Project description:Dimethyl sulfoxide (DMSO) is widely used as a solvent for water-insoluble substances and has biological properties such as inducing cellular differentiation, scavenging free radicals, and acting as a radioprotectant. DMSO has also been reported to enhance mineralization in osteoblastic cells. However, its molecular mechanisms have not yet been fully elucidated. In this study, we identified the differentially expressed genes associated with dimethyl sulfoxide-enhanced mineralization in mouse MC3T3-E1 preosteoblast cells using GeneChip® oligonucleotide microarrays.

Common differentiating agents facilitate transcriptional reprogramming by bromodomain inhibition

Project description:Dimethyl sulfoxide (DMSO) and hexamethylene bisacetamide (HMBA) are biologically active molecules that have shown clinical efficacy in cancer and inflammation, though their therapeutic application has been limited, partly because their mechanism of action is unknown. We demonstrate that DMSO and HMBA facilitate rapid transcriptional reprogramming via competitive inhibition of bromodomain-containing proteins. These results identify novel therapeutic avenues and provide context to over 50 years of biomedical research with these agents.

Project description:Adult male marine fish were exposed to 100ng/L of ethinyl estradiol (EE2) for 7 days in 1.8L vessels, dimethyl sulfoxide (DMSO) was used as carrier never reaching concentrations > 0.001% v/v. The same volume of DMSO was added to the solvent control vessels. Culture conditions, monitored daily, were as follows: temperature 18.8 ± 0.2°C, pH 7.94±0.03, dissolved oxygen saturation 99.3% ± 1.1 and 16:8 hours of light:dark photoperiod. 90% of the water was renewed daily and EE2 and DMSO concentrations were adjusted after that.
