Project description:We explored the transcriptomic changes of synthetic Brassica allohexaploid by comparing to its parents using a high-throughput RNA-Seq method. A total of 35644409 sequence reads were generated, and 32642 genes were aligned from the data. There were 29260, 29060 and 29697 genes identified in Brassica rapa, Brassica carinata, and Brassica allohexaploid, respectively. We screened differentially expressed genes (DEGs) by a standard of two-fold or greater change in expression and false discovery rate (FDR) no more than 0.001. As a result, 7397 DEGs were detected between Brassica hexaploid and its parents. A large proportion of the 3184 DEGs between Brassica hexaploid and its paternal parent B. rapa was involved in biosynthesis of secondary metabolites, plant-pathogen interaction, photosynthesis, and circadian rhythm. Between Brassica hexaploid and its maternal parent B. carinata, 2233 DEGs were screened. A lot of them had functions of plant-pathogen interaction, plant hormone signal transduction, ribosome, limonene and pinene degradation, photosynthesis, and also biosynthesis of secondary metabolites. In addition, we found many transcription factor genes, methyltransferase and methylation genes that showed differential expression between Brassica hexaploid and its parents. Project description:Taraxacum koksaghyz is currently one of the most investigated plants as a potential source of natural rubber. This poly(cis-1,4-isoprene) polymer and further valuable secondary metabolites are produced in laticifer cells, which exist as a network of elongated tubules throughout the plant and have a milky cytoplasm known as latex. However, a detailed analysis of the overall laticifer system and a deeper knowledge about the interaction with the surrounding tissues has not been performed. Therefore, we used a bacterial ribonuclease for the development of transgenic dandelion plants with omitted latex production. Comprehensive histological, biochemical and proteomic analyses served to characterize these plants. Thereby, we were able to generate a molecular map based on the protein expression pattern of laticifers and whole Taraxacum koksaghyz roots at different stages of plant development. This enables us to understand the interplay and tissue specificity of different biosynthesis routes and narrow down biological functions of latex.

Project description:Contamination with enterohemorrhagic Escherichia coli O157:H7 (EHEC) is a worldwide problem but there is no effective therapy available for EHEC infection. Biofilm formation is closely related with EHEC infection and is one of the mechanisms of antimicrobial resistance. Antibiofilm screening of 560 plant secondary metabolites against EHEC shows that ginkgolic acids C15:1 and C17:1 at 5 μg/ml and Ginko biloba extract at 100 μg/ml significantly inhibited EHEC biofilm formation on the surface of polystyrene, nylon membrane, and glass. Importantly, the working concentration of ginkgolic acids and G. biloba extract did not affect bacterial growth and has been known to be non-toxic to human. Transcriptional analyses showed that ginkgolic acid C15:1 repressed curli genes and prophage genes in EHEC, which were corroborated by reduced fimbriae production and biofilm reduction in EHEC. Interestingly, ginkgolic acids and G. biloba extract did not inhibit the biofilm formation of commensal E. coli K-12 strain. The current study suggests that plant secondary metabolites are important resource of biofilm inhibitors, as well as other bioactive compounds. E. coli GeneChip Genome 2.0 Array (Affymetrix, P/N 900551, Santa Clara, USA) was used to study the differential gene expression of the E. coli O157:H7 cells after the treatment with ginkgolic acid C15:1 (0.005 mg/ml). Cells were inoculated into 25 ml LB medium in 250 mL shake flasks with a starting OD600 of 0.05, and cultured at 37oC for 8 h without shaking in the presence or absence of ginkgolic acid C15:1 (5 μg/ml). To prevent RNA degradation, RNase inhibitor (RNAlater, Ambion, TX, USA) was added, and the EHEC cells were collected by centrifugation at 10,000 rpm for 1 min. The cell pellets obtained were immediately frozen with dry ice and stored at -80°C. Total RNA was isolated using a Qiagen RNeasy mini Kit (Valencia, CA, USA).

Project description:The aim of this study is to investigate the effects of dietary plant and animal proteins on gut metabolism and markers for colorectal cancer as well as blood protein metabolites and markers for type 2 diabetes in healthy adults. The study participants will be stratified into three groups with different protein composition in diets: 1) animal 70%/plant 30%; 2) animal 50%/plant 50% and 3) animal 30%/plant 70%. The participants will get part of their diet as ready foods or raw material to promote their compliance. The participants will also get personal advice for their diets. Blood, stool and urine samples will be collected in the beginning and in the end of the 12 week intervention, as well as phenotype measures like BMI, blood pressure and body composition. The participants will also fill food diary before and in the end of the intervention.
