were obtained by mechanical pretreatment, nanofibers showed higher thermal, mechanical and barrier properties, compared to TEMPO-oxidized nanofibers. Furthermore, lignin-containing elm nanofibers presented the most promising characteristics, with slightly lower transparencies.

Proteome response in maize to methyl jasmonate

Project description:Jasmonic acid (JA) and methyl jasmonate (MeJA) regulate plant development, resistance to stress, and insect attack by inducing specific gene expression. However, little is known about the mechanism of plant defense against herbivore attack at a protein level. Using a high-resolution 2-DE gel, we identified 60 MeJA-responsive proteins and measured protein expression level changes. Among these 62 proteins, 43 proteins levels were increased while 11 proteins were decreased. We also found eight proteins uniquely expressed in response to MeJA treatment. The proteins identified in this study have important biological functions including photosynthesis and energy related proteins (38.4%), protein folding, degradation and regulated proteins (15.0%), stress and defense regulated proteins (11.7%), and redox-responsive proteins (8.3%). We found MeJA could not only induce plant defense mechanisms to insects, it also enhanced toxic protein production that potentially can be used for bio-control of Asian corn borer.
