Project description:Alfalfa (Medicago sativa L.) is a forage legume with significant agricultural value worldwide. MicroRNAs (miRNAs) are key components of post-transcriptional gene regulation and essentially control almost all aspect of plant growth and development. Although miRNAs have been reported from alfalfa but their expression profiles in different tissues and novel miRNAs as well as their targets have not been confirmed in this plant species. Therefore, we sequenced small RNAs in whole plantlets, shoots and roots of three different alfalfa genotypes (Altet-4, NECS-141 and NF08ALF06) to identify tissue-specific profiles. After comprehensive analysis using bioinformatics methods, we have identified 100 miRNA families, of which 21 belongs to the highly conserved families whereas the remaining 79 families are conserved between M. truncatula and M. sativa. The profiles of the six highly expressed conserved miRNA families (miR156, 159, 166, 319, 396, 398,) were relatively similar between the plantlets, roots and shoots of three genotypes. Contrastingly, the differenecs were robust between shoots and roots for miR160 and miR408 levels, which were low in roots compared to shoots. The study also has identified 17 novel miRNAs that also differed in their abundanecs between tissues of the alfalfa genotypes. Additionally, we have generated and analyzed the degradome libraries from three alfalfa genotypes that has confirmed 69 genes as targets for 31 miRNA families in alfalfa. The identification of conserved and novel miRNAs as well as their targets in different tissues of three genotypes not only enhanced our understanding of miRNA-mediated gene regulation in alfalfa but could also be useful for practical applications in alfalfa as well as related legume species.

Project description:We have characterized the changes in miRNA expression profiles in rice leaves under drought stress and As stress and compared these to unstressed leaves. 10 pairs of drought responsive and 8 pairs of As responsive miRNA-gene were identified and validated by qRT-PCR. This study identifies putative specific miRNA-mRNA regulatory modules with roles during drought and As stress. Putative microRNAs identified in this study are involved in hormone signaling, lipid and carbohydrate metabolism, and antioxidant defence. The results of this study will assist in elucidating the role of miRNAs in post-transcriptional regulation of target genes during abiotic stress and may contribute to the development of strategies to engineer drought and heavy metal resistance.

Project description:The central part of Brazil, consisting mostly of the Cerrado Biome, is considered to be the new frontier for increasing Brazilian wheat production. However, rainfed wheat production in that area must cope with drought stress. In order to better understand the drought response, we analyzed the mRNA profiling under drought in roots and leaves of the cultivar MGS1 Aliança (a well-adapted cultivar to the Cerrado). We identified 4,422 candidate genes in roots and leaves.

Project description:Two alfalfa varieties, 'Chilean' (M. sativa ssp. sativa var. Chilean, drought sensitive) and 'Wisfal' (M. sativa ssp. falcata var. Wisfal, drought tolerant), with contrasting water use efficiency were subjected to water withholding for 11 days followed by re-watering. Samples were taken for well-watered plants and plants after five, eight, eleven days of drought stress as well as plants after recovery for one day following drought stress. Roots and shoots were sampled and analyzed separately by expression profiling using Affymetrix Medicago GeneChip.

Project description:Drought is one of the major factor that limits crop production and reduces yield. To understand the early response of plants under nearly natural conditions, pepper plants were grown in a greenhouse and drought stressed by withholding water for one week. Plants adapted to the decreasing water content of the substrate by adjustment of their osmotic potential in roots by accumulation of raffinose, glucose, galactinol and proline. In contrast in leaves levels of fructose, sucrose and also galactinol increased. Due to the water deficit cadaverine, putrescine, spermidine and spermine accumulated in leaves whereas the concentration of polyamines was reduced in roots. These polyamines are suggested to rather act as stress protectants than for osmotic adjustment. To understand the molecular basis of the response to this early drought stress better, four suppression subtractive hybridisation libraries from leaves and roots were constructed. Microarray technique was used to identify differentially expressed genes. A total of 109 unique ESTs were detected. The diversity of the putative functions of all identified genes confirms the complexity of the plant response to drought stress. Project description:Plants have evolved to possess adaptation mechanism to cope with drought stress by reprograming transcriptional networks through drought responsive transcription factors, which in turn mediate morphological and physiological changes. NAM, ATAF1-2, and CUC2 (NAC) transcription factors are known to be associated with various developmental processes and stress tolerance. In this study, we functionally characterized the rice drought responsive NAC transcription factor OsNAC14. OsNAC14 was predominantly induced at meiosis stage, and induced by drought, high salinity, ABA and low temperature in leaves than roots. Overexpression of OsNAC14 resulted in drought tolerance at the vegetative growth stage and enhanced filling rate at vegetative growth. OsNAC14 overexpression elevated expression of genes related to DNA damage repair, defense response, strigolactone biosynthesis, which correlated with resistance to drought tolerance. Furthermore, OsNAC14 directly bound to the promoter of drought inducible OsRAD51A1, a key component in homologous recombination in DNA repair system. These results indicate that OsNAC14 mediate drought tolerance by recruiting factors involved in DNA damage repair and defense response to enable plant to protect from cellular damage caused by drought stress, thereby provide mechanism for drought tolerance.

Project description:The data present global gene expression profiling of roots of two barley genotypes with contrasting ability to cope with drought stress. We used the whole root system and the second leaf of CamB and Maresi genotypes subjected to 10-days of mild drought at seedling stage. The transcriptomes of leaves served in the presented study as a background to depict, which genes may respond with the expression changes in an organ-specific manner. Our data indicate that the mild drought resulted in more changes in the transcriptomes of roots than in the leaves and more differentially expressed genes (DEGs) were root-specific. We also found that similar number of DEGs were induced or repressed by the stress in roots of CamB genotype, whereas in roots of more drought susceptible Maresi a down-regulation of gene expression prevailed. We identified 88 genes encoding transcription factors and gene expression regulators that were differentially expressed in roots and the majority of them were root-specific. We discuss their probable function in drought response and tolerance and predicted a possible regulatory network downstream of selected transcription factors.
