Project description:The aim of the study was to identify gene expression profiles that distinguished different stages of osteoblast commitment, and determine whether the same osteoblast cell populations from mice of different ages had distinguishable expression profiles that may indicate functional changes are apparent for the cell populations during different stages of skeletal development. Pre-osteoblastic and mature osteoblastic cells were isolated from collagenase digested bones from 5 week old and 8 week wild type C57BL6 mice by FACS based on Lin, Sca-1, CD51, and CD31 cell surface markers.

Project description:Tightly regulated and cell-specific NADPH-oxidases (Nox) represent one of the major sources of reactive oxygen species (ROS) signaling molecules that are involved in tissue development and stem cell self-renewal. We have characterized the role of Nox4 in osteo-progenitors during postnatal bone development. Nox4 expression in bone and ROS generation were increased during early osteoblast differentiation and bone development. Stromal osteoblastic cell self-renewal, proliferation and ROS production were significantly lower in samples from whole body Nox4 knockout mice (Nox4-/-) and conditional knockout (CKO) mice with depletion of Nox4 in the limb bud mesenchyme compared with those from control mice (Nox4fl/fl), but they were reversed after 9 passages. In both sexes, bone volume, trabecular number and bone mineral density were significantly lower in 3-week old CKO and Nox4-/- mice compared with Nox4fl/fl controls. This was reflected in serum levels of bone formation markers alkaline phosphatase (ALP) and procollagen 1 intact N-terminal propeptide (P1NP). However, under-developed bone formation in 3-week old CKO and Nox4-/- mice quickly caught up to levels of control mice by 6-week of age, remained no different at 13-week of age, and was reversed in 32-week old male mice. Osteoclastogenesis showed no differences among groups, however, CTX1 reflecting osteoclast activity was significantly higher in 3-week old male CKO and Nox4-/- mice compared with control mice, and significantly lower in 32-week old Nox4-/- mice compared with control mice. These data suggest that Nox4 expression and ROS signaling in bone and osteoblastic cells coordinately play a role on osteoblast differentiation, proliferation and maturation.

Project description:To determine the metabolic requirements of key autoimmune populations in a spontaneous lupus-prone mouse model, we carried out RNA-seq of BXSB.Cg-Cd8atm1Mak Il15tmImx Yaa (Yaa DKO) mice following glycolytic inhibition with 2-deoxyglucose (2DG). Pre-symptomatic (6-week old) mice were used as controls for symptomatic (10-week old) mice with or without long-term (4-week) treatment with 2DG. As a result, we found that 2DG-mediated glycolytic inhibition preferentially affected gene signatures of activated B cells and not T cells or other immune cell populations.

Project description:The relationships between cancer cells and the microenvironment play a critical role in cancer growth and development. The bone stroma consists of mesenchymal stem cells (MSCs) and mature osteoblasts that promote cancer growth. Yet it is not completely understood what are the molecular processes guiding cancer cells progression to the bone. In this study, a co-culture assay and subsequent gene profiling arrays were used to compare the gene expression profile of a pre-osteoblastic cell line (MBA-15) with that of a mammary adenocarcinoma (DA3) cells. Following co-culture, cells were separated by magnetic beads based on the expression of CD326 antigen. RNA was purified and hybridized on gene expression array. The gene expression pattern changes were followed by qRT-PCR. We demonstrate that co-cultured DA3 cells express elevated levels of genes that regulate growth and responses to both hormonal stimulus and wounding, as well as reduced expression of genes related to lipid metabolism. Also, co-cultured pre-osteoblastic cells showed reduced expression of cell-junction genes. The study presents a simplified model system, composed of pre-osteoblastic and mammary cancer cells, that potentially mimics the molecular interactions in the tumor microenvironment which contribute to tumor-progression. Gene expression was studied in 2 mouse cell lines: MBA-15, a pre-osteoblastic stromal cell line, and DA3, a mammary adenocarcinomal cell line. The interaction between these 2 cell lines was also analysed by gene expression. The experiment was performed in duplicates (see Date in the Sample characteristics field).

Project description:the intervention period was 4 weeks, and treadmill exercises were conducted for all exercise groups. Two sessions per day were administered. A minimum of a one-hour break was allocated between sessions. the 9-week-old C57BL/6 young male mice and 84-week-old mice were purchased from Central Lab. The groups were divided into young control (YC), young exercise (YE), old control (OC), and old exercise (OE) groups. Each group comprised three mice and all mice were subjected to RNA sequencing analysis.

Project description:The aim was to identify pathways and genes that are transcriptionally deregulated in osteosarcoma due to changes in CpG island DNA methylation. In order to identify candidates, we compared low passage cell cultures derived from a mouse model of osteosarcoma to mature osteoblasts derived by in vitro differentiation of the mouse bone marrow stromal cell line, Kusa4b10. Under cell culture osteoblastic differentiating conditions, Kusa4b10 cells acquire a mature osteoblastic phenotype (21 days).
